Book excerpt: Retinoids have received considerable attention in recent years and due cognizance has been given to their versatility as biological response modifiers, as evidenced by the virtually explosive growth of literature in this field in the past few years. This volume has been designed to give a current state-of-the-art picture of retinoids. The perceived potential of retinoids in the treatment of certain disease stated has initiated attempts at identifying and synthesizing new retinoid derivatives with definable and selective effects on aberrant biological phenomena. Appropriately, therefore, we begin with the chemistry of retinoids and their derivatives together with discussions of their biological activity. Major advances have been made in understanding the mechanisms by which retinoids modulate physiological and phenotypic traits of cells. The transduction of retinoid signaling by the mediation of nuclear receptors of the steroid/thyroid receptor superfamily has now been studied extensively and the cloning and defining the characteristics of these receptors has been a focus of discussion in this volume. Retinoids also markedly modulate the transduction of extracellular signals such as those imparted by growth factors and hormones, and thus actively influence and control cellular proliferative patterns. Retinoids can alter epidermal growth factor receptor expression (Kawaguchi et al., 1994), responsiveness to thyroid hormone (Esfandiari et al., 1994; Pallet et al., 1994), inhibit the proliferative responses of hematopoietic progenitor cells to granulocyte colony stimulating factor (Smeland et al., 1994), and modulate secretion on interleukins by leukaemic cells (Balitrand et al., 1994), among other things. This has obvious implications for pharmacological manipulation of deregulated growth (Dickens and Colletta, 1993; Mulshine et al., 1993). Apoptosis is another component in the regulation of growth control. Apoptotic cell death is influenced by several agents and retinoids may function by interfering with apoptotic pathways of regulation of growth control and quite legitimately, therefore, the importance of this aspect of retinoid function has been duly recognized here.

Book excerpt: This book presents the current state of knowledge on the origin and differentiation of cell lines involved in the development of the vertebrate male and female gonads with particular emphasis on the mouse. It also discusses the processes leading to the testis- and ovary-specific structures and functions. The individual chapters review the origin and differentiation of the somatic cells of the genital ridges; the formation and migration of primordial germ cells in mouse and man; the gonadal supporting cell lineage and mammalian sex determination; differentiation of Sertoli and granulosa cells; mesonephric cell migration into the gonads and vascularization; origin and differentiation of androgen-producing cells in the gonads; germ cell commitment to the oogenic versus spermatogenic pathway and the role of retinoic acid; ovarian folliculogenesis; control of oocyte growth and development by intercellular communication within the follicular niche; biology of the Sertoli cell in the fetal, pubertal and adult mammalian testis; mechanisms regulating spermatogonial differentiation; stem cells in mammalian gonads; the role of microRNAs in cell differentiation during gonad development; human sex development and its disorders; as well as methods for the study of gonadal development.

Book excerpt: The first of its kind, this reference gives a comprehensive but concise introduction to epigenetics before covering the many interactions between hormone regulation and epigenetics at all levels. The contents are very well structured with no overlaps between chapters, and each one features supplementary material for use in presentations. Throughout, major emphasis is placed on pathological conditions, aiming at the many physiologists and developmental biologists who are familiar with the importance and mechanisms of hormone regulation but have a limited background in epigenetics.

Book excerpt: Some investigators have hypothesized that estrogens and other hormonally active agents found in the environment might be involved in breast cancer increases and sperm count declines in humans as well as deformities and reproductive problems seen in wildlife. This book looks in detail at the science behind the ominous prospect of "estrogen mimics" threatening health and well-being, from the level of ecosystems and populations to individual people and animals. The committee identifies research needs and offers specific recommendations to decision-makers. This authoritative volume: Critically evaluates the literature on hormonally active agents in the environment and identifies known and suspected toxicologic mechanisms and effects of fish, wildlife, and humans. Examines whether and how exposure to hormonally active agents occursâ€"in diet, in pharmaceuticals, from industrial releases into the environmentâ€"and why the debate centers on estrogens. Identifies significant uncertainties, limitations of knowledge, and weaknesses in the scientific literature. The book presents a wealth of information and investigates a wide range of examples across the spectrum of life that might be related to these agents.

Book excerpt: The Retinoids, Volume 1 covers the chemistry and biology of retinoids, with an emphasis on the role of retinoids in nutrition and in vision. After briefly discussing the discovery and nomenclature of retinoids, this six-chapter volume describes the chemical and physical properties of natural and synthetic retinoids, as well as the retinoidal benzoic acid derivatives. The book goes on describing various reactions with radioisotopes for the synthesis of retinoids and related compounds. Considerable chapters explain the chemical, physical, and biological methodologies for separating and measuring retinoids. A discussion on the relationships between structure and activity of retinoids is included. The last chapter addresses the role of vitamin A in animal and human nutrition. This volume also discusses the metabolism of vitamin A in normal and disease states, as well as its interaction with hormones, micronutrients, drugs, and alcohol. This volume is an ideal source for nutritionists, clinicians, and researchers who are interested in the progressing field of retinoid research.
