Book excerpt: Since the pioneering discovery of cyclic AMP four decades ago, a multitude of signaling pathways have been uncovered in which an extracellular signal (first messenger) impacts the cell surface, thereby triggering a cascade that ultimately acts on the cell nucleus. In each cascade the first messenger gives rise to the appearance of a second messenger such as cyclic AMP, cyclic GMP, or diacylglycerol, which in turn triggers a third messenger, a fourth messenger, and so forth. Many advances in elucidating such pathways have been made, including efforts to link messenger molecules to brain processes operative in health or disease. However, the latter type of information, relating signaling pathways to brain function, is scattered across a variety of publication media, which makes it difficult to integrate the multiple roles of different signaling cascades into our understanding of brain function in health and disease. The primary aim of Cerebral Signal Transduction: From First to Fourth Messengers, therefore, is to offer a comprehensive picture of the recent advances made in the signaling field as it relates to neuronal and cere bral function. The current state of progress provides an exciting opportunity for such a comprehensive focus because molecular tools have become available to selectively remove, reduce, or enhance spe cific components in the signaling pathways, e. g. , by interfering with the genes encoding key proteins. In addition, the increased awareness of crosstalk between different signaling cascades has revealed many possibilities for changes in gene expression underlying long-term changes in brain function.

All mental phenomena, from basic sensory-motor to higher abstract-verbal, are the result of neural encoding of the external world signals and internal bodily signals into representations constituting the model of reality for the purpose of controlling the body and adapting to this reality. Thus, any theory of the brain faces the question of the nature of the neural code which could explain the observed speed and efficiency, scope and complexity of the computational process that we call the Mind. The mainstream theories of neuroscience that consider neural activity as trains of discrete identical spikes (various firing rate coding and temporal coding models) contradict the reality of the information density of neural computing. That is why, despite huge efforts by generations of researchers, this approach did not lead to deciphering the neural code. We know the details of the neural processes down to the molecular level but the brain remains a ‘black box’ that we cannot read. It is the outcome of the wrong theoretical assumption that should be revised. The author creates the concept of a neural code that overcomes the shortcomings of old models. There is another problem that cannot be avoided by any theory of consciousness. It is not enough to say that the brain creates the psyche as this leaves an explanatory gap. We need to show how this physiological system generates mental phenomena physically. The Mind is a technological process that works according to a certain algorithm based on physical laws. The author’s theory offers a fundamentally new approach to the nervous system that bridges physiology and psychology by illuminating the algorithm and the physics of the Mind from the intracellular to the system-wide level.

The resistance status of malignancy to treatment is determined by an inextricable connection between P-gp expression in malignant cell membranes exposed to drugs, blood-brain barrier developmental cells in human foetal cells, and ALL blood cells undergoing chemotherapy. A new type of concept is introduced here as a human P-gp resistance amalgamation (P-gp-MDR) is conceived. Multidrug resistance is created due to cellular and barrier programs through the expression of a membrane drug efflux pump with an ability to create downstream signal transduction systems and which regulates its function and activity. This includes cellular and barrier capacities to generate unforeseen, innovative signalling pathways on multiple levels that can jeopardise treatment pathways at key stages in a fatal way. At present, these systems were unrecognised as potential elements for targeting the killing of deadly malignancies, both in primary and secondary forms. The fact that the blood-brain barrier is such a robust, intelligent entity, interfaced between blood and brain, indicates that it is the essential guardian for the survival of the brain, and thus all human biological systems depend on it. Its expression is seen in developing foetal brain tissues. However, it is also a stumbling block to the eradication of malignant cells. These findings of inducible P-gp expression in cancer cells and those that will constitute the blood-brain barrier creates intense tension both in the malignant systems as well as in the natural systems surrounding them, raising questions as to how to intervene to preserve the normal and retain its autonomy whilst destroying destructive actions, like hijackings by the malignant cells; the source of these abnormal responses remains to be answered. This book dynamically argues that P-gp MDR 1 and MDR 3 and the expressive Pg-p brain-blood functions are necessary to address for new chemotherapeutic challenges. It is important to study in general and specific, intelligent malignant, and natural systems that create multidrug resistance to treatment. The creativity and actions of P-gp and its polymorphisms are distinct in natural cells and malignant cells, and P-gp and its polymorphisms are involved in the creation of acute multidrug resistance, and they act as deadly programmed protectors of the biological environments of malignant cells and the cells exposed to drug treatments. As such the structure and function of P-gp together constitutes fatal ends for the host and its systems - based on studies of human cell lines, in vitro systems, in vivo foetal systems, in vivo blood cell systems - and exhibits non-cooperation of cells to multidrug treatments. In short, ALL is a multifaceted disease and displays abnormal activation of signalling pathways, which in turn activate drug-resistance pumps. The ability to target elements that activate the drug transporter P-gp will then be key to success to overcome cancers in this cutting edge field of nano-micro-molecular genetics. Examples of phytochemicals include catechins, resveratrol, ginkgo biloba, curcumin, and sulfur compounds found in garlic. Although, the precise molecular mechanisms associated with beneficial effects of phytochemicals still remain the subject of intense investigations, but it is becoming increasingly evident that phytochemicals mediate their effects by counteracting, reducing, and repairing the damage caused by oxidative stress and neuroinflammation. In addition, phytochemicals also stimulate the synthesis of adaptive enzymes and proteins through the stimulation of a transcription factor called Nrf2 and induction of phase II detoxifying enzymes. Consumption of phytochemicals induces neurohormetic response that results in the expression of adaptive stress-resistance genes that are responsible for encoding antioxidant enzymes, protein chaperones, and neurotrophic factor (BDNF).
