Available in PDF, EPUB and Kindle. Book excerpt: This book provides microscopic insights into chemical properties of NO on metal surfaces. NO/metal systems have been studied intensively to understand heterogeneous catalysis to detox exhaust NOx gas. The identification and componential analysis of various and mixed chemical species of NO adsorbed onto the surfaces have been significant challenges faced by conventional experimental techniques, such as vibrational spectroscopies. The author investigated "individual" NO molecules on Cu surfaces using low-temperature scanning tunneling microscopy (STM). STM not only provides information on the geometric, electronic, and vibrational properties at the single-molecule level; it is also able to manipulate molecules on surfaces to induce chemical reaction. Exploiting those techniques, the author chemically identified individual NO-related species on the surfaces and discovered new reaction processes for NO reduction, which provides microscopic insights into the catalytic mechanisms. The author also visualized wave functions of electrons in a valence orbital of NO and demonstrated that the wave functions are modified by the formation of covalent bonding or hydrogen bonding. This is, namely, "the visualization of quantum mechanics in real space," which is certainly worth reading. Furthermore, the book demonstrates that direct observation of valence orbitals helps to elucidate the reactivity of molecules adsorbed onto surfaces. This innovative approach to studying molecular properties will contribute to further development of STM and its related methods.

Available in PDF, EPUB and Kindle. Book excerpt: The significant role that nitric oxide plays in human physiology is linked to the ability of NO to bind to iron forming mono-nitrosyl iron complexes. Protein-bound and low-molecular-weight dinitrosyl iron complexes (DNICs) are known to form in excess NO. Studies of such biological DNICs have relied on their paramagnetism and characteristic EPR signal of g value of 2.03. It has been suggested that DNICs act in vivo as NO storage (when protein-bound) and transfer agents (when released by, for example, free cysteine). Biological DNICs, mainly resulting from iron-sulfur cluster degradation, are difficult to extract and isolate, thereby preventing their full characterization. Thus, development of synthetic DNICs is a promising approach to model and better understand the formation and function of biological DNICs, the scope of donor ligands that might coexist with Fe(NO)2 units, the redox levels of bio-DNICs, and establish other spectroscopic techniques appropriate for characterization. A series of N-heterocyclic carbene (NHC) and imidazole (Imid) complexes has been characterized as mimics of histidine-containing DNICs. The pseudo-tetrahedral L2Fe(NO)2 complexes have NO stretching frequencies and redox potentials that suggest the NHCs are slightly better donors than Imids, however the two types of ligands have similar steric properties. Both the EPR-active, {Fe(NO)2}9 and the EPR-silent, {Fe(NO)2}10 states can be accessed and stabilized by the NHC. Nitric oxide transfer studies have shown that only the {Fe(NO)2}9 complexes are capable of transferring NO to a suitable NO trapping agent. Deprotonation of the distal nitrogen functionality in the imidazolate ligands of [(Imidazole)2Fe(NO)2]- leads to aggregation forming molecular squares of {Fe(NO)2}9 units bridged by the imidazolates. These interesting tetrameric complexes are examined by X-ray diffraction, EPR, and Mössbauer studies. The paramagnetic tetrameric complexes have multiple redox events observed by cyclic voltammetry. Mössbauer spectral data of the tetrameric complexes are compared with Mössbauer data obtained for a series of NHC-containing DNICs. Iron and cobalt-containing mononitrosyl N2S2 model complexes of the nitrile hydratase enzyme active site demonstrate sulfur-based reactivity resulting in the formation of polymetallic complexes. In all cases, shifts in the nitrosyl stretching frequencies demonstrate substantial transfer of electron density from the (NO)M(N2S2) moiety to the metal-acceptor site.

Book excerpt: Nitric oxide (NO) is a gas that transmits signals in an organism. Signal transmission by a gas that is produced by one cell and which penetrates through membranes and regulates the function of another cell represents an entirely new principle for signaling in biological systems. NO is a signal molecule of key importance for the cardiovascular system acting as a regulator of blood pressure and as a gatekeeper of blood flow to different organs. NO also exerts a series of other functions, such as acting a signal molecule in the nervous system and as a weapon against infections. NO is present in most living creatures and made by many different types of cells. NO research has led to new treatments for treating heart as well as lung diseases, shock, and impotence. Scientists are currently testing whether NO can be used to stop the growth of cancerous tumors, since the gas can induce programmed cell death, apoptosis. This book is the first comprehensive text on nitric oxide to cover all aspects--basic biology, chemistry, pathobiology, effects on various disease states, and therapeutic implications. Edited by Nobel Laureate Louis J. Ignarro, editor of the Academic Press journal, Nitric Oxide Authored by world experts on nitric oxide Includes an overview of basic principles of biology and chemical biology Covers principles of pathobiology, including the nervous system, cardiovascular function, pulmonary function, and immune defense

