Available in PDF, EPUB and Kindle. Book excerpt: In the summer of 1988, my developmental biology professor announced to the class that hematopoietic stem cells (HSCs) had finally been purified. Somehow, I never forgot the professor’s words. When I started working in Dr. Irv Weissman’s labo- tory at Stanford as a postdoctoral fellow, I realized that the findings mentioned by the professor were from Weissman’s laboratory and had been published in a 1988 edition of the journal Science. It has been over 20 years since the publication of that seminal paper, and since then tremendous advances in understanding the biology and maturation of HSCs, namely the process of hematopoiesis, which includes lymphocyte development, have been made. These discoveries were made possible in part by advancements in technology. For example, recent availability of user friendly fluorescence activated cell sorting (FACS) machines and monoclonal an- bodies with a variety of fluorescent labels has allowed more scientists to sort and analyze rare populations in the bone marrow, such as HSCs. All classes of hematopoietic cells are derived from HSCs. Stem cell biology draws enormous attention not only from scientists, but also from ordinary people because of the tremendous potential for development of new therapeutic application to diseases that currently lack any type of effective therapy. Thus, this type of “regenerative medicine” is a relatively new and attractive field in both basic science and clinical medicine.

Book Synopsis Stem Cells: A Cellular Fountain of Youth by : M.P. Mattson
Download or read book Stem Cells: A Cellular Fountain of Youth written by M.P. Mattson and published by Elsevier. This book was released on 2002-08-21 with total page 240 pages. Available in PDF, EPUB and Kindle. Book excerpt: The developmental capabilities and therapeutic potential of stem cells are being revealed in studies of cellular signaling mechanisms that regulate their proliferation, differentiation and survival. "Stem Cells: A Cellular Fountain of Youth" reviews the current state of understanding of the molecular mechanisms that regulate embryonic and adult stem cells with an emphasis on how aging and age-related disease impact on these mechanisms. Leading authorities detail the properties and therapeutic potential of embryonic stem cells, and stem cell precursors of blood, nervous and muscle and bone cells. Recent advances in deciphering the environmental signals and intrinsic signal transduction pathways that regulate embryonic stem cells are described, and the potential therapeutic uses of these totipotent cells is considered. Analyses of hematopoietic stem cells during aging suggest an important genetic component to the control of their self-renewing capability which may contribute to determination of lifespan. The contribution of lymphocyte depletion to impaired immune function during aging is considered, as is the potential of hematopoietic cells to form other types of cells including neurons. Several chapters cover the remarkable and rapidly advancing field of neural stem cells. The adult brain contains populations of stem cells capable of forming new neurons and glial cells; the signals that regulate these neural stem cells and the involvement of neurogenesis in normal brain function is described. Because of their potential to replace lost or damaged neurons, there has been intense interest in determining the therapeutic potential of stem cells for the treatment of patients with Parkinson's and Alzheimer's diseases, stroke and traumatic brain and spinal cord injuries. Heart and skeletal muscle contain stem cells and the impact of aging and disease on these stem cell populations and the potential of stem cell therapy to recover function of these organs is reviewed. A final example of the fascinating world of stem cells is a review of the roles of stem cells in bone formation and remodeling. Collectively, this book provides a comprehensive, yet concise, view of stem cell molecular biology in the context of aging and age-related disease. This book will be a valuable reference for graduate students and senior scientists interested in the fascinating world of stem cells and their potential use in the clinic.
Book Synopsis Stem Cells & Regenerative Medicine by : Krishnarao Appasani
Download or read book Stem Cells & Regenerative Medicine written by Krishnarao Appasani and published by Springer Science & Business Media. This book was released on 2010-11-01 with total page 632 pages. Available in PDF, EPUB and Kindle. Book excerpt: Defined as, “The science about the development of an embryo from the fertilization of the ovum to the fetus stage,” embryology has been a mainstay at universities throughout the world for many years. Throughout the last century, embryology became overshadowed by experimental-based genetics and cell biology, transforming the field into developmental biology, which replaced embryology in Biology departments in many universities. Major contributions in this young century in the fields of molecular biology, biochemistry and genomics were integrated with both embryology and developmental biology to provide an understanding of the molecular portrait of a “development cell.” That new integrated approach is known as stem-cell biology; it is an understanding of the embryology and development together at the molecular level using engineering, imaging and cell culture principles, and it is at the heart of this seminal book. Stem Cells and Regenerative Medicine: From Molecular Embryology to Tissue Engineering is completely devoted to the basic developmental, cellular and molecular biological aspects of stem cells as well as their clinical applications in tissue engineering and regenerative medicine. It focuses on the basic biology of embryonic and cancer cells plus their key involvement in self-renewal, muscle repair, epigenetic processes, and therapeutic applications. In addition, it covers other key relevant topics such as nuclear reprogramming induced pluripotency and stem cell culture techniques using novel biomaterials. A thorough introduction to stem-cell biology, this reference is aimed at graduate students, post-docs, and professors as well as executives and scientists in biotech and pharmaceutical companies.

Download or read book Hematopoiesis written by Leonard I. Zon and published by Oxford University Press. This book was released on 2001-04-12 with total page 864 pages. Available in PDF, EPUB and Kindle. Book excerpt: Hematopoiesis, or the process of blood formation, has been extensively studied at both basic and clinical levels. Human diseases such as thalassemia, immunodeficiency, and leukemia represent defects in this process. Approaches to treat these disorders have required a basic understanding of the biology of blood cells. For instance, hemapoietic stem cell replacement or bone marrow transplantation has been used to ameliorate disease. This volume focuses on hematopoiesis at a cellular and molecular level, and establishes the basis for clinical manipulation of hematopoietic cells for therapeutic benefit. In Part I, the cellular characteristics of progenitors and stem cells are explored. Emphasis is placed on purification of stem cells and both in vitro and in vivo assays. The regulation of normal and leukemis stem cells is illustrated. An excellent discussion of potential use of these cells for gene therapy concludes this section. Hemapoiesis is easily studied during embryogenesis. Part II develops the concept of the waves of hemapoiesis during development. Comparative hematology is making a major comeback as a field in the 1990's. One hope is that general principles of hematopoiesis will be established by studying many models and systems. Part III delves into critical factors that regulate hematopoiesis, including both intracellular and extracellular signals. Part IV and V describe lineage programs for myeloid and lymphoid lineages. These chapters are meant to be illustrative of the different cell fates, but are not exhaustive. Part VI examines the genetics of hematopoisis, particularly in animal models. The hematopoietic system is in constant contact with stromal cells and endothelial cells during development and in the adult. Evidence suggests that endothelial cells and blood cells may arise from a common progenitor, the hemangioblast. Part VII and VIII discuss the stromal and endothelial cells with the emphasis on their interaction with hematopoietic cells.
