Book excerpt: ABSTRACT: Cryptosporidium parvum is of growing public health concern due to its ability to survive typical water treatment processes. In order to protect the public from infection, the Environmental Protection Agency developed Method 1623 for the detection of Cryptosporidium oocysts in environmental water samples. Execution of this method is time consuming, and the results do not provide an accurate estimation of viability. Therefore, current research is focused on creating a real-time PCR method for the accurate detection of viable Cryptosporidium parvum in environmental water samples. This thesis presents the development of a real-time PCR method, and the results obtained in its use on field samples. The assay was standardized using multiple dilution series in addition to positive and negative controls. Environmental water samples were tested using this method and Method 1623 for comparison. The results were compared statistically to determine the degree of correlation between methods. The data show that the real-time PCR method correlates well to Method 1623. In addition, the assay was determined to be more cost effective and less labor intensive than Method 1623. Although these early findings are promising, additional research and development are needed before the proposed assay can be used in industry.

The researchers tested the assumption that improved survival of host cells enhances growth of C. parvum. Although certain modifications to the culture methodology improved host cell survival, enhancing host cell survival was not always accompanied by an increase in parasite density in the cell monolayer. Some cultures grown on collagen or laminin showed improved parasite growth. However, this effect was not consistent, indicating that other variables affect parasite growth. C. parvum was more prevalent in cells in the mitotic cycle than in non-dividing cells. This is consistent with the absence of certain biosynthetic pathways in C. parvum, and suggests that dividing cells may be more favorable to parasite development. Although current culture methods enable the detection of infectious Cryptosporidium oocysts, limited parasite growth reduces the sensitivity of the assay, making it necessary to apply sophisticated methods for the detection of the intracellular parasites. This study identified methods that slightly improve growth of Cryptosporidium in culture and also highlighted methods that have no effect. This report is available only as a pay-per-view item.

Book excerpt: Cryptosporidium parvum is a protozoal pathogen transmitted through water by the fecal-oral route as oocysts. Because the oocysts are more resistant to environmental stresses than the bacteria conventionally used as indicators of fecal contamination, they can be present in water when indicator organisms, such as E. coli, are not found. In addition, because they are resistant to chlorine, they can pass from source water through water treatment into drinking water systems. The EPA method for detection of Cryptosporidium oocysts consists of identifying oocysts with fluorescently labeled antibodies, staining with 4',6-diamidino-2-phenylindole and examining slides with epifluorescent microscopy and differential interference contrast microscopy. This protocol is labor intensive and subject to technician error. A new method was developed for the rapid detection of Cryptosporidium parvum oocysts using fluorescent in situ hybridization (FISH) and the ScanRDI, a solid phase laser cytometer. Optimization of the FISH protocol for use with the ScanRDI was done with E. coli cells and known Cryptosporidium oocysts as a model. Source water and treated drinking water from the water treatment plant at Crow Agency on the Crow Indian Reservation in Montana was collected over the course of a year and concentrated using the EPA protocol for collection of oocysts. The samples were then examined for Cryptosporidium oocysts using both the ScanRDI method and the standard US EPA method. The combination of FISH for labeling Cryptosporidium and the ScanRDI for examination results in significantly higher numbers of Cryptosporidium detected as well as greater ease in identification. A statistical comparison was done that determined there was no correlation between the number of E. coli cells found in the water samples and the number of Cryptosporidium oocysts present. Additionally, although not tested on environmental samples, the FISH/ScanRDI method allowed for simultaneous detection of Cryptosporidium parvum oocysts and E. coli cells on the same membrane filter. Membranes were incubated before hybridization, hybridized concurrently with a Cryptosporidium specific probe and a probe specific for E. coli, followed by detection for both organisms with the ScanRDI.
