Available in PDF, EPUB and Kindle. Book excerpt: The principal objective of this symposium was to bring together investigators from all over the world who work in diverse areas of research but share a common interest in the physiologic and pathophysiologic funetions of the nueleoside adenosine. In 1978, the "International Conference on Physiological and Regulatory Funetions of Adenosine and Adenine Nueleotides," held in Banff, Canada, served as an effective catalyst in enabling scientists with differ ent points of view to exchange information and ideas on this subject. Since 1978, the amount of research on different as peets of adenosine metabolism, mechanism of action, transport, and function in various tissues (in both nor maI and abnormal states) has increased exponentially, and there was an urgent need for another meeting for updating information on this burgeoning subject. The focus of the meeting was extremely sharp, but the scope of the subject material was very broad. Scientists categorized as chemists, biochemists, physiologists, pharmacologists, and immunologists, working with cells and eelI fragments, with tissues such as heart, brain, musele, fat, kidney, and with human beings having immune deficiency diseases participated in the sym posium. They had an opportunity to air their differences, reinforce their areas of agreement, and, most important, they educated each other with respeet to research on adenosine; it is hoped that they also enriched their own research programs from the information they gained from their fellow participants.

Book excerpt: Extracellular purines and pyrimidines (ATP, ADP, UTP and adenosine) are released into the extracellular milieu in response to a variety of stress conditions and act as important regulators of vascular homeostasis. This new book is uniquely focused on the signaling actions of extracellular purines in endothelial cells and the crucial role of extracellular purines in regulation of angiogenesis, vascular tone, cell permeability, wound healing, inflammation and cell-to-cell communication. This book examines the responses of endothelial cells, originating from various tissues (such as cornea, pancreas and uterus), to extracellular nucleotides and adenosine under physiological and pathological conditions, i.e. pregnancy, hypoxia, hypertension, inflammation and diabetes. In the book’s 12 chapters, the role of purinergic signaling in endothelium-dependent tissue perfusion, regulation of endothelial barrier function, and angiogenesis are discussed. The mechanisms of ATP release and its role in intercellular communication are also presented. In addition, the book provides the most up to date mechanisms of extracellular nucleotide metabolism by purine-converting ecto-enzymes and their contribution to purinergic signaling in endothelial cells originating from various vascular beds. This book is a valuable resource for biomedical research scientists, clinical scientists, graduate students and health science professionals interested in the mechanisms of extracellular purine function in endothelial cells under physiologic and pathologic conditions.

Available in PDF, EPUB and Kindle. Book excerpt: In their classic paper in 1929, Drury and Szent-Gyorgyi described a number of the important cardiovascular actions of adenosine. Another thirty years were to pass before the possible physiological role of adenosine in coronary vasodilation was studied by Berne and others. Since then, there has been a tremendous increase in research into the actions of adenosine. Workers from many disciplines have employed a wide variety of techniques, since adenosine is a product of and a substrate for a number of metabolic pathways, is transported into cells, and acts at discrete receptor sites to modulate the activity of adenylate cyclase and to produce important actions on many cells and tissues including platelets, adipo cytes, heart, blood vessels, and other smooth muscles. International symposia on the actions of adenosine were held in 1978, 1981, and 1982, and the proceedings of these symposia have been published (Baer and Drummond, 1979; Daly et at., 1983; Berne et at., 1983). Since it is not the primary purpose of the present volume to review our current understanding of the nu merous actions of adenosine, these volumes should be consulted for such details. Rather, the present volume has been planned to provide both graduate students and investigators in pharmacology and related disciplines with a summary of some of the methods now available for the study of the actions of adenosine and, in particular, to highlight their possible uses and limitations.

Available in PDF, EPUB and Kindle. Book excerpt: Homeostasis of key metabolites and metabolic health affects all bodily systems. Not surprisingly, altered metabolic function is associated with a wide spectrum of dysfunctions in the central nervous system – including developmental disorders, acute nervous system injury, and neurodegenerative disorders. Accordingly, metabolism-based therapies offer significant promise as new category of treatment options designed to limit, delay or reverse the disease process by reconstructing homeostatic functions. Increasingly it is appreciated that restoring metabolic health could promote normal nervous system activity, and improve behavior and cognition. Adenosine: A Key Link Between Metabolism and Central Nervous System Activity focusses on diverse aspects of adenosine, an evolutionarily conserved homeostatic bioenergetic regulator in the central nervous system. Because of its interrelationship with ATP (adenosine triphosphate), adenosine is integral to cell metabolism. At the same time, adenosine influences neuronal activity directly via receptors, and is involved in biochemical processes related to gene expression. Thus, adenosine is uniquely placed as a reciprocal and rapid link between changes in metabolism and changes in neuronal activity, and, on a longer time scale, to changes in gene expression and long term changes in cell function. Leaders in the field feature basic research on adenosine at the cellular level in the central nervous system, and relate these findings to its recognized potential in diverse acute and chronic disorders. This comprehensive overview of adenosine also highlights emerging adenosine-based treatments and associated opportunities for central nervous system disorders.

Book excerpt: Adenine Nucleotides play a major role in cellular metabolism and functions, serving as high-potential phosphate transfer compounds in energy metabolism and as substrates and co-factors for proteins involved in signal transduction. During the last few years definite advancement has been made in elucidating the molecular and genetic aspects of the enzyme involved in oxidative phosphorylation, the so-called FoFl H~-ATP synthase. Non-invasive NMR technologies have been developed to monitor in vivo the energy level of tissues based on determination of the concentrations of adenine nucleotides, phosphate and phosphate esters. Thus it became clear that the capacity of oxidative phosphorylation adapts itself to the ATP demand which changes continuously with the physiological state in various tissues. This is achieved by regulation of the enzyme activity as well as by regulation of its biogenesis. The reversible phosphorylation of proteins is re cognised as a major regulatory mechanism in eukaryotic as well as in prokaryotic cells for cellular signal transduction and control of gene expression, cell growth, differentation and oncogenesis. The same applies to the role played by cAMP. A further topic of growing interest concerns the discovery of the ATP binding cassette (ABC) superfamily of transport proteins which includes systems of primary importance in medicine such as the multi-drug resistance P glycoprotein, the cystic fibrosis transmembrane conductance regulator (CFTR) and the 70 kd peroxisomal membrane protein. Finally, much attention is being devoted in many laboratories to the molecular structure and role of ATP modulated channels.

Once thought only as an intracellular molecule that provides energy, ATP is also now considered an essential autocrine/paracrine agonist that acts extracellularly within tissues and tissue microenvironments. Receptors for ATP and its metabolites, so-called "purinergic receptors," are essential membrane receptors that transduce the extracellular signal carried by ATP. Many cell types or tissues express multiple types of these receptors, often on the same cell. This volume will cover the history and overall impact of extracellular ATP signaling, methods to detect ATP release and signaling, the molecular and cell biology of purinergic receptors, the pharmacology of purinergic receptors, the ion channel biophysics and biochemistry of P2X purinergic receptor channels, and the physiology and pathophysiology of purinergic receptors. Key Features* Organized and written by world-renowned experts* Covers state-of-the-art experimental methods* Has broad appeal to basic and clinical scientists, the biotch industry and pharmaceutical research companies
