Book excerpt: From within complex structures of organisms and cells down to the molecular level, biological processes all involve movement. Muscular fibers slide on each other to activate the muscle, as polymerases do along nucleic acids for replicating and transcribing the genetic material. Cells move and organize themselves into organs by recognizing each other through macromolecular surface-specific interactions. These recognition processes involve the mu tual adaptation of structures that rely on their flexibility. All sorts of conformational changes occur in proteins involved in through-membrane signal transmission, showing another aspect of the flexibility of these macromolecules. The movement and flexibility are inscribed in the polymeric nature of essential biological macromolecules such as proteins and nucleic acids. For instance, the well-defined structures formed by the long protein chain are held together by weak noncovalent interac tions that design a complex potential well in which the protein floats, permanently fluctuating between several micro- or macroconformations in a wide range of frequencies and ampli tudes. The inherent mobility of biomolecular edifices may be crucial to the adaptation of their structures to particular functions. Progress in methods for investigating macromolecular structures and dynamics make this hypothesis not only attractive but more and more testable.

Book excerpt: This volume is a collection of the contributions presented at the 42nd Erice Crystallographic Course whose main objective was to train the younger generation on advanced methods and techniques for examining structural and dynamic aspects of biological macromolecules. The papers review the techniques used to study protein assemblies and their dynamics, including X-ray diffraction and scattering, electron cryo-electron microscopy, electro nanospray mass spectrometry, NMR, protein docking and molecular dynamics. A key theme throughout the book is the dependence of modern structural science on multiple experimental and computational techniques, and it is the development of these techniques and their integration that will take us forward in the future.

Book excerpt: Essentials of Chemical Biology Discover a detailed knowledge of concepts and techniques that shape this unique multi-discipline Chemical Biology is devoted to understanding the way that Biology works at the molecular level. This is a problem-driven multi-discipline, incorporating as it does Organic, Physical, Inorganic, and Analytical Chemistry alongside newer emerging molecular disciplines. In recent years, Chemical Biology has emerged as a vibrant and growing multi-discipline distinct from Biochemistry that is focused on the quantitative analyses of the structures and functions of biological macromolecules and macromolecular lipid assemblies, at first in isolation, then in vitro and in vivo. The second edition of the Essentials of Chemical Biology begins with a thorough introduction to the structure of biological macromolecules and macromolecular lipid assemblies, before moving on to the principles of chemical and biological synthesis, followed by descriptions of a comprehensive variety of research techniques and experimental methods. In addition, the second edition now includes new sections on the behaviour of biological macromolecules and macromolecular lipid assemblies in cells in vitro and in organisms in vivo. Given this, the second edition of the Essentials of Chemical Biology promises to cement itself as the leading introduction to Chemical Biology, incorporating descriptions of cutting-edge research wherever appropriate. Hence, readers of the second edition of the Essentials of Chemical Biology will find: a general expansion in understanding of basic molecular mechanisms in Biology moving towards cellular and organismal mechanisms entirely new chapters covering miniaturization and array technologies, Chemical Cell Biology, and the interface between Chemical Biology and Nanotechnology updates to chapters reflecting recent research developments an increased engagement with medical applications Essentials of Chemical Biology is ideal for advanced undergraduates or (post) graduate students in Chemical Biology and adjacent fields.

Book excerpt: This volume collects a number of the invited lectures and a few selected contrib utions presented at the International Symposium on Structure and Dynamics of Nucleic Acids, Proteins and Membranes held August 31st through September 5th, 1986, in Riva del Garda, Italy. The title of the conference as well as a number of the topics covered represent a continuation of two previous conferences, the first held in 1982 at the University of California in San Diego, and the second in 1984 in Rome at the Accademia dei Lincei. These two earlier conferences have been documented in Structure and Dynamics: Nucleic Acids and Proteins, edited by E. Clementi and R. H. Sarma, Adenine Press, New York, 1983, and Structure and Motion: Membranes, Nucleic Acids and Proteins, edited by E. Clementi, G. Corongiu, M. H. Sarma and R. H. Sarma, Adenine Press, New York, 1985. At this conference in Riva del Garda we were very hesitant to keep the name of the conference the same as the two previous ones. Indeed, a number of topics discussed in this conference were not included in the previous ones and even the emphasis of this gathering is only partly reflected in the conference title. An alternative title would have been Structure and Dynamics of Nucleic Acids, Proteins, and Higher Functions, or, possibly, "higher components" rather than "higher functions.

Book excerpt: Everything in a living organism relies on biological macromolecules, which have the role of enzymatic chemical transformations, formation of structures, transportation, catalysis, and regulation of biological processes. They are complex biological structures that require an atomistic understanding. A molecular understanding of biological macromolecules has had a massive impact on the pharmaceutical, biotechnological, and chemical industries. Specifically, new enzymatic structures are being discovered through various experimental and computational methods, by describing an atomistic-level insight into function, mechanism, role in reactions and their inhibition. Those atom-level illustrations are mainly focused through enzyme kinetics, enzyme inhibition, mutational and conformational analysis through quantum mechanical and molecular dynamics methods.
