Available in PDF, EPUB and Kindle. Book excerpt: Handbook of Microscopy is a manual that deals mainly with the basic instruments and techniques used in light microscopy and its biological applications. A large section is devoted to the study of organic matter in microfossils preserved in rocks, in view of its stratigraphic importance in mining and oil prospecting. This text is comprised of six chapters; the first of which introduces the reader to the basic principles as well as to the instruments and techniques used in light microscopy. This book also discusses the microscopes and electronic flashlights for photomicrography, along with the use of monochromatic light, stereological and physicochemical microanalysis, microanalysis by electron microscopy, and microdetermination of physical values. Attention then turns to staining and impregnation and methods of fixation, examination, cutting, and mounting. The remaining chapters focus on the microscopy of topological stains and non-specific cytological stains, with emphasis on special methods used in animal and plant histology and protistology and mycological methods in pathology. This book is written specifically for microscopists.

Book excerpt: The user This manual is designed for the use of geo-scientists with an interest and need in developing palaeobiological materials as a potential source of data. To meet this objective practical procedures have been formatted for use by both professional and semi professional students with an initial understanding of palaeo biological research aims as a primary source of scientific data. I have attempted to provide an explanation and understanding of practical procedures which may be required by students undertaking palaeobiological projects as part of a degree course. The layout of this manual should be particularly beneficial in the instruction and training of geotechnologists and museum preparators. Graduate students and scientists requiring an outline of a preparation procedure will also be able to use the manual as a reference from which to assess the suitability of a procedure. This manual is also intended for use by the "committed amateur". Many of the techniques described in this manual have been devised by non-palaeontologists, and developed from methods used in archaeology, zoology and botany, as well as other areas of geology. A considerable number of the methods can be undertaken by the amateur, and in the case of many of the field procedures, should be used. This will ensure that specimens and samples can be conserved in such a manner as to facilitate any later research, and not invalidate the results of subsequent geochemical analytical techniques which might be employed.

Book excerpt: In recent years microstructural analysis has been a rapidly changing field of scien tific endeavor. No longer are the efforts of the microstructural analysts (sometimes referred to as metallographers, materialographers, ceramographers, and similar desig nations) limited to the tasks of polishing, etching, and photographing specimens of materials. The performance demanded of materials used for many current applica tions requires much more complete characterizations than were possible only a scant few years ago. Although the individuals who have been expected to develop new and improved techniques to permit these required characterizations have been severely challenged, in large part they have met the challenge. In view of the many new developments in the field of microstructural analysis and recognizing the requirements to communicate these developments to the wide audience that might make use of them, the American Society for Metals and the In ternational Metallographic Society joined forces to co-sponsor a symposium that was intended to bring participants and attendees up to date on the subject "Inter pretive Techniques for Microstructural Analysis". This symposium was held in Min neapolis, Minnesota, USA, June 29 and 30, 1975. It followed two earlier symposia co-sponsored by the same two societies on other subjects of current interest to the metallographic community, Microstructural Analysis - Tools and Techniques, 1972, and Metallographic Specimen Preparation - Optical and Electron Micros copy, 1973.

Available in PDF, EPUB and Kindle. Book excerpt: Introduces readers to the enlightening world of the modern light microscope There have been rapid advances in science and technology over the last decade, and the light microscope, together with the information that it gives about the image, has changed too. Yet the fundamental principles of setting up and using a microscope rests upon unchanging physical principles that have been understood for years. This informative, practical, full-colour guide fills the gap between specialised edited texts on detailed research topics, and introductory books, which concentrate on an optical approach to the light microscope. It also provides comprehensive coverage of confocal microscopy, which has revolutionised light microscopy over the last few decades. Written to help the reader understand, set up, and use the often very expensive and complex modern research light microscope properly, Understanding Light Microscopy keeps mathematical formulae to a minimum—containing and explaining them within boxes in the text. Chapters provide in-depth coverage of basic microscope optics and design; ergonomics; illumination; diffraction and image formation; reflected-light, polarised-light, and fluorescence microscopy; deconvolution; TIRF microscopy; FRAP & FRET; super-resolution techniques; biological and materials specimen preparation; and more. Gives a didactic introduction to the light microscope Encourages readers to use advanced fluorescence and confocal microscopes within a research institute or core microscopy facility Features full-colour illustrations and workable practical protocols Understanding Light Microscopy is intended for any scientist who wishes to understand and use a modern light microscope. It is also ideal as supporting material for a formal taught course, or for individual students to learn the key aspects of light microscopy through their own study.
