Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

Book excerpt: This publication sets outs the findings of an IARC Working Group, held in Lyon, France in November 2003, which considered the benefits of a diet rich in cruciferous vegetables in helping to reduce the risk of various cancers. Cruciferous vegetables, such as broccoli, cabbage, cauliflower, watercress and brussel sprouts, contain substantial amounts of compounds which have been shown to inhibit the growth of cancers. This publication reviews current knowledge on the topic, including data from human, experimental and mechanistic studies, as well as making recommendations for future research and public health policy options.

Available in PDF, EPUB and Kindle. Book excerpt: Cancer care today often provides state-of-the-science biomedical treatment, but fails to address the psychological and social (psychosocial) problems associated with the illness. This failure can compromise the effectiveness of health care and thereby adversely affect the health of cancer patients. Psychological and social problems created or exacerbated by cancer-including depression and other emotional problems; lack of information or skills needed to manage the illness; lack of transportation or other resources; and disruptions in work, school, and family life-cause additional suffering, weaken adherence to prescribed treatments, and threaten patients' return to health. Today, it is not possible to deliver high-quality cancer care without using existing approaches, tools, and resources to address patients' psychosocial health needs. All patients with cancer and their families should expect and receive cancer care that ensures the provision of appropriate psychosocial health services. Cancer Care for the Whole Patient recommends actions that oncology providers, health policy makers, educators, health insurers, health planners, researchers and research sponsors, and consumer advocates should undertake to ensure that this standard is met.

Book excerpt: A working group of sixteen experts from seven countries re-evaluated the evidence of the carcinogenicity of betel-quid and areca-nut chewing and some areca-nut related nitrosamines. Betel-quid and areca-nut chewing are widely practised in many parts of Asia and in Asian-migrant communities elsewhere in the world. There are hundreds of millions of users worldwide. They evaluated betel quid with tobacco as carcinogenic to humans (Group 1) on the basis of sufficient evidence of an increased risk of cancer of the oral cavity, pharynx and oesophagus. The working group reviewed epidemiological studies of human cancer, mainly studies from India, Pakistan and Taiwan (China). Studies on betel quid with tobacco and areca nut with tobacco in experimental animals now also provide sufficient evidence of carcinogenicity. The working group also evaluated betel quid without tobacco as carcinogenic to humans (Group 1), on the basis of sufficient evidence of an increased risk of oral cancer. Studies on betel quid without tobacco and areca nut without tobacco in experimental animals now also provide sufficient evidence of carcinogenicity. Areca nut, a common ingredient of betel quid and many different chewing preparations, including those available commercially, has been observed to cause oral submucous fibrosis

The result is a gripping narrative that reveals why the public's immense investment in research has been badly misspent, why scientists seldom collaborate and share their data, why new drugs are so expensive yet routinely fail, and why our best hope for progress-- brilliant young scientists-- are now abandoning the search for a cure.

Available in PDF, EPUB and Kindle. Book excerpt: This Surgeon General's report returns to the topic of the health effects of involuntary exposure to tobacco smoke. The last comprehensive review of this evidence by the Department of Health and Human Services (DHHS) was in the 1986 Surgeon General's report, The Health Consequences of Involuntary Smoking, published 20 years ago this year. This new report updates the evidence of the harmful effects of involuntary exposure to tobacco smoke. This large body of research findings is captured in an accompanying dynamic database that profiles key epidemiologic findings, and allows the evidence on health effects of exposure to tobacco smoke to be synthesized and updated (following the format of the 2004 report, The Health Consequences of Smoking). The database enables users to explore the data and studies supporting the conclusions in the report. The database is available on the Web site of the Centers for Disease Control and Prevention (CDC) at http://www.cdc.gov/tobacco.

