Book excerpt: In recent years, the techniques of electron microscopy have developed so widely and rapidly that they now cover the fields of research once the unique ll:panage of sister research techniques such as biochemistry, physiology, immunology, X-ray diffraction, etc. It is now possible to reach molecular and submolecular levels, making this technique indispensable in every type of research. Electron microscopy alone often provides enough information to solve given problems. In the field of the connective tissue matrix, knowledge of the molecular structure of collagen, pro teoglycans and elastin and their interaction has been to a large extent elucidated by electron microscopy. The field over which electron microscopy ranges in the investigation of the connective tissue matrix is so wide that the aim of this volume is to collect the main ultrastructural acquisitions disseminated in various journals and monographs in one book. The intent ofthis volume is to: (a) integrate different and new microscopic methods and review the results of such an integrative approach; (b) present a comprehensive ultrastructural account of selected aspects of the field; (c) point out gaps or controversial topics in our knowledge; (d) outline pertinent future research and expansion of the subject.

Available in PDF, EPUB and Kindle. Book excerpt: The use of nano-materials in tissue engineering is being widely researched and many subsequent applications currently exist. Recently it was realized that the extra-cellular matrix (ECM) is an intricate interweaving of nanometer sized protein fibers (i.e., mainly collagen). Replicating the ECM remains a challenge. However, it is crucial for applications ranging from skin and organ replacement to testing drug toxicity. Currently there are several fabrication methods for replicating the ECM, each of which have numerous drawbacks including, lack of repeatability and failure to replicate the internal fibrillar and hierarchical collagen structure. The work outlined for this dissertation will utilize a flow-based gel-spinning protocol to assemble collagen fibrils with and without the presence of nano-carbons. This novel synthetic method is aimed at achieving continuous collagen fibers, which exhibit highly aligned fibrillar and organized molecular structures as toward mimicking the native material. This type of collagen fiber fabrication remains a challenge to date. Collagen and collagen/nano-carbon composites were fabricated and characterized in order to determine the effects of the nano-carbon, in terms of geometry, size-scale, and distribution in the matrix, on collagen self-assembly and molecular packing. Nano-carbon dispersions, collagen sonication, fiber incubation, fiber strain and cross-linking were also studied to determine their effects on the overall assembly process. Fundamental studies to determine the structure-property relationship were also conducted using electron microscopy, X-ray scattering/diffraction techniques, and mechanical testing. These characterization methods allow better understanding of the nano-carbons ability to template highly aligned collagen fibrillar organization as well as the capability for these fillers to influence the collagen fiber structure.

A detailed discussion of the lung and liver is included. Another topic of interest is the liver fibrosis development induced by parasitic infection or alcohol. The periodontal ligament is specialized tissue whose basic role is tooth support. This tissue is covered in detail. A section of the book provides an analysis of the elastin gene expression and regulation. Topics such as the microfibrillar components of connective tissue, the regulation of collagen genes, the complexity and the evolution of the collagen molecule, the invasiveness of malignant tumors, and the characteristics of fibroblast are fully explained. The text presents an exhaustive description of the connective tissue of the lung. This chapter includes a comprehensive description of the pulmonary system. The book will provide useful information to doctors, students, researchers and specialists in the field of human anatomy.

Book excerpt: Collagen is a fascinating protein not only because of its ubiquitous occurrence in multicellular animals, but also because of its unique chemi cal structure. As the predominant constituent in bone, cartilage, skin, tendon, and tooth, it is not surprising that collagen is of interest to anatomists, biochemists, biomedical engineers, cell biologists, dermatolo gists, dental surgeons, leather chemists, orthopedic surgeons, physiologists, physicians, zoologists, and a host of others. This book was planned to provide an up-to-date comprehensive survey of all aspects of biochemistry of collagen. The recent discovery of genetically distinct collagens with tissue specificity has opened a new era in collagen biochemistry, and Karl Piez discusses this in the opening chapter on primary structure. In the next chapter, Ramachandran and Rama krishnan deal with the molecular structure of collagen, placing special emphasis on the conformational aspects of its polypeptide chains. Follow ing the consideration of primary and secondary structure of collagen, the three-dimensional arrangement of collagen molecules in the fibrils is covered by Miller in Chapter 3. Collagen is generally in the insoluble state in the living organism due to the cross-linking of individual molecules, and Tanzer describes the various aspects of this cross-linkage in Chapter 4. The biosynthesis of collagen is discussed in depth by Prockop and his colleagues.

Available in PDF, EPUB and Kindle. Book excerpt: International Review of Connective Tissue Research, Volume 7 focuses on the structural aspects of the connective tissues and their compounds. This book reviews the data concerning the primary structure of collagen and explores the physical features of elastin. Organized into six chapters, this volume begins with an overview of the amino acid sequence of collagen. This text then examines the relationship of the structure and function of elastic tissues and reviews the usefulness of immunological methods when applied to structural studies. Other chapters consider the substantial progress in the elucidation of the antigen determinants of the collagen molecule. This book discusses as well the immunological properties of collagen that may have considerable bearing on the causes of some connective tissue diseases. The final chapter deals with the extensive investigations into glycoproteins in various connective tissues of various species at various ages. This book is a valuable resource for biochemists and physicians.

Book excerpt: A brief historical account of the background leading to the publication of the first four editions of the World Directory of Crystallographers was presented by G. Boom in his preface to the Fourth Edition, published late in 1971. That edition was produced by traditional typesetting methods from compilations of biographical data prepared by national Sub-Editors. The major effort required to produce a directory by manual methods provided the impetus to use computer techniques for the Fifth Edition. The account of the production of the first computer assisted Directory was described by S.C. Abrahams in the preface of the Fifth Edition. Computer composition, which required a machine readable data base, offered several major advantages. The choice of typeface and range of characters was flexible. Corrections and additions to the data base were rapid and, once established, it was hoped updating for future editions would be simple and inexpensive. The data base was put to other Union uses, such as preparation of mailing labels and formulation of lists of crystallographers with specified common fields of interest. The Fifth Edition of the World Directory of Crystallographers was published in June of 1977, the Sixth in May of 1981. The Subject Indexes for the Fifth and Sixth Editions were printed in 1978 and 1981 respectively, both having a limited distribution.

Beginning with a section on in vitro models for the characterization of collagen formation, the book continues by highlighting large-scale analysis of collagen with mass spectrometry in order to elucidate the proteomics, degradomics, interactomes, and cross-linking of collagen, high resolution imaging approaches for collagen by the use of scanning electron microscopy and multiphoton imaging, as well as the role of collagen during physiological and pathological conditions. Written for the highly successful Methods in Molecular Biology series, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and tips on troubleshooting and avoiding known pitfalls. Authoritative and practical, Collagen: Methods and Protocols is an ideal guide to high quality and repeatable protocols in this vital field of study.