Reactivity of Tropocoronand-Bound Cobalt(III) Nitrite with Nitric Oxide as a Function of Polymethylene Linker Chain Length The studies on the dependence of cobalt(II) tropocoronand reactivity with nitric oxide (NO) on linker chain length, described in Chapter 4, were expanded to include the chemistry of the cobalt(III) nitrites [Co(NO2)(TC-n,n)] (n = 4 - 6) with NO. Complete conversion from the cobalt(III) nitrite to the cobalt mononitrosyl [Co(NO)(TC-4,4)] is demonstrated upon exposure to NO (g). In contrast, exposure of [Co(N0 2)(TC-5,5)] and [Co(N0 2)(TC-6,6)] to NO (g) results in conversion to a cobalt dinitrosyl adduct. This Chapter aims to broaden the field of cobalt(III) chemistry with NO. Chapter 6. Nitric Oxide Reactivity of Cobalt(III) Triflate and Cobalt(III) Thiolate Tropocoronand Complexes The reactivity of two cobalt(III) tropocoronands with NO (g) is described. [Co(TC-4,4)](OTf) undergoes reductive nitrosylation in the presence of NO (g), forming [Co(NO)(TC-4,4)], N20, and additional reactive species. The reaction releases one equivalent of protons for every equivalent of [Co(TC-4,4)](OTf) consumed. [Co(SC 6F5)(TC-4,4)] forms [Co(NO)(TC-4,4)] and the disulfide F5C6SSC6F5 in the presence of NO (g). We hypothesize that disulfide formation is followed by electrophilic aromatic substitution of F5C6S* onto the tropocoronand aromatic ring. Efforts to prepare substituted tropocoronand ligands and test the electrophilic aromatic substitution hypothesis are described. Chapter 7. Synthesis and Characterization of Mononuclear, Pseudotetrahedral Cobalt(III) Compounds The synthesis and characterization of two mononuclear cobalt(III) tropocoronand complexes, [Co(TC-5,5)](BF 4) and [Co(TC-6,6)](BPh 4), are reported. The cobalt(III) centers exist in rare pseudotetrahedral conformations, with twist angles of 650 and 740 for the [Co(TC-5,5]* and [Co(TC-6,6)]* species, respectively. Structural and electrochemical characteristics are compared with those of newly synthesized [Ga(TC-5,5)](GaCl4) and [Ga(TC-6,6)](GaCl 4) analogs.

Abstract: Abstract of thesis entitled BIOMIMETIC REACTIONS OF NITRIC OXIDE SYNTHASE - STUDY OF THE REACTIONS OF N-SUBSTITUTED-N-HYDROXYGUANIDINES WITH METALLOPORPHYRIN AND NON-HEME COMPLEXES Submitted by Chu Tsun Tung For the Degree of Doctor of Philosophy at The University of Hong Kong in April 2007 Nitric oxide (NO) is one of the most important biological messengers. The generation of NO in animals is carried out by nitric oxide synthase (NOS), an enzyme which catalyzes the two-step. The first step is a 2-electron oxidation of L- Arg to N-hydroxyarginine (NHA), which similar to cytochrome P450 monoxidase type reaction. The second step is a three 3-electron oxidation of N-hydroxyarginine to NO and citrulline. Continuous studies of unusual aerobic oxidation of NHA catalyzed by NOS lead to several proposed mechanisms for the reaction. One of the proposed mechanisms features the ligation of NHA to Fe(III)-heme followed by Fe- O bond homolysis to generate Fe(II)-heme and iminoxy radical. This mechanism was supported by a chemical model system that employs fluorenone oxime and [Fe(OH)(Por)]. Inspired by this model, some N-hydroxyguanidines (NHGs), instead of an oxime, were used as substrate in our studies because N-hydroxyguanidine is the characteristic function group of NHA. All these NHGs were found ligating to [Fe(TMP)], and an X-ray crystal structure of NHG ligated [Fe(TMP)] has been determined, which supports that NHGs can ligate to iron via oxygen. Mechanistic studies of the aerobic oxidation of NHGs support that the NHG oxidation is initiated by O -assisted Fe-O bond homolysis. Guanidine and urea were thought to be the reaction products of iminoxy radical with other iminoxy radical or NHGs. HNO was thought to be one of the nitrogen oxide products, which was supported by the detection of N O in the reaction headspace gas. A modified mechanism of NOS catalyzed oxidation of NHA to citrulline and NO was proposed according to the findings of our studies and other new mechanistic findings about NOS reaction. The aerobic oxidations of some trisubstituted NHGs catalyzed by [Fe(OH)(TMP)] generated 3-amino-1,2,4-oxadiazoles as products. Some non-heme iron complexes and metalloporphyrins were tested for the catalytic properties towards the oxidative cyclization of NHGs. Complexes [nBu N][Fe(L)(Cl) ] (L = 4 2 bispyridylamide type ligands) were found to be good catalysts, which gave the corresponding oxadiazole in 80% yield within 1.5 hours from N, N-dimethyl-N'- benzyl-N"-hydroxyguanidine (DMBH). Mechanistic studies for the oxidative cyclization of NHGs catalyzed by the [nBu N][Fe(L)(Cl)] complexes were 4 2 performed and a mechanism for the reaction was proposed. Available in PDF, EPUB and Kindle. Book excerpt: The Chemistry and Biology of Nitroxyl (HNO) provides first-of-its-kind coverage of the intriguing biologically active molecule called nitroxyl, or azanone per IUPAC nomenclature, which has been traditionally elusive due to its intrinsically high reactivity. This useful resource provides the scientific basis to understand the chemistry, biology, and technical aspects needed to deal with HNO. Building on two decades of nitric oxide and nitroxyl research, the editors and authors have created an indispensable guide for investigators across a wide variety of areas of chemistry (inorganic, organic, organometallic, biochemistry, physical, and analytical); biology (molecular, cellular, physiological, and enzymology); pharmacy; and medicine. This book begins by exploring the unique molecule’s structure and reactivity, including important reactions with small molecules, thiols, porphyrins, and key proteins, before discussing chemical and biological sources of nitroxyl. Advanced chapters discuss methods for both trapping and detecting nitroxyl by spectroscopy, electrochemistry, and fluorescent inorganic cellular probing. Expanding on the compound’s foundational chemistry, this book then explores its molecular physiology to offer insight into its biological implications, pharmacological effects, and practical issues. Presents the first book on HNO (nitroxyl or azanone), an increasingly important molecule in biochemistry and pharmaceutical research Provides a valuable coverage of HNO’s chemical structure and significant reactions, including practical guidance on working with this highly reactive molecule Contains high quality content from recognized experts in both industry and academia

