Book excerpt: The work described in this thesis focuses on the role of morphologies and surface chemistry on the properties of nanomaterials. First, the role of organic surface ligands in the synthesis of nanoheterostructures is discussed. It shows that the length of the alkyl chain of surface ligands can shift the equilibrium between the wurtzite and zinc blende polytypes of CdSe nanocrystals (NCs). In-situ wide-angle X-ray scattering measurements reveal that short-chain (e.g., propyl) phosphonic acids stabilize CdSe NCs with the zinc blende phase whereas octadecylphosphonic acid stabilize NCs with the wurtzite phase. It is also demonstrated how this effect can be used to improve the shape selectivity in the synthesis of anisotropic CdSe/CdS and ZnSe/CdS nanoheterostructures. Then we describe the effect of morphologies on the electronic and optical properties of NCs. The quantum-confined stark effect of CdSe/CdS nanoheterostructures embedded in polymer matrix is studied. Single-particle spectroscopy of CdSe/CdS nanoheterostructures is used to explore interfacial energy transfer in NCs. We further discuss inorganic ligands capped NCs and the enhanced electronic coupling in all-inorganic NCs arrays. Oxo-ligands is explored as a new group of inorganic ligands to functionalize NCs. The chemistry of ligand exchange is discussed in detail and the binding affinity of different inorganic ligands is compared. We also demonstrated the catalytic properties of POMs retain after capping on NCs.

Colloidal nanochemistry has emerged as one of the most effective methods for generating high quality, monodisperse nanoparticles with diverse structural features and highly complex geometries. These wet-chemical approaches offer an array of synthetic levers that can be used to tailor nanoparticles for targeted applications, and deliver solution-dispersible solids that are easily integrated onto device architectures. Additionally, colloidal nanoparticles can be used as building blocks for constructing periodic superlattices and multicomponent hybrid nanoparticles, which offer unique properties that can support next-generation technologies. As the applications for colloidal nanoparticles continue to expand, the architectural and compositional requirements for these materials are becoming increasingly rigid. Conventional colloidal methods are effective for generating diverse nanoparticle systems, but rely on complex nucleation and growth processes, which are often poorly understood and difficult to control in dynamic reaction environments. For these reasons, there are a number of high profile nanoparticle targets that remain out of reach. Accordingly, new approaches are needed that can circumvent these synthetic bottlenecks and narrow the growing disconnect between nano-design and synthetic capability. In this dissertation, I present several colloidal strategies for engineering synthetically challenging nanomaterials using multistep reaction sequences that, in many ways, parallel the total-synthesis framework that organic chemists use to access complex molecules. A variety of approaches are discussed, including nanoscale ion exchange transformations and seeded-growth protocol for constructing multicomponent hybrid nanoparticles. First, I demonstrate that solution-mediated anion and cation exchange can be integrated into one multistep reaction sequence, which leads to a complete material transformation of a pre-synthesized nanotemplate. Importantly, although the final product does not contain any of the original elements, the morphology is retained, effectively decoupling morphology and composition control. Next, I demonstrate that both anion and cation sublattice features of preformed Cu2-xS nanocrystals can be retained during cation exchange with Co2+ and Mn2+, yielding wurtzite-type CoS and MnS polymorphs that are metastable in bulk systems. This study was enabled by new cation exchange chemistry with previously unexplored 3d transition metal systems, and offers new guidelines for predictably targeting sublattice features in colloidal nanomaterials. To conclude the dissertation, I offer two studies investigating the seeded-growth synthesis of three-component Ag-Pt-Fe3O4 heterotrimer nanoparticles, which are generated by adding a Ag domain to preformed Pt-Fe3O4 heterodimers. First, to gain access to the alternate Pt-Fe3O4-Ag configuration, I demonstrate that a thin iron oxide shell can be applied to the Pt surface of the Pt-Fe3O4 seeds, which acts as a solid-state protecting group that can direct Ag growth onto the Fe3O4 domain, producing the otherwise inaccessible Pt-Fe3O4-Ag architecture. This strategy is inspired by similar techniques used in organic synthesis, and represents an important addition to the total synthesis toolbox used to construct colloidal hybrid nanoparticles. Finally, I offer an in-depth microscopic investigation that probes the chemoselective addition of Ag to Pt-Fe3O4 to form the Ag-Pt-Fe3O4 heterotrimer product. This study reveals that Ag indiscriminately nucleates on both the Pt and Fe3O4 domains of the Pt-Fe3O4 seeds during the early stages of the reaction, followed by a surface-mediated coalescence of Ag onto the Pt domain to yield the Ag-Pt-Fe3O4 configuration. The mechanistic insights gained in this work provide new design criteria for synthesizing multicomponent hybrid nanoparticle systems with targeted configurations.

