Book excerpt: The field of cell cycle regulation is based on the observation that the life cycle of a cell progresses through several distinct phases, G1, M, S, and G2, occurring in a well-defined temporal order. Details of the mechanisms involved are rapidly emerging and appear extraordinarily complex. Furthermore, not only is the order of the phases important, but in normal eukaryotic cells one phase will not begin unless the prior phase is completed successfully. Che- point control mechanisms are essentially surveillance systems that monitor the events in each phase, and assure that the cell does not progress prematurely to the next phase. If conditions are such that the cell is not ready to progress—for example, because of incomplete DNA replication in S or DNA damage that may interfere with chromosome segregation in M—a transient delay in cell cycle progression will occur. Once the inducing event is properly handled— for example, DNA replication is no longer blocked or damaged DNA is repaired—cell cycle progression continues. Checkpoint controls have recently been the focus of intense study by investigators interested in mechanisms that regulate the cell cycle. Furthermore, the relationship between checkpoint c- trol and carcinogenesis has additionally enhanced interest in these cell cycle regulatory pathways. It is clear that cancer cells often lack these checkpoints and exhibit genomic instability as a result. Moreover, several tumor suppressor genes participate in checkpoint control, and alterations in these genes are as- ciated with genomic instability as well as the development of cancer.

Book excerpt: Cell cycle checkpoints are the means by which cells maintain their genomic integrity and resist factors that are potentially mutagenic. It is well known that there are different checkpoints for different stresses a cell could face, one of them being the DNA damage checkpoint. For long the activity of this checkpoint was thought to be limited to the nucleus. In this and other emerging studies it has now been established that the DNA damage checkpoint can regulate cell cycle arrest, nuclear positioning and movement by controlling cytoplasmic factors and events. This study establishes a novel regulatory mechanism of the DNA damage checkpoint involving vesicular transport system and cytoplasmic, non-proteosomal protein degradation. The study also focuses on the connection between the spindle assembly checkpoint (SAC) and the DNA damage checkpoint. To study this we generate an acentric chromosome with a single DNA double strand break on it. We find that budding yeast cells that have one chromosome with conditional GAL1-Cen centromere turned off cannot activate the SAC and both the sister chromatids of this chromosome get preferentially segregated to the mother cell.

Book excerpt: Checkpoint signaling and DNA repair are closely connected processes that protect the cell's genetic material from exogenous and endogenous stress and are collectively referred to as the DNA damage response (DDR). Relocalization of checkpoint proteins to chromatin flanking DNA double-strand breaks (DSBs) is critical for DDR. Schizosaccharomyces pombe Crb2, which mediates Chk1 activation by Rad3 ATR, forms ionizing radiation-induced nuclear foci (IRIF) at DSBs. Crb2 C-terminal BRCT domains (BRCT 2 ) bind histone H2A phosphorylated by Tel1 ATM and Rad3 ATR ([gamma]-H2A). This work proposes that cooperative phosphate interactions in the BRCT 2 [gamma]-H2A-binding pocket of Crb2, coupled with Tudor domain interactions with lysine-20 dimethylation of histone H4, facilitate stable recruitment of Crb2 DSBs, which in turn mediates efficient phosphorylation of Chk1. This mechanism of cooperative interactions with the [gamma]-H2A/X phosphate is likely conserved in S. pombe Brc1 and human MDC1 genome maintenance proteins.