Available in PDF, EPUB and Kindle. Book excerpt: The culture of cancer cells is routinely practiced in many academic research centers, biotechnology companies, and hospital laboratories. Cancer Cell Culture: Methods and Protocols describes easy-to-follow methods to guide both novice and more experienced researchers seeking to use new techniques in their laboratories. Our present understanding of the cell and molecular biology of cancer has been derived mainly from the use of cultured cancer cells and we cover a number of the most widely used assays to study function in current use. Part I introduces the basic concept of cancer cell culture and this is followed by a description of the general techniques used in many cell culture facilities. The importance of cell line characterization is now widely recognized and methods to characterize and authenticate cell lines are described in Part II. Part III covers the isolation and development of specific cancer cell types and provides valuable tips for those wishing to derive new cell line models. A wide range of procedures encompassing many of the key functional features of cancer cells are described in Part IV including assays to evaluate clonogenicity, cell proliferation, apoptosis, adhesion, migration, invasion, senescence, angiogenesis, and cell cycle parameters. Methods to modify cancer cells are described in Part V, including protocols for transfection, development of drug-resistance, immortalization, and transfer in vivo. In Part VI methods of coculture of different cell types and contamination of cell lines are covered.

This book was released on 2004 with total page 540 pages. Available in PDF, EPUB and Kindle. Book excerpt: A working group of 23 experts from 13 countries met in Lyon to evaluate the evidence for carcinogenicity of arsenic (mostly naturally occurring) as a contaminant of drinking-water, and of the water-disinfectant chloramine. The working group also evaluated or re-evaluated four chlorination by-products found in drinking-water, namely chloral hydrate, di- and trichloroacetic acids, and 3-chloro-4-(dichloromethyl)-5-hydroxy-2(5H)-furanone (also known as MX). High-level exposure to arsenic in drinking-water occurs in some regions such as China, Latin America, Bangladesh and West Bengal. The Working Group reviewed epidemiological studies of human cancer (mainly ecological studies in Taiwan and Chile, and several case-control and cohort studies) in relation to arsenic in drinking-water. Arsenic in drinking-water (primarily inorganic, as arsenate and to a lesser extent arsenite) was evaluated as carcinogenic to humans (Group 1) on the basis of sufficient evidence for an increased risk for cancer of the urinary bladder, lung and skin. Studies on inorganic arsenic in experimental animals provided limited evidence for its carcinogenicity, but sufficient evidence was found in experimental animals for the carcinogenicity of dimethylarsinic acid (an organic form of arsenic), which produced urinary bladder tumours in rats and lung tumours in mice after oral administration.

Book excerpt: "It began with a promising cancer drug, the brainchild of a gifted researcher, and grew into an insider trading scandal that ensnared one of America's most successful women. The story of ImClone Systems and its "miracle" cancer drug, Erbitux, is the quintessential business saga of the late 1990s. It's the story of big money and cutting-edgescience, celebrity, greed, and slipshod business practices; the story of biotech hype and hope and every kind of excess. At the center of it all stands a single, enigmatic figure named Sam Waksal. A brilliant, mercurial, and desperate-to-be-liked entrepreneur, Waksal was addicted to the trappings of wealth and fame that accrued to a darling of the stock market and the overheated atmosphere of biotech IPOs. At the height of his stardom, Waksal hobnobbed with Martha Stewart in New York and Carl Icahn in the Hamptons, hosted parties at his fabulous art-filled loft, and was a fixture in the gossip columns. He promised that Erbitux would "change oncology," and would soon be making $1 billion a year. But as Waksal partied late into the night, desperate cancer patients languished, waiting for his drug to come to market. When the FDA withheld approval of Erbitux, the charming scientist who had always stayed just one step ahead of bankruptcy panicked and desperately tried to cash in his stock before the bad news hit Wall Street. Waksal is now in jail, the first of the Enron-era white-collar criminals to be sentenced. Yet his cancer drug has proved more durable than his evanescent profits. Erbitux remains promising, the leading example of a new way to fight cancer, and patients and investors hope it will be available soon.
