Book excerpt: Present-day drug design and testing includes experimental in-vitro and in-vivo measurements of the drug candidate's adsorption, distribution, metabolism, elimination and toxicity (ADMET) properties in the initial stages of drug discovery. Just a poor percentage of the recommended drug candidates get the authorization of government and reach the market place. Disadvantageous pharmacokinetic properties, bad bioavailability and efficiency, negative side effects, poor solubility and toxicity matters are responsible for most of the drug failures confronted in the pharmaceutical industry. Authors from across the globe have provided information elucidating pharmaceutical concerns, regulatory policies and clinical properties in their respective countries hoping that the open trade of scientific ideas and outcomes compiled in this book will result in enhanced pharmaceutical products. This book has a section on drug design covering several research chapters.

Book excerpt: This volume represents the proceedings of the Fifth Congress of the International Society of Ocular Toxicology (ISOT), which was held at the Grove Park Inn and Resort in Asheville, North Carolina, October 13-17, 1996. We are delighted to present this volume to the ophthalmic community, especially those with a significant interest in ocular toxicol ogy. The Fifth Congress was developed around themes relating to ocular drug metabolism, the ocular pathophysiological effects of nitric oxide, government issues relating to the use of alternative methods for toxicity testing, and a workshop that encompassed comparisons of both in vitro versus in vivo models as well as different animal models. The outcome of this congress, embodied in this volume, is a contribution to the methodologies currently employed or under development and to various drug or physical effects on different ocular tissues. While the focus of this proceedings is on ocular effects of drugs or other materials, many of the contributions deal with topics that have a much broader interest. The workshop concerning the use of different model systems and the choice of the best animal model for drug testing covers a wide range of interests that ex tends far beyond specific ocular effects. This is especially true in the area of alternative methods and in the choice of the best animal model for examination of different disease entities.

Book excerpt: Advances in molecular biology and toxicology are paving the way for major improvements in the evaluation of the hazards posed by the large number of chemicals found at low levels in the environment. The National Research Council was asked by the U.S. Environmental Protection Agency to review the state of the science and create a far-reaching vision for the future of toxicity testing. The book finds that developing, improving, and validating new laboratory tools based on recent scientific advances could significantly improve our ability to understand the hazards and risks posed by chemicals. This new knowledge would lead to much more informed environmental regulations and dramatically reduce the need for animal testing because the new tests would be based on human cells and cell components. Substantial scientific efforts and resources will be required to leverage these new technologies to realize the vision, but the result will be a more efficient, informative and less costly system for assessing the hazards posed by industrial chemicals and pesticides.

Book excerpt: Improving and Accelerating Therapeutic Development for Nervous System Disorders is the summary of a workshop convened by the IOM Forum on Neuroscience and Nervous System Disorders to examine opportunities to accelerate early phases of drug development for nervous system drug discovery. Workshop participants discussed challenges in neuroscience research for enabling faster entry of potential treatments into first-in-human trials, explored how new and emerging tools and technologies may improve the efficiency of research, and considered mechanisms to facilitate a more effective and efficient development pipeline. There are several challenges to the current drug development pipeline for nervous system disorders. The fundamental etiology and pathophysiology of many nervous system disorders are unknown and the brain is inaccessible to study, making it difficult to develop accurate models. Patient heterogeneity is high, disease pathology can occur years to decades before becoming clinically apparent, and diagnostic and treatment biomarkers are lacking. In addition, the lack of validated targets, limitations related to the predictive validity of animal models - the extent to which the model predicts clinical efficacy - and regulatory barriers can also impede translation and drug development for nervous system disorders. Improving and Accelerating Therapeutic Development for Nervous System Disorders identifies avenues for moving directly from cellular models to human trials, minimizing the need for animal models to test efficacy, and discusses the potential benefits and risks of such an approach. This report is a timely discussion of opportunities to improve early drug development with a focus toward preclinical trials.

Book excerpt: The increased exposure to toxins, toxicants and novel drugs has promoted toxicology to become one of the most important areas of research with emerging innovative toxicity testing protocols, techniques, and regulation being placed. Since the bioactivation of many toxins and toxicants and its consequences on human health are not clearly known, this book offers a quick overview of cellular toxicology through the cell, drug and environmental toxicity. This book does not strive to be comprehensive but instead offers a quick overview of principle aspects of toxins and toxicants in order to familiarize the key principles of toxicology. The book is divided into three main sections,; the first one discusses the role of mitochondrial dysfunction, oxidative stress and mitochondrial drug development. The second and third sections bring light to forensic toxicology and drug poisoning followed by environmental toxicity.

This book was released on 2006-04-07 with total page 270 pages. Available in PDF, EPUB and Kindle. Book excerpt: Toxicity testing in laboratory animals provides much of the information used by the Environmental Protection Agency (EPA) to assess the hazards and risks associated with exposure to environmental agents that might harm public health or the environment. The data are used to establish maximum acceptable concentrations of environmental agents in drinking water, set permissible limits of exposure of workers, define labeling requirements, establish tolerances for pesticides residues on food, and set other kinds of limits on the basis of risk assessment. Because the number of regulations that require toxicity testing is growing, EPA called for a comprehensive review of established and emerging toxicity-testing methods and strategies. This interim report reviews current toxicity-testing methods and strategies and near-term improvements in toxicity-testing approaches proposed by EPA and others. It identifies several recurring themes and questions in the various reports reviewed. The final report will present a long-range vision and strategic plan to advance the practices of toxicity testing and human health assessment of environmental contaminants.
