Book excerpt: Dual specificity phosphatases (DUSPs) constitute a heterogeneous group of protein tyrosine phosphatases with the ability to dephosphorylate Ser/Thr and Tyr residues from proteins, as well as from other non-proteinaceous substrates including signaling lipids. DUSPs include, among others, MAP kinase (MAPK) phosphatases (MKPs) and small-size atypical DUSPs. MKPs are enzymes specialized in regulating the activity and subcellular location of MAPKs, whereas the function of small-size atypical DUSPs seems to be more diverse. DUSPs have emerged as key players in the regulation of cell growth, differentiation, stress response, and apoptosis. DUSPs regulate essential physiological processes, including immunity, neurobiology and metabolic homeostasis, and have been implicated in tumorigenesis, pathological inflammation and metabolic disorders. Accordingly, alterations in the expression or function of MKPs and small-size atypical DUSPs have consequences essential to human disease, making these enzymes potential biological markers and therapeutic targets. This Special Issue covers recent advances in the molecular mechanisms and biological functions of MKPs and small-size atypical DUSPs, and their relevance in human disease.

Book excerpt: Glucocorticoids are steroid hormones that are used synthetically for their anti-inflammatory and immunosuppressive effects. However, treatment can result in unwanted side effects, such as insulin resistance, osteoporosis, and muscle atrophy. The effects of glucocorticoids are mediated through an intracellular glucocorticoid receptor (GR). The wanted anti-inflammatory effects of synthetic glucocorticoids, like dexamethasone (DEX), occur through gene repression by indirect GR association with NF-[kappa]B and AP-1 transcription factors while the side effects are associated with gene activation. In the pharmaceutical field, it would be ideal to find a GR modulator that can distinguish between transactivation and transrepression in both tissue-specific and promoter-specific contexts. Muscle atrophy can be triggered by different conditions--disease, nutritional deprivation, and altered hormone levels--and has been associated with increased proteasome activity. The MuRF1 and MAFbx genes were found to be upregulated in different atrophy-inducing conditions, including DEX treatment, and serve as key atrophy markers. In MuRF1 and MAFbx knockout mice, significant muscle mass sparing was observed. In efforts to discover novel GR ligands with selective effects, a series of drugs (NSR compounds) were created, displaying promising results in distinguishing repression of NF-[kappa]B and AP-1 dependent promoters from activation of GRE-containing promoters in transfected reporter gene assays and GR target cells. Previously, our lab studied the effects NSR1, NSR2, and NSR5 in the C2C12 mouse skeletal cell line. While these compounds efficiently suppressed pro-inflammatory IL-6 expression, they poorly regulated the levels of MuRF1 and MAFbx induction. NSR1 had weak or no effects in the upregulation of endogenous MuRF1 and transfected MuRF1 promoter-containing or MuRF1 GRE-containing reporter genes. We found that the NSR compounds induced myotube atrophy to similar degrees as measured MuRF1 and MAFbx expression levels, with NSR1 resulting in minimal atrophy. Additionally, the NSR compounds were effective at resulting in nuclear translocation of the GR. Attention was given to NSR1, which suppressed IL-6 expression but weakly upregulated MuRF1 and MAFbx expression, thus a potential GR ligand. NSR1 weakly activated glucocorticoid-responsive genes due to poor association of the GR to GRE-containing promoters. Our studies have demonstrated the complexity of GR gene regulation, which is cell-specific and promoter-specific.
