The physiological effect of cycling temperatures on electrolyte balance in skeletal muscle and plasma of 1% - year-old rainbow trout ( Salmo gairdneri ) was studiedc The "cycling fish" were maintained in a large tank in which the water temperature cycled from S^C to 18°C daily, closely simulating daily temperature conditions in several Alberta mountain streams. After 42 days of exposure to the cycle, the fish were sampled at various temperatures. Muscle and plasma samples were analyzed for sodium, potassium, chloride, and water content. Six other groups of control trout were acclimated for 42 days at six different constant temperatures which coin cided with the same temperatures of the cycling fish. Muscle water content in the cycling fish was signifi cantly lower at 8°C of the cycle, but did not change signi ficantly throughout the rest of the cycle. The control fish showed a significant drop in muscle water content at 16°C and 18°C which corresponded to a rapid growth rate at these temperatures. Muscle chloride content in the cycling and control fish showed nearly constant levels even though large fluctuations in plasma chloride levels occurred. A reciprocal relationship was found between the muscle sodium and potassium levels in the cycling fish. Levels of plasma sodium, chloride, and potassium in the cycling fish were similar to the levels in the control fish held at low temperatures. Cycling fish appeared to be like cold adapted fish, in their water and electrolyte metabolism, which had acclimated to the lower end of the daily temperature cycleo It was also found that plasma chloride binding occurred in fish acclimated to temperatures of 4°C but not at 12°C. Because it appears that chloride does not distribute itself according to a Donnan distribution, the chloride space and chloride-potassium space cannot serve as valid estimates of extracellular fluid volumec The sodium space in almost all cases was smaller than the chloride space or the chloride- potassium space and was therefore used as an estimate of extracellular fluid volumeo In the controls and cycling fish the sodium space decreased significantly with either an increase in holding temperature or an increase in temperature of the cycle.

Book excerpt: The study of thermoregulation in endotherms has contributed much to the emergence of the concept of control theory in biology. By the same token, the study of tempera ture adjustment in ectotherms is likely to have a far-reaching influence on ideas on the regulation of metabolism in general. The reason for this is that ectotherms, in adapting to the vagaries of a thermally unstable environment, deploy a range of subtle molecular and organismic strategies. Thus the experimenter, using temperature changes as a tool, is well equipped to analyze some of these strategies. This approach has enabled some important mechanisms of temperature-induced adaptation to be elucidated; the most striking of these are the effects on metabolism of changes in the conformation of enzymes and the transfer properties of membranes. Furthermore, there is a vague but persistent feeling among those working in this field that changes in the nervous system will ultimately prove to be the agency by which many of the molecular mechanisms of temperature adaptation are controlled. Should this indeed be the case, a new phase would soon begin in our understanding of the interactions between the systemic and the cellular levels of organization. However, it is not only questions about the causes of temperature adaptation that can provide answers of potential importance to the general biologist; of equal significance are questions as to the meaning of temperature adaptation in a particular organism.

Book excerpt: The primary objective of this investigation was that of providing a comprehensive tissue-by-tissue assessment of water-electrolyte status in thermally-acclimated rainbow trout, Salmo gairdneri. To this end levels of water and the major ions, sodium, chloride and potassium were evaluated in the plasma, at three skeletal muscle sites, and in cardiac muscle, liver, spleen, gut and brain of animals acclimated to 2°, 10° and 18°C. The occurrence of possible seasonal variations in water-electrolyte balance was evaluated by sampling sununer and late fall-early winter populations of trout. On the basis of values for water and electrolyte content, estimates of extracellular and cellular phase volumes, cellular electrolyte concentrations and Nernst equilibrium potentials were made. Since accurate assessment of the extracellular phase volume is critical in the estimation of cellular electrolyte concentrations and parameters based on assumed cellular ion levels, [14 C]-polyethylene glycol-4000, which is assumed to be confined to the extracellular space, was employed to provide comparisons with various ion-defined spaces (H20~~s, H20~~/K and H20~~s). Subsequently, the ion-defined space yielding the most realistic estimate of extracellular phase volume for each tissue was used in cellular electrolyte calculations. Water and electrolyte content and distribution varied with temperature. Tissues, such as liver, spleen and brain appeared to be the most thermosensitive, whereas skeletal and cardiac muscle and gut tissue were less influenced. 'Summer' series trout appeared to be more capable of maintaining their water- electrolyte balance than the ~fall-winter' series animals. i The data are discussed in terms of their possible effect on maintenance of appropriate cellular metabolic and electrophysiological functions.
