Available in PDF, EPUB and Kindle. Book excerpt: Warfarin (also known under the brand names of Coumadin, Jantoven, Marevan, and Waran) is an anticoagulant medication that is administered orally or, very rarely, by injection. It is used for the prophylaxis of thrombosis and embolism in many disorders. Its activity has to be monitored by frequent blood testing for the international normalised ratio (INR). It is named for the Wisconsin Alumni Research Foundation. Warfarin is a synthetic derivative of coumarin, a chemical found naturally in many plants, notably woodruff (Galium odoratum, Rubiaceae), and at lower levels in liquorice, lavender and various other species. Warfarin was originally developed as a rat poison; however, more modern poisons are much more potent and toxic (e.g., brodifacoum). Warfarin and contemporary rodenticides belong to the same class of drugs (coumarins) and both decrease blood coagulation by interfering with vitamin K metabolism. For this reason, drugs in this class are also referred to as vitamin K antagonists.

Available in PDF, EPUB and Kindle. Book excerpt: Global dietary recommendations emphasize the consumption of plant-based foods for the prevention and management of chronic diseases. Plants contain many biologically active compounds referred to as phytochemicals or functional ingredients. These compounds play an important role in human health. Prior to establishing the safety and health benefits of these compounds, they must first be isolated, purified, and their physico-chemical properties established. Once identified, their mechanisms of actions are studied. The chapters are arranged in the order from isolation, purification and identification to in vivo and clinical studies, there by covering not only the analytical procedures used but also their nutraceutical and therapeutic properties.

Available in PDF, EPUB and Kindle. Book excerpt: Coumarins are widely distributed in nature and can be found in a large number of naturally occurring and synthetic bioactive molecules. The unique and versatile oxygen-containing heterocyclic structure makes them a privileged scaffold in Medicinal Chemistry. Many coumarin derivatives have been extracted from natural sources, designed, synthetized, and evaluated on different pharmacological targets. In addition, coumarin-based ion receptors, fluorescent probes, and biological stains are growing quickly and have extensive applications to monitor timely enzyme activity, complex biological events, as well as accurate pharmacological and pharmacokinetic properties in living cells. The extraction, synthesis, and biological evaluation of coumarins have become extremely attractive and rapidly developing topics. A large number of research and review papers have compiled information on this important family of compounds in 2020. Research articles, reviews, communications, and concept papers focused on the multidisciplinary profile of coumarins, highlighting natural sources, most recent synthetic pathways, along with the main biological applications and theoretical studies, were the main focus of this book. The huge and growing range of applications of coumarins described in this book is a demonstration of the potential of this family of compounds in Organic Chemistry, Medicinal Chemistry, and different sciences related to the study of natural products. This book includes 23 articles: 17 original papers and six review papers.

Available in PDF, EPUB and Kindle. Book excerpt: The available parenteral and oral anticoagulants have a large clinical use. Understanding biochemistry of anticoagulants may help to improve therapeutic strategies. Resistance to vitamin K antagonist drugs might be a problem for rodent populations. Patients who have thrombogenic risk factors should be anticoagulated. The need for cardiac implantable electronic devices is increasing, and there is a substantial number of patients who are on oral anticoagulant therapy. Prothrombin complex concentrate and other plasma concentrates are useful to deal with over-coagulated situations. The efficacy and safety of non-vitamin K antagonist oral anticoagulants have been proven in large phase III trials. The real-world data suggest even better outcomes with these agents compared to vitamin K antagonists.

Almost 100 years have passed since the initial discovery of this complex substance and, during this time, understanding of the nature and uses of heparin and related molecules has grown dramatically. The aim of this volume is to summarise the developments that have led to the current status of both heparins as drugs and the field of heparin research, with a focus on the particularly rapid progress that has been made over the past three decades. Individual sections are dedicated to the nature of heparin as a biological molecule, the current approaches and techniques that are used to ensure the safety and reliability of heparin as a medicine, the clinical pharmacology of heparin as an anticoagulant drug, effects and potential applications of heparin aside of those involving haemostasis and, finally, the nature and potential uses of heparin-like materials from both natural and synthetic sources.

Included are contributions by pioneers in the discovery of P450, with chapters on the molecular and functional properties of P450 and cutting-edge applications knowledge from various fields. P450 research has its roots in metabolism, but the true beginning was in 1962 with the publication by Tsuneo Omura and Ryo Sato in The Journal of Biological Chemistry on their discovery of the cytochrome. Following this groundbreaking study, over the last half-century, research has revealed that many forms of P450 exist in animals, plants and microorganisms. P450 research has expanded into many different fields including medicine, agriculture and biotechnology and has drawn the attention of industries for its bioengineering applications, such as drug development and creation of the “blue rose”. Also, research on nuclear receptors, which has grown out of research on the regulatory mechanisms of P450 genes, has become an important area in biology, medical science, pharmacology and clinical medicine—for example, with recent developments in personalized medicines. This book will draw readers into the important and exciting world of P450 and will encourage young students and scientists in P450 research to continue expanding the field via new approaches.
