Book excerpt: The fields of protein crystallography, cryo-electron microscopy and cryopreservation require that aqueous biological samples or aqueous solutions be cooled to very low temperatures. The outcomes of experiments in those fields also depend on the behavior of the water contained in such samples. This makes understanding that behavior central to such experiments, even if that understanding is not the aim of the research. In particular, samples often must be cooled so that the water is in a metastable amorphous state, i.e. so that no crystalline ice forms. This dissertation covers topics ranging from the very practical matter of how this metastable state can be achieved and maintained to more fundamental questions surrounding its formation. A cheap and simple method by which tiny samples can be cooled at very high rates is demonstrated, which will allow cooling at rates previously requiring expensive and complicated apparatus. The relationship between the cooling rate required to produce an amorphous sample and the amount of solute in the sample is demonstrated to be a simple exponential, which may indicate the fundamental mechanism by which ice forms from supercooled solution. Finally, the radiation sensitivity of protein crystals is shown to be linked with the diffusive motions of the supercooled solvent within them: radiation sensitivity is suppressed by lowering the temperature because diffusive motion is also suppressed.

The past few years have wit nessed an increasing interest in supercooled water and amorphous ice. If the properties of liquid water in the normal temperature range are already eccentric, then they become exceedingly so below the normal freezing point, in the metastable temperature range. Water can be supercooled to -39°C without too much effort, and most of its physical properties show a re markable temperature dependence under these conditions. Although ade quate explanations are still lacking, the time has come to review available knowledge. The study of amorphous ice, that is, the solid formed when water vapor is condensed on a very cold surface, is of longer standing. It has achieved renewed interest because it may serve as a model for the liquid state. There is currently a debate whether or not a close structural relation ship exists between amorphous ice and supercooled water. The nucleation and growth of ice in supercooled water and aqueous solutions is also still one of those grey areas of research, although these topics have received considerable attention from chemists and physicists over the past two decades. Even now, the relationships between degree of supercooling, nucleation kinetics, crystal growth kinetics, cooling rate and solute concentration are somewhat obscure. Nevertheless, at the empirical level much progress has been made, because these topics are of considerable importance to biologists, technologists, atmospheric physicists and gla ciologists.

Dr. Krijnen was intentionally honoured with the invitation to open this symposium on cryopreservation and low temperature biology in blood transfusion and be the guest of honour at this event. Unfortunately, Dr. Krijnen suddenly died on the first of June 1989. In honour and mem ory of Dr. Krijnen this symposium will therefore be dedicated to him. Since the lOth International Symposium on Blood Transfusion in 1985 highlighted the theme of "Future developments in blood banking", major changes have occurred in the blood banking world. Most of these changes were forced upon the Blood Banks by the fear of spreading AIDS through contaminated donations. This not only led to the wide spread testing of blood, but also to a more appropriate counselling of the community and the blood donors in specific. Additionally, virus inacti vation techniques were introduced for those components derived from multiple donations and intended for a regular transfusion in haemophi lia patients and others.
