Book excerpt: Developmental biology deals with the regulatory principles underlying the generation of a new organism. The up-to-date knowledge of the molecular and cellular mechanisms that guide mammalian development from a single cell through a complex and integrated process of cell multiplication, differentiation and organisation of cells into tissues and organs is presented in this volume. Aspects of the molecular nature and developmental significance of cell-to-cell signals focus on: - hoxgenes, their products and potential role in mammalian morphogenesis; - growth factors, their receptors and signal transduction, and their role in guiding cell multiplication, differentiation and transformation; and - cell adhesion and extracellular matrix molecules, as short-range cell-to-cell signals in mammalian development.

Book excerpt: The book considers signaling events from the zygote embryo through to the blastocyst with relevant data from embryonic stem (ES) cells, including dialogue with the extracellular environment and with the maternal tract during the implantation process. Application of the knowledge described to improve the success of human and animal assisted conception is considered where appropriate, but the focus is largely on fundamental rather than applied cell/molecular biology, as this is the area that has historically been neglected. While the general features of metabolism during preimplantation development are well established, especially in terms of nutrient requirements, uptake and fate, remarkably little is known about early embryo signaling events, intracellular or intercellular, between individual embryos in vitro or with the female reproductive tract in vivo. This contrasts with the wealth of information on cell signaling in somatic cells and tissues, as a glance at any textbook of biochemistry illustrates. This lack of information is such that our understanding of the molecular cell biology of early embryos -- a prerequisite to defining the mechanisms which regulate development at this critical stage of the life cycle -- is seriously incomplete. This volume is the first to address this issue by describing the current state of knowledge on cell signaling during mammalian early embryo development and highlighting priority areas for research.

Available in PDF, EPUB and Kindle. Book excerpt: Cell-Cell Signaling in Vertebrate Development provides a comprehensive discussion of cell-cell interactions in vertebrate development and the molecular signals that mediate them. The book is divided into six parts, arranged according to major developmental phenomena demonstrated in illustrative systems derived from amphibian, avian, mammalian, and piscine sources. Part I introduces the mechanisms of gene activation in the context of early vertebrate development. Part II is concerned with cellular contacts and the induction process. Cell-cell interactions are illustrated through analyses of neurogenesis in the mouse; embryonic induction is considered in the frog and in the chick. Part III deals with cell migration and differentiation. It examines cell lineages in the frog eyebud; migration phenomena in connection with axon guidance in the embryonic rat spinal cord and mouse visual system; pathfinding by primary motoneurons; and the formation of terminal arbors in zebrafish embryos. Part IV discusses developmental processes that depend on diffusible signals and signal gradients. Part V illustrates pattern formation as exemplified in the developing chick hindbrain and in urodele limb regeneration. Part VI highlights gene expression and its regulation by transcription factors or growth factors in rodent development.

The players in signal transduction are diverse, from small molecules as first messengers, to proteins, receptors, transcription factors, among many others. The different signaling pathways and the crosstalk between them originates the unique signaling profile of every cell type in the human body. The cell signaling specificity depends on several aspects including protein composition, subcellular localization and complexes and gene promoters. This textbook provides a comprehensive overview of the specific signaling pathways on a variety of human tissues. This information can be of great value for health science researchers, professionals and students to understand key pathways for tissue-specific functions in the plethora of signals, signals receptors, transducers and effectors. Emphasis is placed on the insulin family of peptides, including insulin and the structurally and functionally related insulin-like growth factors. The initial chapters provide a review of basic topics, including developmental genetics, energy metabolism and hormonal signaling mechanisms, which are important prerequisites to the central theme that follows. The book concludes with a brief review of oncogene expression in early development; this new field has contributed significantly to our understanding of how mitogenic signals activate genetic elements responsible for embryonic growth and development. This book presents information important to cell biologists, endocrinologists, biological chemists, and developmental biologists.

Available in PDF, EPUB and Kindle. Book excerpt: Developmental biologists have been driven to investigate growth factor signaling in embryos in order to understand the regulatory mechanisms underlying a given developmental process. Thus, it is critical to explore the technical methods and experimental designs for growth factor signaling in embryos. Focusing on specific pathways or pathway components, Analysis of Growth Factor Signaling in Embryos provides the methods and guidelines for experimental design to study major aspects of cell signaling in vertebrate embryos. The book covers a broad range of topics in signaling and a variety of current model organisms. Section I explores specific signaling pathways or pathway components. In this section, some chapters highlight the biochemistry of signaling pathways during development, which is often distinctive from that observed in cell culture systems. Section II discusses ionic regulatory mechanisms and the two chapters in Section III examine ways of investigating gene regulation in response to extracellular signals. Finally, Section IV addresses emerging strategies that facilitate integrated analyses of cell signaling in vivo in embryonic systems. Featuring contributions from expert researchers, Analysis of Growth Factor Signaling in Embryos will provide a foundation for further explorations of the cellular regulatory mechanisms governing vertebrate embryonic development.

Book excerpt: Because of the recent advances in embryo modeling techniques, and at the request of the Office of Science Policy in the Office of the Director at the National Institutes of Health, the National Academies of Sciences, Engineering, hosted a 1-day public workshop that would explore the state of the science of mammalian embryo model systems. The workshop, which took place on January 17, 2020, featured a combination of presentations, panels, and general discussions, during which panelists and participants offered a broad range of perspectives. Participants considered whether embryo model systems - especially those that use nonhuman primate cells - can be used to predict the function of systems made with human cells. Presentations provided an overview of the current state of the science of in vitro development of human trophoblast. This publication summarizes the presentation and discussion of the workshop.
