Some fields like basic biochemistry, organic reaction mechanisms, and chemical thermody namics are well represented by many excellent texts, and new or revised editions are published sufficiently often to keep up with progress in research. However, some areas of chemistry, especially many of those taught at the graduate level, suffer from a real lack of up-to-date textbooks. The most serious needs occur in fields that are rapidly changing. Textbooks in these subjects usually have to be written by scientists actually involved in the research which is advancing the field. It is not often easy to persuade such individuals to set time aside to help spread the knowledge they have accumulated. Our goal, in this series, is to pinpoint areas of chemistry where recent progress has outpaced what is covered in any available textbooks, and then seek out and persuade experts in these fields to produce relatively concise but instructive introductions to their fields. These should serve the needs of one semester or one quarter graduate courses in chemistry and biochemistry. In some cases, the availability of texts in active research areas should help stimulate the creation of new courses.

The first chapter comes from a laboratory that was one of the first to visualize the distribution of transplantation antigens on cell membrane surfaces. Various methods are described for visualizing these antigens by electron microscopy. Davis and his colleagues then proceed to show how the antibody-induced redistribution of antigenic macromolecules led to the formulation of the fluid mosaic model. From Hakomori's labora tory comes a methodological paper describing a novel method of labeling the carbohydrate portions of the membrane glycoproteins that are exposed on the outer surfaces of cells. The two chapters reviewing the changes found on carcinogen-induced and virus-induced malignant cells complete the survey of the structures associated with surface mem branes. Other methods, by their very nature, apply to organized bilayers comprised of both protein and lipid. Van Meer and van Genderen provide us with an introduction to the field (Chapter I). From their personal perspective regarding the distribution, trans port, and sorting of membrane lipids, they formulate a number of biologically relevant questions and show that the physicochemical methods described in this book may contribute in great measure to solving these issues. The methods of analytical ultracentrifugation have served faithfully for 60 years in the study of water-soluble proteins. The use of detergent extraction of membrane proteins, and the manipulation of density with H20/D20 mixtures, has extended this technique to the study of proteins, and in particular their interactions, from biological membranes. As described by Morris and Ralston in Chapter 2, this technique can be used to determine a number of important properties of proteins.

The symposium brought together 80 of the leading investigators concerned with ATP-utilizing and ATP-generating systems associated with cation fluxes across membranes to discuss biochemical mechanisms in depth and their relation to cation transport functions. The papers presented focused on three types of membrane systems. The first two membrane systems are classified as ATP-utilizing systems. These include the plasma membrane, associated with the ATP dependent Na+-K+ transport system, which draws upon most of the cell's energy for cation fluxes; and the sarcoplasmic recticulum membrane associated with Ca++ transport, which plays a key role in excitation-contraction coupling in muscle. The third type of membrane system falls under ATP-generating systems. These include the inner membranes of mitochondria, chloroplasts, and bacteria associated with H+ fluxes generated by oxidation-reduction reactions, and their coupling to secondary ion flows and oxidative and photosynthetic phosphorylation. H+ transport associated with the photoreaction cycle of bacteriorhodopsin, the light energy converted in halobacteria was also considered.
