Book excerpt: This book reviews collagen-based biomaterials that have been applied broadly to tissue engineering and local drug delivery applications and lays out a landscape for developing a multifunctional biograft material from collagen polymers. The book also discusses current shortcomings in collagen based drug delivery opportunities, including poor mechanical properties, rapid proteolytic degradation, and cursory control over physical properties and molecular release profiles. Finally, a review of application of the collagen biograft materials for promoting neovascularization and tissue regeneration is presented, using examples of established in-vivo chicken egg chorioallantoic membrane (CAM) model. Use of heparin for affinity-based vascular endothelial growth factor (VEGF) retention in collagen constructs is also discussed for promoting neovascularization. Reviews state-of-the-art strategies for drug incorporation and retention in collagen; Covers collagen based material applications for improving vascularization and tissue regeneration; Illustrates how to tailor collagen architecture for soft tissue engineering and controlled drug delivery.

Book excerpt: As collagen is the main structural component of body tissues, there has been much effort expended, both in the past and today, to use it in cell culture applications. Indeed, its importance is further stressed by the wide use of collagen biomaterials in tissue repair and regeneration, including the treatment of burns, chronic wounds or bone repair. This book represents a journey through recent scientific history, and describes the massive implications of the research carried out by Bornstein, Ehrmann and Gey on collagen preparations in the 1950s for cell culture and more recently, tissue engineering and personalised medicine.

Book excerpt: The chapter describes the regulatory framework applicable to medical devices, and specifically to biotextiles, principally in the United States, but also in the European Union. With the emergence of new technologies and materials, new forms of regulation have become necessary, and some of the traditional classifications no longer match present-day applications. Problems encountered by manufacturers with regard to premarket approval clearance and material supply quality standards assurance are discussed, as well as issues increasingly arising when one biotextile-based medical device is evolved into another for a different use.

The use of collagen-based biomaterials in tissue engineering applications has increased dramatically over the past few years, owing to extensive research in the field. Multiple cross-linking strategies for collagen have been examined. Various combinations of collagen with other biopolymers have also been investigated in an attempt to increase the tissue function of the collagen biomaterials in their various formulations. Collagen Biomaterials provides a thorough overview of the different uses of collagen-based biomaterials produced for tissue engineering, to offer a functional material for use in regenerative medicine from the laboratory bench to the patient bedside.

Available in PDF, EPUB and Kindle. Book excerpt: Tissue grafting is mostly used for repair and replacement of severely damaged tissues, the key challenges are compatibility, availability of the grafts, complex surgical process and post-operative complications. Hence, additive technologies such as three-dimensional (3D) bioprinting have emerged as promising alternative for tissue engineering in order to ensure safety, compatibility, and rapid healing. The aim of this chapter is to give an elaborate account of 3D printed scaffolds for bone, cartilage, cardio-vascular and nerve tissue engineering. Various components such as polycaprolactone, poly (lactic-co-glycolic acid), and β-tricalcium phosphate, bioglass 45S5, and nano-hydroxyapatite are combined with collagen and its derivatives to achieve specific pore size in the scaffolds for effective restoration of the defects of soft or hard tissues. Likewise, proanthocyanidin, oxidized hyaluronic acid, methacrylated gelatin, are used in collagen based 3D printed scaffolds for cartilage tissue engineering. Bioink with collagen as active component is also used for developing cardio-vascular implants with recellularizing properties. Collagen in combination with silk fibroin, chitosan, heparin sulphate and others are ideal for fabrication of elastic nerve guidance conduits. In view of the background, collagen-supplemented hydrogels can revolutionize future biomedical approaches for the development of complex scaffolds for tissue engineering.

Book excerpt: This book summarizes the properties and applications of conventional and commercially available fiber-forming, bioresorbable polymers, as well as those currently under study, for use as biotextiles. Factors affecting the performance of these biomaterials are presented, and precautionary measures to reduce premature, hydrolytic degradation during manufacturing and processing are discussed. Because of the structural requirements of medical devices and the technological advancements in synthetic fibers and textile technology, the new field of "Biotextiles" has evolved to exploit the potential of various woven, knitted, braided and non-woven textile structures for biomedical applications. Textile substrates provide certain unique mechanical properties to the medical device and because of an inherently high level of porosity, they can encourage cell growth and promote migration and proliferation. Bioresorbable devices that assist in the repair and regeneration of damaged tissues have in recent years replaced many of the permanent prosthetic devices. Thus, the topic of “Bioresorbable Biomaterials” generates much interest and research activity in the field of biomaterials science today. For this reason, the use of bioresorbable polymers as fibers is currently dominating the field of resorbable biomaterials for applications from sutures to tissue engineering scaffolds.
