Available in PDF, EPUB and Kindle. Book excerpt: With its focus on drugs so recently introduced that they have yet to be found in any other textbooks or general references, the information and insight found here makes this a genuinely unique handbook and reference. Following the successful approach of the previous volumes in the series, inventors and primary developers of successful drugs from both industry and academia tell the story of the drug's discovery and describe the sometimes twisted route from the first drug candidate molecule to the final marketed drug. The 11 case studies selected describe recent drugs ranging across many therapeutic fields and provide a representative cross-section of present-day drug developments. Backed by plenty of data and chemical information, the insight and experience of today's top drug creators makes this one of the most useful training manuals that a junior medicinal chemist may hope to find. The International Union of Pure and Applied Chemistry has endorsed and sponsored this project because of its high educational merit.

As a comprehensive guide to this rapidly growing area of research, it covers the structure and characteristic properties of contractile and regulatory proteins, with special emphasis on their predicted function in the live muscle. Also included in this book are intermediate filament proteins, and desmin and vimentin, whose function in smooth muscle is unknown; and several enzymes involved in the phosphorylation-dephosphorylation of contractile and other proteins.

Available in PDF, EPUB and Kindle. Book excerpt: In recent years, CD38 has emerged from a simple differentiation marker to a multifunctional cell surface protein. It can serve as an ectoenzyme that catalyzes the synthesis and hydrolysis of a Ca2+mobilizing agent, cyclic ADP-ribose. Since intracellular calcium plays a key role in cell signaling pathways leading to cell growth, apoptosis, and differentiation, the enzymatic functions of CD38 may contribute to an array of immunoregulatory functions. Ligation of CD38 with agonistic antibodies induces diverse effects in hematopoietic cells which range from growth stimulation to induction and prevention of apoptosis, induction of cytokines, and activation of certain kinases. Based on these observations a ligand for CD38 was recently identified and characterized. The natural ligand is capable of transducing signals similar to the agonistic antibody, following its interaction with cell surface CD38. Recent reports have suggested an association of CD38 in diverse human pathologies such as HIV, chronic B lymphocytic leukemia, myeloma, lymphoma, diabetes, and retinoic acid syndrome. This book offers the first single source on CD38 antigen and related molecules for new and experienced investigators. It will allow them to pose new and right questions in an attempt to elucidate the biological relevance and significance of these molecules in cancer, immunology, pharmacology and biochemistry.

Book excerpt: Sphingolipids are fundamental to the structures of cell membranes, lipoproteins, and the stratum cornea of the skin. Many complex sphingolipids, as well as simpler sphingoid bases and derivatives, are highly bioactive as extra- and intracellular regulators of growth, differentiation, migration, survival, senescence, and numerous cellular responses to stress. This book reviews exciting new developments in sphingolipid biology/sphingolipidology that challenge our understanding of how multicellular organisms grow, develop, function, age, and die.

Available in PDF, EPUB and Kindle. Book excerpt: Twenty-five years have passed since J. MILLER'S basic work on the central role of the thymus for the generation of immunological reactivi ty. During this time, the abundance of fascinating literature on the immunological and functional aspects of this organ has been in contrast to the paucity of reports on its role in pathology. The causal or formal pathogenesis even of many of the well-documented pathological fea tures is so far unexplained or at least uncertain. In spite of some concep tual progress related to the work of CASTLEMAN, LEVINE, and ROSAI, we regrettably have to say that in pathology the thymus remains almost as inconspicuous as 25 years ago. Only the new techniques developed in recent years have made it possible to look as closely into the complicated structural organization of the thymus as it appears to be necessary to observe and document abnormalities. Major steps have been taken with the advent of mono clonal antibody immune histochemistry and with detailed ultrastructur al studies in embryology. At this point, pathologists and researchers with a special interest in the thymus were asked to give overviews of their respective fields of interest in light of recent findings in immunology and basic insights into the structural-functional interrelationship of the human thymus. The results of this initiative have been brought together in this volume.

Book excerpt: This much-needed book is the first definitive volume on Euglena in twenty-fire years, offering information on its atypical biochemistry, cell and molecular biology, and potential biotechnology applications. This volume gathers together contributions from well-known experts, who in many cases played major roles in elucidating the phenomenon discussed. Presented in three parts, the first section of this comprehensive book describes novel biochemical pathways which in some instances have an atypical subcellular localization. The second section details atypical cellular mechanisms of organelle protein import, organelle nuclear genome interdependence, gene regulation and expression that provides insights into the evolutionary origins of eukaryotic cells. The final section discusses how biotechnologists have capitalized on the novel cellular and biochemical features of Euglena to produce value added products. Euglena: Biochemistry, Cell and Molecular Biology will provide essential reading for cell and molecular biologists with interests in evolution, novel biochemical pathways, organelle biogenesis and algal biotechnology. Readers will come away from this volume with a full understanding of the complexities of the Euglena as well as new realizations regarding the diversity of cellular processes yet to be discovered.

It explores the metabolic requirements of the infectious agents, the microbial metabolic pathways that are dedicated to circumvent host immune mechanisms as well as the molecular mechanisms by which pathogens hijack host cell metabolism for their own benefit. Finally, it provides insights on the possible clinical and immunotherapeutic applications, as well as on the available experimental and analytical methods. The contributions break new ground in understanding the metabolic crosstalk between host and pathogen.

Available in PDF, EPUB and Kindle. Book excerpt: I entered the gene therapy field in the mid-1990s, being fascinated by the immense potential of genes as drugs for the treatment of human disease. Since then, I have experienced the ups and downs of this discipline, and tried to contribute with my work and that of my laboratory to the development of innovative approaches to the treatment of cardiovascular disorders. During these years, I have had several opp- tunities to speak on gene therapy at lectures and academic lessons, and have often noticed that the field is very attractive to scientists of all disciplines. However, as yet no comprehensive book on the subject has been published. Indeed, most books in the field are either a collection of gene transfer laboratory protocols or deal with the subject in a rather superficial manner. Hence the idea to write a gene therapy textbook that is broad and comprehensive, but at the same time provides sufficient molecular and clinical detail to be of interest to students, professors, and specialists in the various disciplines that contribute to gene therapy. I have tried to keep the language plain and, whenever possible, non-technical. Since the book is intended to be a textbook in the field of gene therapy in both the basic science and clinical areas, whenever technical descriptions are required, they are provided.
