Différentes techniques analytiques peuvent être utilisées pour la validation des procédés telles que l'HPLC, le travail dans l'UV/visible (UVNis), et la méthode par mesure du Carbone Organique Total (COT). - Les méthodes HPLC et UVNis sont des méthodes spécifiques qui identifient et quantifient des substances actives définies. La méthode COT est une méthode non spécifique qui détecte tous les composés contenant du carbone organique, tels que les substances actives, les excipients et les agents de nettoyage. - L'inconvénient majeur d'une méthode spécifique est qu'il faut créer un nouveau protocole pour chaque substance active utilisée dans la production de médicament. Ce procédé nécessite un temps précieux et peut se révéler pénalisant pour l'activité industrielle. - A l'inverse une même méthode COT peut être potentiellement utilisée pour tous les produits fabriqués. La méthode d'analyse par mesure du taux de COT est précise jusqu'au ppb (partie par billion) et nécessite moins de temps que les méthodes HPLC ou UVNis. Des méthodes développées par l'USP et la Pharmacopée Européenne concernant la recherche de COT sont des standards pour l'analyse de l'eau pour préparations injectables et de l'eau purifiée (3) (4) (5); de simples applications de ces méthodes peuvent être utilisées pour la validation du nettoyage. - Le but de cette étude est de démontrer comment développer et valider une méthode d'analyse COT pour la validation du nettoyage. - Parmi les différents paramètres étudiés sont inclus la linéarité, la détermination de la limite de détection, de la limite de quantification, l'exactitude, la répétabilité, la reproductibilité, et le taux de récupération.
