Book excerpt: Traffic jam (tj) encodes a soma-specific Maf transcription factor required for morphogenesis of the gonads in Drosophila melanogaster. I examined the requirement of tj function throughout oogenesis by studying the expression of follicle cell type specific markers in tj mutant cell clones. I also examined the interaction between tj and JAK-Stat pathway genes using a variety of loss- and gain-of-function mutations. As well as regulating the expression of a number of adhesion molecules, TJ appears to be a general requirement in follicle cells for their proper differentiation throughout oogenesis. Depending on when cells lose tj, their subsequent differentiation is blocked or defective. This suggests that like other Mafs, TJ is a regulator of differentiation. Furthermore, my findings suggest that TJ interacts with the JAK-Stat pathway during oogenesis. I have also identified a possible interaction between TJ and the EGF receptor signaling pathway.

Book excerpt: During animal development precise coordination and regulation of cell proliferation, differentiation and cell death is required for proper tissue organization. This is achieved through specific cell communication by intercellular signals. Cell death, for example, is a mechanism utilized by multicellular organisms for several developmental processes such as elimination of damaged cells or morphological shaping. Apoptosis can be induced by intrinsic signals generated within the cells or from extrinsic signals received from the surrounding environment. This work centered on the analysis of the mechanisms and signals that trigger apoptosis during Drosophila oogenesis. Drosophila ovaries are composed of approximately 16-20 ovarioles, each of which contains a series of egg chambers that are proceeding through the 14 stages of oogenesis moving from the germarium toward the oviduct. For the mature egg to be formed cell death has to occur at specific stages both in the germline (nurse cells) and in the somatic cells (follicle cells). However, whether this apoptosis is caused by intrinsic or extrinsic signals is not known. In addition to this developmentally controlled cell death, apoptosis can also be induced by environmental cues. Under starvation, for example, there is an increase of apoptosis at particular stages: in the germarium and at mid-oogenesis. Mid-oogenesis (stage8/9) is when vitellogenesis starts. At these stages the state of the egg chambers are checked in order to eliminate, through apoptosis of the germline, defective egg chambers. In starved flies, through activation of this "check-point", oogenesis is blocked before vitellogeneis starts. It is not clear what are the signals that prevent apoptosis at mid-oogenesis in well-fed flies. In this study I have analyzed the function of a newly identified signaling molecule CG7447 during Drosophila oogenesis. My results indicate that CG7447 is required to prevent apoptosis at mid-oogenesis in well-fed flies. CG7447 R.

It plays a particularly important role in early embryonic development in organisms that rely heavily on maternally supplied mRNAs. The Drosophila developing oocyte serves as a valuable model for studying translational control. The Drosophila posterior determinant, nanos (nos), is translationally repressed throughout the oocyte cytoplasm except at the posterior pole. A multi-functional RNA-binding protein Glorund (Glo), the homolog of the mammalian hnRNP F/H family of proteins, represses translation of nos during oogenesis by targeting both translation initiation and translation elongation. To elucidate the molecular mechanism by which Glo regulates nos, I identified dFMRP as a Glo-interacting protein. By biochemically dissecting repression of nos translation in vitro, I demonstrated that dFMRP specifically inhibits translation elongation. Furthermore, I combined mutational analysis and in vivo and in vitro binding assays to show that Glo's qRRM2 domain specifically and directly interacts with dFMRP, suggesting that Glo's RNA-binding domains can also function as protein-protein interaction interfaces critical for its regulatory functions. Additionally, I applied ribosome footprint profiling to the Drosophila ovary at different developmental stages to identify maternal transcripts regulated during the translation elongation phase. A footprint peak-finding tool has been developed to detect transcriptome-wide ribosome stalling sites. My preliminary results suggest that developmentally regulated ribosome stalling sites may be widely present on Drosophila maternal transcripts. An A-/P-site mapping algorithm is currently under development to facilitate mechanistic analysis of ribosome stalling during oogenesis.

Book excerpt: In this dissertation, I examined the molecular mechanism of function of the Drosophila melanogaster protein Tribbles (Trbl) during oogenesis and larval development. Trbl is the founding member of an evolutionarily conserved family of kinase proteins that play diverse roles in cell signaling and energy homeostasis. In addition to the central Serine/Threonine kinase domain, members of the Tribbles gene family (Trib) shares C terminal mitogen activated protein kinase kinase MEK1 and E3 ubiquitin ligase COP1 binding motifs, the latter required for the degradation of target proteins via proteasome. During oogenesis, Trbl controls border cell (BC) cluster migration by mediating degradation of C/EBP transcription factor Slbo. I first investigated Trbl's role during oogenesis using a Trbl specific antisera. Trbl localizes to the nucleus of main body follicle cells (MBFC) up to stage 10 of oogenesis. In the case of BC, Trbl expresses in a complementary pattern to Slbo expression. The Trbl level decreases gradually as the BC cluster delaminates from the epithelium and starts migrating when Slbo protein level increases. Moreover, Slbo was found to be essential but not sufficient to decrease the Trbl level required for BC migration. In a wing misexpression screen for Trbl interacting proteins, I identified the Serine/Threonine protein kinase Akt, a major mediator of insulin signaling. In recent years, mammalian Trib3 and Trib2 proteins have been implicated in the regulation of Insulin signaling by inhibiting the activating phosphorylation of Akt, Given the central role of Akt in insulin signaling, I tested whether the function of Trib family in insulin signaling is evolutionarily conserved. Using Drosophila larval development as a model system, I found that Trbl has a conserved role in binding and inhibiting Akt phosphorylation-activation, implicating Trib proteins as novel sites of signaling pathway integration that link nutrient availability with cell growth and proliferation. Finally, I have identified a previously unknown motif (R141) in Trib proteins essential for their function to regulate insulin signaling mediated growth and metabolism.
