Today, however, whole classes of pesticide chemistry have fallen by the wayside due to misuse which generated resistance problems reaching crisis proportions, severe adverse effects on the environment, and public outcry that has led to increasingly stricter regulation and legislation. It is with this background, demanding the need for safer, environmentally friendly pesticides and new strategies to reduce resistance problems, that this book was written. The authors of the various chapters have a wealth of experience in pesticide chemistry, biochemical modes of action, mechanism of resistance and application, and have presented concise reviews. Each is actively involved in thedevelopment of new groups of pesticide chemistry which led to the development of novel insecticides with special impact in controlling agricultural pests. Emphasis has been given to insecticides with selective properties, such as insect growth regulators hormone mimics, ecdysone agonists), (chitin synthesis inhibitors, juvenile chloronicotinyl insecticides (imidacloprid, acetamiprid), botanical insecticides (neem, plant oils), pymetrozine, diafenthiuron, pyrrole insecticides, and others. It is against this background that the authors of this book present up-to-date findings-relating to biochemical sites that can serve as targets for developing insecticides with selective prop erties, and as the basis for the elucidation of resistance mechanisms and countermeasures. The book consists of eight chapters relating to biochemical targets for insec ticide action and seven chapters relating to biochemical modes of resistance and countermeasures. The authors of the chapters are world leaders in pesti cide chemistry, biochemical modes of action and mechanisms of resistance. Biochemical sites such as chitin formation, juvenile hormone and ecdysone receptors, acetylcholine and GABA receptors, ion channels, and neuropeptides are potential targets for insecticide action. The progress made in recent years in molecular biology (presented in depth in this volume) has led to the iden tification of genes that confer mechanisms of resistance, such as increased detoxification, decreased penetration and insensitive target sites. A combina tion of factors can lead to potentiation of the resistance level. Classifications of these mechanisms are termed gene amplification, changes in structural genes, and modification of gene expression.

He obtained grow ing cells from pupal ovaries of the emperor gum moth, Antheraea euca lypti. At that time, I was trying to obtain growing cells from leafhoppers. Grace's method could not be applied directly to my culture because of the differences in species, the size of the insects, and the tissue to be cul tured. The vertebrate tissue culture methods gave me some ideas for pre paring cultures from leafhoppers, but those could not be used directly either. There were no textbooks and no manuals for invertebrate tissue culture, so I had to develop a method by myself. First, I considered what type and what size of vessels are suitable for insect tissue culture. Also, I had to look for suitable materials to construct the culture vessels. Sec ond, I had to examine various culture media, especially growth-promot ing substances, such as sera. Then I had to improve culture media by trial and error. The procedure to set up a primary culture was also a problem. How could I sterilize materials? How could I remove tissues from a tiny insect? How many tissues should I pool in order to set up one culture? I had to find out the answers. Naturally, it took a lot of time.

Book excerpt: The application of organic insecticides in the agrotechnical praxis resulted in a great and unexpected progress in the control of insect pests, and was of a great economical value all over the world. The widespread application of these agents, however, is also accompanied by negative effects. The principal drawback of classical insecticides consists in the lack of their specificity, the useful insects being killed together with insect pests. Furthermore, the broad-scale application for many years led to the formation of more resistant insect strains requiring higher and higher doses of insecticides. The residues of the mostly used chlorinated compounds accumulate in human and animal foods producing directly or indirectly harmful effects in human sub jects. The critical situation led in many developed countries to the restriction in the usage of some types of classical insecticides. Under these circumstances it is quite natural that novel routes for the control of insect pests are investigated. In this connection, attention has been paid especially to the insect endocrinology and insect hormones which regulate the admirable and in many regards specific development from the egg to the adult insect. The recent successful discoveries in this field are thus in close relation to the practical requirements. Isolation and identification of moulting hor mones and juvenile-hormone-like naturally occurring substances not only made possible an exact investigation of their physiological effects but also stimulated the chemical research.
