Book Synopsis A toolkit to study nuclear transport pathways: nanobody-mediated inhibition of importin β-related nuclear transport receptors (NTRs) by : Dilantha Perera

This book was released on 2023-08-07 with total page 136 pages. Available in PDF, EPUB and Kindle. Book excerpt: Eukaryotic cells are divided into a nuclear and a cytoplasmic compartment. This separates transcription from translation and makes gene expression dependent on nucleocytoplasmic transport. The members of the importin β superfamily function as shuttling nuclear transport receptors (NTRs) that recognize and actively transport cargoes through nuclear pores. An estimated 5 000 to 10 000 different human proteins are subject to active nuclear transport. Numerous cargo/NTR pairs have been identified, however, we are still far from a complete understanding as it has been very challenging to setup a systematic in vivo analysis that integrates the impact of all transport pathways. In this study, we obtained anti-NTR nanobodies against TRN1, Xpo4, Xpo7, and CAS. Our aim was to identify nanobodies, and prepare nanobody fusions, that impede nuclear pore-passage of the targeted NTR and thus, interrupt a given transport cycle. These nanobody fusions were observed to inhibit the partition of NTR/cargo complexes into a reconstituted FG phase. We also observed that the nanobodies and nanobody fusions inhibit NTR transport in permeabilized cells.
Book Synopsis Far-Field Optical Nanoscopy by : Philip Tinnefeld
Download or read book Far-Field Optical Nanoscopy written by Philip Tinnefeld and published by Springer. This book was released on 2015-02-07 with total page 335 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book describes developments in the field of super-resolution fluorescence microscopy or nanoscopy. In 11 chapters, distinguished scientists and leaders in their respective fields describe different nanoscopy approaches, various labeling technologies, and concrete applications. The topics covered include the principles and applications of the most popular nanoscopy techniques STED and (f)PALM/STORM, along with advances brought about by fluorescent proteins and organic dyes optimized for fluorescence nanoscopy. Furthermore, the photophysics of fluorescent labels is addressed, specifically for improving their photoswitching capabilities. Important applications are also discussed, such as the tracking and counting of molecules to determine acting forces in cells, and quantitative cellular imaging, respectively, as well as the mapping of chemical reaction centers at the nano-scale. The 2014 Chemistry Nobel Prize® was awarded for the ground-breaking developments of super-resolved fluorescence microscopy. In this book, which was co-edited by one of the prize winners, readers will find the most recent developments in this field.

Book Synopsis Protein Structure by : Robert Austin
Download or read book Protein Structure written by Robert Austin and published by Springer. This book was released on 2011-10-13 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book is concerned with the physical aspects of molecular and electronic tunneling in biological systems, and the extent to which protein structure controls these events. The scope is very broad and this volume could almost be a textbook in biophysics. Both fundamental processes and the extrapolation to physiological events are stressed. The discussion sections are remarkably frank and offer insight into the basic problems confronting physists and chemists as they seek to apply their techniques to biological systems. This book on the physics of biomolecules reflects recent progress in understanding the biological function of the key protein molecules from detailed knowledge of their physics. New and exciting are the glasslike aspects of protein structures and the discussion of proteins as fractals. Other topics dealt with are low-temperature kinetics and reactivity, structure and charge exchange, and charge separation in photosynthetic reaction centers.
