Book excerpt: Honey bees (Apis melliferaL.) are the most agriculturally beneficial eusocial insects for crop pollination. Chemical communication is critical in maintaining colony structure and activity, which may be exploited by parasites. Varroa destructor (Anderson and Trueman; Acari: Varroidae) is regarded as one of the biggest threats to apiculture, blamed for annual colony mortalities of over 30% in some regions. The objectives of this thesis were to identify odorants important to V. destructor for further development of techniques to disrupt its lifecycles through in-colony treatment. Honey bee colony volatile collections involving ex-situ techniques were used to identify individual compounds and odor detection sensitivity of V. destructorto these compounds through gaschromatography-mass spectrometry and gas chromatography-linked electrotarsal detection, respectively. Volatile components identified in this and previous research were then tested for concentration-dependent responses using electrophysiology. Electrotarsogram responsesindicated significant difference among odorants in eliciting responses, suggesting the potential application of this procedure in screening putative repellents or odors that disrupt host detection (disruptants) to live V. destructor. Results from this research can be applied o colony-wide testing of active odorants in developing effective alternative methods for V. destructor control as well as developing methods for future research exploring chemical ecology of social insects.

Despite its great potential, beekeeping industry is facing several constraints, which needs immediate attention. Among these, Varroa destructor Anderson and Trueman, an ectoparasitic mite of brood and adult bees, is a serious pest of Apis mellifera L.). In the last three years, beekeeping in India is adversely affected by this mite. In the present scenario, 90 per cent apiaries and 50 per cent colonies are affected by this mite. Varroa was first described on its native host, the Asian honey bee (Apis cerana Fab.) in 1904 in Java (Oudemans, 1904). Some of behavioural traits which were acquired by honey bees against Varroa are tendency to swarm and willingness to abandon their hives may have effectively countered the mite, but these traits also suggested difficulty in domestication of this species.Genus Varroa consists of at least four but possibly seven distinct species. Among the four recognized species, the most destructive and largest among these is Varroa destructor.

Book excerpt: Honey bees suffer from parasitism from Varroa destructor, which spreads between colonies via several transmission routes. Mites infest the brood cells of developing bees, feeding on and transmitting pathogens to their hosts. When mites are inside capped brood cells, they are protected from beekeeper applied acaricides. In this study, I used indoor storage to halt brood production, with the goal of reducing the protective brood cells and force mites into a phoretic state to increase susceptibility to treatment. I tested the efficacy of a novel strain of an entomopathogenic Metarhizium fungus as a mite control method against oxalic acid (OA). I observed that indoor storage effectively reduced the quantity of capped brood. However, I did not observe a proportional associated increase in phoretic mite population. Metarhizium reduced Varroa populations at similar rates when compared with OA, but neither OA nor Metarhizium performed significantly better after indoor storage compared to outdoor storage. This research shows promise for novel tools to reduce mite infestations and combat pesticide resistance when treating V. destructor.

They are important crop pollinators; contributing $15-20 billion to the agricultural economy in the United States. Many factors can lead to a decline in these utilitarian insects, and Varroa destructor, an ectoparasitic mite (Anderson & Trueman) is one of the most important worldwide. This pest, feeds on fat body tissue of honey bees, and cause collapse of untreated colonies within two years. Although beekeepers have used acaricides to control these mites, they continue to face resistance problem to the chemicals. Emerging acaricide resistance in varroa mites have compelled beekeepers to explore alternative ways to control varroa mites. The effectiveness of three non-acaricidal approaches to control varroa mites were tested under laboratory conditions. Varroa mites used for testing collected from the research beehives in Moscow, Idaho, USA. We tested application of spores of Metarhizium brunneum at concentrations of 5.7 x 10^4, 2.24 x 10^5, 5.05 x 10^5, 9.2 x 10^5, and 2.05 x 10^6 viable conidia in per ml−1, compared with oxalic acid and thymol. Materials were sprayed with a Potter Precision Spray Tower machine (Burkard Scientific, UK) and evaluated for their effectiveness on varroa mite mortality. The same number of varroa mites for each objective was used and recorded daily mite mortalities. 3.0% solution of oxalic acid and thymol killed varroa mites quickly than entomopathogenic fungi in 48h. No significant differences were observed in the virulence of mites between fungal concentrations. For the second experiment, 1.5% solution of oxalic acid and 10^4, 10^5, 10^6, 10^7 and 10^8 concentrations of Metarhizium brunneum were applied by mixing (500 μl oxalic acid and 500 μl M. brunneum). The optimal effect on varroa mite mortality was observed for the mixture solution oxalic acid with 10^8, 10^7 and 10^6 concentrations of M. brunneum. There was a noticeable difference between the mixture solutions with 10^8, 10^7 and 10^6 concentration of M. brunneum and 1.5% solution of pure oxalic acid application in 24h. The effectiveness of 105 and 104 mixture solutions were lower than other solutions.
