Book excerpt: Detailed methods of the most widely used and best established animal models and related approaches to the study of rheumatic diseases and their treatment are presented in this major reference book. The detailed description of each model allows the reader to select the model most appropriate for his research and to reproduce the model without resorting to additional references. In addition, other methods for induction of the model, sources for procurement of suitable animals and validation of the model are highlighted. Methods for eliciting non-spontaneous phenomena, follow-up examinations and data collection are also described. Specific techniques such as joint-injection, obtaining synovial tissue, methods of assessment and similar non-specific information are emphasized. This easy-to-read resource text is essential for scientists, graduate students, research assistants, physicians and other invbestigators who use animal models for the study of rheumatic diseases.

Rheumatoid arthritis (RA) is an autoimmune disease of unknown etiology, characterized by the presence of chronic inflammatory infiltrates, the development of destructive arthropathy, bone erosion, and degradation of the articular cartilage and subchondral bone. There is currently no treatment that resolves the disease, only the use of palliatives, and not all patients respond to pharmacologic therapy. According to RA multifactorial origin, several in vivo models have been used to evaluate its pathophysiology as well as to identify the usefulness of biomarkers to predict, to diagnose, or to evaluate the prognosis of the disease. This chapter focuses on the most common in vivo models used for the study of RA, including those related with genetic, immunological, hormonal, and environmental interactions. Similarly, the potential of these models to understand RA pathogenesis and to test preventive and therapeutic strategies of autoimmune disorder is also highlighted. In conclusion, of all the animal models discussed, the CIA model could be considered the most successful by generating arthritis using type II collagen and adjuvants and evaluating therapeutic compounds both intra-articularly and systemically.

Available in PDF, EPUB and Kindle. Book excerpt: Rheumatoid arthritis is a bewilderingly complex disease involving the interactions of many, and varied, cell populations and multiple families of low and high molecular mass mediators. We are only slowly beginning to understand the mechanisms that produce the local and systematic pathology clinically recognized as rheumatoid arthritis. Increasingly, use is being made of experimental models of this disease in an effort to test hypotheses about putative pathological mechanisms and to investigate the effect of novel therapeutic agents. A major section of this book covers these experimental models in great detail from their development through to reviews of the most recent information on each model.Mechanisms and Models in Rheumatoid Arthritis brings together a group of eminent researchers from the fields of clinical rheumatology, pathology, experimental pathology, immunology, connective tissue biochemistry, microbiology, pharmacology and developmental biology to describe the current views of the cellular and humoral mechanisms that drive the pathology of rheumatoid arthritis and experimental models of rheumatoid arthritis. Key Features:* The book is divided into five sections:* The clinical spectrum, aetiopathogenesis, the role of microbial superantigens in pathology and the present and future therapies for rheumatoid arthritis* The pathology of rheumatoid arthritis, the development and role of synovial pannus and the use of immunohistochemistry in defining synovial pathology* Cell populations involved in synovitis with chapters on synovial cells, chondrocytes, bone cells, leukocytes and leukocyte trafficking* Mediators, with coverage of: cytokines and cytokine inhibitors, growth factors, free radicals and inflammatory lipid mediators, neuropeptides and proteases* Current studies on experimental models of rheumatoid arthritis

Available in PDF, EPUB and Kindle. Book excerpt: This book provides a comprehensive and up-to-date review of the basic and clinical aspects of infections and infectious processes that trigger or aggravate a variety of autoimmune and inflammatory musculoskeletal disorders. It delivers a timely update on the interactions between infection and rheumatic diseases in order to provide clinicians with the tools they need for proper diagnosis and treatment. The book is divided into five main sections. The first examines the basic aspects of bacterial infections with a discussion of molecular biology, the microbiome, various types of bacteria that cause infection, and recent advances in biologic therapy. The next section focuses on different forms of viral arthritis including hepatitis arthritis, arthritis associated with alpha viruses, and parvovirus-related arthritis. Subsequent chapters then discuss arthritis caused by mycobacteria, fungi, spirochete, and other miscellaneous arthritides. Following this are various analyzations on reactive arthritis and infection-related rheumatic diseases such as Whipple’s Disease, SAPHO Syndrome, rheumatic fever, and HIV-associated rheumatic manifestations. Finally, the book closes with three chapters on external impacts on infectious diseases such as climate change, vaccinations and systemic lupus erythematosus. Infections and the Rheumatic Diseases is an essential resource for physicians and related professionals, residents, fellows, graduate students, and nurses in rheumatology, clinical immunology, and internal medicine.

Available in PDF, EPUB and Kindle. Book excerpt: The world has recorded losses in terms of human life as well as extensive time spent in experimentation with development of new drugs, elucidation of disease mechanism(s), and therapeutic agent discovery. Ethical and legal issues cojoin in slowing down scientific discoveries in medicine and biology. The past two (2) decades, therefore, have seen tremendous attempts that largely are successful in developing animal models with the characteristics of mimicking, approximating, or expressing transplanted human organs/tissues. These models or rather approaches seem to be fast, cost-effective, and easy to maintain compared to primates. This book is a collection of expert essays on animal models of human diseases of global interest. A visible objective of the book is to provide real-time experimental approach to scientists, clinicians, ethicists, medicolegal/medical jurisprudence workers, immunologists, postgraduate students, and vaccinologists and informative and multidisciplinary approach for the identification of new therapeutic targets and biomarkers using animal models as well as investigating the pathogenesis and therapeutic strategies of human diseases. An increased understanding of the genetic, molecular, and cellular mechanisms responsible for the development of human diseases has laid out the foundation for the development of rational therapies mainly with animal models.

Book excerpt: Principles of Immunopharmacology provides a unique source of essential knowledge on the immune response, its diagnosis and its modification by drugs and chemicals. The 4th edition of this internationally recognized textbook has been revised to include recent developments, but continues the established format, dealing with four related fields in a single volume, thus obviating the need to refer to several different textbooks. The first section of the book, providing a basic introduction to immunology and its relevance for human disease, has been updated to accommodate new immunological concepts, particularly the role of epigenetics and the latest understanding of cancer immunology. The second section on immunodiagnostics offers a topical description of widely used molecular techniques and a new chapter on imaging techniques. This is followed by a systematic coverage of drugs affecting the immune system, including natural products. This third section contains 15 updated chapters, covering classical immunopharmacological topics such as anti-asthmatic, anti-rheumatic and immunosuppressive drugs, but also deals with antibiotics, plant-derived and dietary agents, with new chapters on monoclonal antibodies, immunotherapy in sepsis and infection, drugs for soft-tissue autoimmunity and cell therapy. The book concludes with a chapter on immunotoxicology and drug safety tests. Aids to the reader include a two-column format, glossaries of technical terms and appendix reference tables. The emphasis on illustrations is maintained from the first three editions. The book is a valuable single reference for undergraduate and graduate medical and biomedical students, postgraduate chemistry and pharmacy students, researchers in chemistry, biochemistry and the pharmaceutical industry and researchers lacking basic immunological knowledge, who want to understand the actions of drugs on the immune system.