Book excerpt: The series Structure and Bonding publishes critical reviews on topics of research concerned with chemical structure and bonding. The scope of the series spans the entire Periodic Table and addresses structure and bonding issues associated with all of the elements. It also focuses attention on new and developing areas of modern structural and theoretical chemistry such as nanostructures, molecular electronics, designed molecular solids, surfaces, metal clusters and supramolecular structures. Physical and spectroscopic techniques used to determine, examine and model structures fall within the purview of Structure and Bonding to the extent that the focus is on the scientific results obtained and not on specialist information concerning the techniques themselves. Issues associated with the development of bonding models and generalizations that illuminate the reactivity pathways and rates of chemical processes are also relevant. The individual volumes in the series are thematic. The goal of each volume is to give the reader, whether at a university or in industry, a comprehensive overview of an area where new insights are emerging that are of interest to a larger scientific audience. Thus each review within the volume critically surveys one aspect of that topic and places it within the context of the volume as a whole. The most significant developments of the last 5 to 10 years should be presented using selected examples to illustrate the principles discussed. A description of the physical basis of the experimental techniques that have been used to provide the primary data may also be appropriate, if it has not been covered in detail elsewhere. The coverage need not be exhaustive in data, but should rather be conceptual, concentrating on the new principles being developed that will allow the reader, who is not a specialist in the area covered, to understand the data presented. Discussion of possible future research directions in the area is welcomed. Review articles for the individual volumes are invited by the volume editors. Readership: research scientists at universities or in industry, graduate students Special offer for all customers who have a standing order to the print version of Structure and Bonding, we offer free access to the electronic volumes of the Series published in the current year via SpringerLink.

Book excerpt: Nitric oxide (NO) is a molecule that is essential for life and regulates both beneficial and harmful processes. Because this gaseous radical influences many aspects of health and disease, we wish to explore the relationship between NO and physiology/pathophysiology. To this end, we seek to create tools for the fluorescent imaging of NO in vivo. We have adapted an existing small molecule-based sensor for more biologically relevant applications by including it within a polymeric film. We have also developed turn-on fluorescent sensors for NO based on conjugated polymers, which demonstrated good selectivity and sensitivity for this analyte. In addition, we have prepared a related sensor that will detect nitroxyl (HNO) but not NO. These systems demonstrate the versatility and value of the conjugated polymer scaffold for sensing applications. Many targets of the diatomic radical NO contain redox active units, such as transition metals or thiolate ligands. To gain insight into how NO might regulate biological processes by interacting with these redox active species, we have initiated a fundamental study of the reactivity of NO with transition metal thiolate model complexes. Our explorations in this field have yielded unique nickel and cobalt nitrosyl species with atypical electronic and structural parameters. These studies have suggested intermediates for the more biologically relevant iron nitrosyl complexes that have not yet been observed may exist. Furthermore, the NO chemistry of these small molecule nickel and cobalt thiolate complexes may guide future biological investigations into the regulation of nickel and cobalt metalloproteins by NO.