Many efforts were made starting from the choice of appropriate nanoparticles (NPs) that provide desired photophysical and chemical properties, tailoring the surface coating with modular organic ligands, ensuring compact size and biocompatibility, to finally realizing their bio-applications. The aim of this dissertation is to provide a better understanding of the synthesis and growth of inorganic NPs, to explore various surface conjugations strategies in aqueous media, and to use the control over optical features for application in biomedical sensing. The dissertation is organized into seven chapters, as follows: Chapter 1 provides basic optical, physical and chemical properties of quantum dots (QDs) and gold colloids. The synthetic chemistry and surface coating strategies are briefly introduced, with emphasis on the recent progress on ligand design. We then discuss several effective bioconjugation approaches on the particle surfaces and review representative examples of using energy transfer-based bio-nano hybrids for sensing and intracellular delivery purpose. Chapter 2 first presents the exact transformation of the anhydride rings, in a poly(isobutylene-alt-maleic anhydride) block copolymer, PIMA, upon reaction with primary amino nucleophiles. We find that the ring opening reaction occurs under mild conditions. Conversely, cyclic imide formation is promoted when the reactions are carried out at high temperatures, or the use of a protic solvent, such as methanol, or addition of thionyl chloride. Based on this work, in Chapter 3 the as-prepared multi-coordinating polymer ligands, LA-PIMA-PEG under ring-opening conditions, are then applied for coating on various gold colloids and the effectiveness of a photoligation strategy is tested. We find that photoligation on gold surfaces occurs within 1 h and consumes a fraction of ligands often required in a large amount using a conventional ligation strategy. In Chapter 4, we show that the natural swelling of polymer macromolecules on particle surfaces leaves a fraction of the lipoic acid groups in coordination free, which are targeted for activation and conjugation to other molecules, using the reliable sulfhydryl-to-maleimide reaction. The next part of the dissertation shows the application of nanomaterials (e.g., QDs and AuNPs) for biosensing purpose. In Chapter 5, we exploit the use of PEGylated AuNPs, covalently coupling with dye-labelled peptides, as optical-addressed nanoprobes for detecting the activities of MMP-14 enzyme. Quantitative analyses based on a jammed Michaelis-Menten model show a slow proteolysis at interfaces and theoretical calculations within diffusion-collision framework is employed to describe the complexity and irregularity of interfacial enzymatic reactions. Furthermore, Chapter 6 presents a Förster resonance energy transfer (FRET)-based QD-peptide-dye assemblies that are able to detect the proteolytic activity of matrix metalloproteinases-14 (MMP-14) enzyme. The sensing capability is enabled by correlating the FRET efficiency changes with the alternation of donor-acceptor spatial separation, which is induced by the enzyme proteolysis on the surface-docked peptides. The last part of the dissertation focuses on investigating the growth mechanisms of anisotropic gold nanorods. In Chapter 7, ex-situ TEM images indicate that the shape anisotropy is due to truncated octahedral single crystals favor the deposition of gold precursors on (111) facets and therefore present preferential growth along (100) direction. In-situ liquid-cell TEM show the complex dynamics of growth which includes oriented attachment and formation of filament structures, yielding striking aspects on the symmetry breaking at early growth stages. This emphasizes the importance of integrating nonclassical crystallization models to the traditional interpretations of how post-nucleates grow in synthetic systems.

Book excerpt: As the roles of inorganic nanomaterials continue to advance, precise control over property defining features such as composition, crystal structure, morphology and surface functionality becomes increasingly crucial. Colloidal synthesis employs solubilized chemical precursors to build materials from the bottom-up, providing an attractive method for generating nanoscale materials with extraordinary levels of control over material dimensions and morphology. In addition to the range of tunable synthetic parameters, colloidal syntheses offer solution dispersible nanoscale solids, which allows for convenient and high throughput liquid phase processing of the desired materials.Under the appropriate synthetic conditions, colloidally dispersed nanoparticle seeds can be used as structural foundations to nucleate and grow new material domains. Through this strategy, nanoparticles consisting of chemically distinct materials connected through solid state interfaces can be accessed. Dubbed hybrid nanoparticles, these materials constitute an expanding class of multi-component nanostructures that can exhibit multifunctionality as well as emergent phenomena not observed for the constituent materials independently. Moreover, these hybrids can adopt structures that expose multiple material domains to the surrounding media, allowing for additional synergies to be realized due to the distinct chemical surfaces arranged in close proximity. The unique features offered by these hierarchal nanostructures underpin, among many others, advanced applications in photovoltaics, biomedical theranostics, catalysis and chemical sensing.Although colloidal hybrid nanoparticles are attractive for a variety of applications, there are still significant hurdles that impair their widespread use. For example, the complex pathways that govern the heterogeneous nucleation and growth of these materials are not well understood, and accordingly, a robust synthetic framework for the modular design of hybrid structures from a diverse library of material combinations and connectivities has yet to be developed. In addition, current synthetic protocols for known colloidal hybrid architectures are nontrivial to execute, with run-to-run variability commonly observed among the product particle populations. For these reasons, studies that provide fundamental insights into colloidal reaction pathways, or offer robust strategies to access these materials in high quality, will enable key advancements in the future design and application of hybrid nanoparticle systems.In this dissertation, I present several contributions focusing on various aspects of inorganic colloidal nanoparticle chemistry. These contributions include chapters describing rigorous synthetic details as well as insights into the mechanistic pathways that govern the outcome of colloidal hybrid nanoparticle syntheses. Specifically, these studies center on the well known heterotrimer nanoparticle systems Ag-Pt-Fe3O4 and Au-Pt-Fe3O4, using them as model systems to provide generalizable insights into hybrid nanoparticle design and synthesis. In a subsequent chapter of this work, I describe new developments in the colloidal nanoparticle purification technique differential magnetic catch and release. This chapter demonstrates an alternative strategy to accessing predictable and high quality colloidal structures, by focusing on post synthetic purification rather than judicious synthetic control. Finally, an additional chapter of this dissertation describes a study investigating the selective hydrogenation of functionalized nitroarenes. These selective transformations typically require the application of complex nanoscale catalysts, which have complementary chemical surfaces in close proximity that are capable of cooperatively activating distinct reagent functionalities. The structural commonalities between heterostructured hybrid nanoparticles and these catalytic systems motivated us to investigate these transformations further and resulted in the unexpected discovery of bulk iron pyrite (FeS2) as an effective catalyst for these transformations.
