Book excerpt: With a few notable exceptions, mammalian preimplantation embryos grown in vitro are likely to exhibit sub-optimal or retarded development. This may be manifested in different ways, depending on the species and on the stage(s) of embryonic development that are being examined. For example, bovine embryos often experience difficulty in cleaving under in vitro conditions, and usually cease development at about the 8-cell stage (Wright and Bondioli, 1981). The block to development is stage-dependent; embryos cultured for 24 hr from the I-cell stage are much more capable of developing into viable blastocysts after transfer to oviducts than embryos cultured for 24 hr from the 4-cell stage prior to transfer (Eyestone et oZ. , 1985). Similar problems with in vitro embryo development are encountered in other species. Pig embryos can be grown up to the 4-cell stage in vitro but usually no further (Davis and Day, 1978). In the golden hamster, in the rat and in many outbred strains of mice, development of zygotes in vitro is blocked at the 2-cell stage (Yanagimachi and Chang, 1964; Whittingham, 1975). Even with some inbred mouse strains, embryo development is reduced if very early cleavage stages are used as the starting point for in vitro culture (Spielmann et oZ. , 1980). A common finding is that embryos grown in vitro have reduced cell counts (Harlow and Quinn, 1982; Kane, 1985) and their viability is reduced (Bowman and McLaren, 1970; Papaioannou and Ebert, 1986) compared to equivalent developmental stages recovered from mated animals.

Book excerpt: The study of birth defects has assumed an importance even greater now than in the past because mortality rates attributed to congenital anomalies have declined far less than those for other causes of death, such as infectious and nutritional diseases. It is estimated that as many as 50% of all pregnancies terminate as miscarriages. In the majority of cases this is the result of faulty development. Major congenital malformations are found in at least 2% of all liveborn infants, and 22% of all stillbirths and infant deaths are associated with severe congenital anomalies. Teratological studies of an experimental nature are neither ethical nor justifiable in humans. Numerous investigations have been carried out in laboratory animals and other experimental models in order to improve our understanding of abnormal intrauterine development. In less than two decades the field of experimental teratology has advanced phenomenally. As a result of the wide range of information that is now accumulating, it has become possible to obtain an insight into the causes, mechanisms, and prevention of birth defects. However, considerable work will be needed before these problems can be resolved. The comributions in this volume include some of the more recent and exciting observations on the cellular and molecular aspects of developmental defects. It is not only a documentation of the latest experimental work, but it also indicates new and important areas for future research.

In most higher organisms, the analysis of the cell lineage has to rely on indirect methods. Only in the most suitable systems, like the nematodes, can the pattern of cell division be determined by direct observation under the microscope. In cases where this is not possible, the fate of the cells has to be examined by using cell markers. Most suitable for this purpose are genetic markers, provided that they do not interfere with the developmental pathway to be studied. However, suitable genetic markers and techniques for generating genetic mosaics are available in a few organisms only. Therefore, this volume is largely concerned with Drosophila and the mouse, which have been studied most extensively. In 1929 STURTEVANT introduced the analysis of gynandromorphs into devel opmental genetics. However, this important contribution remained largely unnoticed until the late sixties, when the potential of this technique for deter mining embryonic fate maps and the number of primordial cells was exploited, and the methodology extended to the mapping of mutational foci. In pregnancy, the uterus acquires this impor tance with the arrival of the fertilized egg, which takes up residence for periods ranging from about 2 weeks in the opossum to about 2 years in the elephant. The arrival of the embryo signals a crucial time for the establishment of pregnancy. For several days the blas tocyst remains free in the uterine lumen, where it depends on uterine secretions for its sur vival and differentiation. During this time, essential changes in the endometrium take place in preparation for attachment of the blastocyst and implantation. Early embryonic loss is an economic problem of global proportions in animal husbandry, where, in pigs and cattle for example, some 30% of all fertilizations fail to result in a pregnancy. In humans this figure may be even higher, and estimates of early spontaneous abortions range from 40 to 60% of all conceptions.
