Available in PDF, EPUB and Kindle. Book excerpt: This book provides a resource of current understandings about various aspects of the biology of spermatogonia in mammals. Considering that covering the entire gamut of all things spermatogonia is a difficult task, specific topics were selected to provide foundational information that will be useful for seasoned researchers in the field of germ cell biology as well as investigators entering the area. Looking to the future, the editors predict that the foundational information provided in this book -- combined with the advent of new tools and budding interests in use of non-rodent mammalian models -- will produce another major advance in knowledge regarding the biology of spermatogonia over the next decade. In particular, we anticipate that the core molecular machinery driving different spermatogonial states in most, if not all, mammals will be described fully, the extrinsic signals emanating from somatic support cell populations to influence spermatogonial functions will become fully known, and the capacity to derive long-term cultures of SSCs and transplant the population to regenerate spermatogenesis and fertility will become a reality for higher order mammals.

The structure-function relationship of sperm has been studied from various viewpoints. Accumulated evidence has shown that the sperm components undergo sequential changes from the beginning of spermatogenesis to the time of fertilization/embryogenesis. Structural analyses have been performed using various new techniques of light and electron microscopy as well as immunohistochemistry and immunocytochemistry in combination with specific probes such as antibodies against sperm components. Recently developed gene-manipulation techniques have accelerated investigations on the events that govern the relationship between the structure and molecular components of sperm. In addition, animal models with gene manipulations have been shown to exhibit various morphological and functional abnormalities that lead to infertility. In this book, I discuss the events that occur in the normal sperm head and govern the structure-function relationship from the time of spermatogenesis to that of fertilization or egg activation. In this regard, I describe dynamic modifications and maturation events occurring in sperm-head components and compare the outcomes of these events with the outcomes of their failure.

Available in PDF, EPUB and Kindle. Book excerpt: In the past thirty years, significant advances have been made in the field of reproductive biology in unlocking the molecular and biochemical events that regulate spermatogenesis in the mammalian testis. It was possible because of the unprecedented breakthroughs in molecular biology, cell biology, immunology, and biochemistry. In this book entitled, Molecular Mechanisms in Spermatogenesis, a collection of chapters has been included written by colleagues on the latest development in the field using genomic and proteomic approaches to study spermatogenesis, as well as different mechanisms and/or molecules including environmental toxicants and transcription factors that regulate and/or affect spermatogenesis. The book begins with a chapter that provides the basic concept of cellular regulation of spermatogenesis. A few chapters are also dedicated to some of the latest findings on the Sertoli cell cytoskeleton and other molecules (e.g., proteases, adhesion proteins) that regulate spermatogenesis. These chapters contain thought-provoking discussions and concepts which shall be welcomed by investigators in the field. It is obvious that many of these concepts will be updated and some may be amended in the years to come. However, they will serve as a guide and the basis for investigation by scientists in the field.

Book excerpt: Spermiogenesis summarizes the process of sperm structure and development in a variety of organisms. It attempts to illustrate the structure of the organelles that characterize mature spermatozoa and to trace the morphogenesis of these organelles during sperm formation. This task is complicated by considerable variations in the structure of spermatozoa among different species and the lack of good morphological studies of sperm structure and spermiogenesis in many animal groups. The general morphological features of mammalian sperm structure differ in some ways from the features of insect sperm, the spermatozoa produced by insects appearing relatively more simple. When the functions and developmental regions of the organelles are considered, homologies between complex structures found in mammalian sperm and simpler insect sperm structures may be deduced even though the organelles in question are quite dissimilar in appearance. Such homologies will be stressed in an effort to present as general a picture of spermiogenesis as seems consistent with the species-specific variations that exist. This book will be a useful, stimulating reference for students of reproductive biology.

Book excerpt: Scientists investigating germ cells have, over the past 15 years, originated discoveries and innovations that give us valuable insights into the mechanisms that regulate not just stem cell function, but human development in its widest sense. With contributions from some of the leading researchers in the field, Male Germline Stem Cells: Developmental and Regenerative Potential assesses the implications of these discoveries for understanding the fundamental biology of germline stem cells as well as their potential for human stem cell-based therapies. This monograph covers many of the fundamental issues now being explored by today’s generation of stem cell researchers, including the field’s potential for regenerative medicine. Ranging from an assessment of the pluripotency of primordial germ cells and their possible applications in treating testicular cancer, to the recovery of once-mordant fertilization-competent sperm, this volume has it all. It is a reference point for any scientist involved in related research as well as being a timely summation of what could prove to be a hugely exciting and very fruitful area of inquiry.

Book excerpt: The conference represented by this book was made possible by support from NICHD and a planning committee headed by Dr. Richard Sherins. Two general areas of research are included: the first encompasses steroid hormone synthesis, metabolism and transport in the testis; and the second relates to hormonal regula tion of the seminiferous tubule with special emphasis on the con trol of Sertoli cell function. In addition, there are sections on the purification of unique testicular proteins and morpho logical studies with particular emphasis on the Sertoli cell. We would like to express our sincere thanks to Dr. Sherins and his staff at NICHD and to all of the people at the University of North Carolina who participated in the Conference arrangements, to Dr. Judson J. Van Wyk, Chief of the Pediatric Endocrinology Division, and Dr. H. Stanley Bennett, Director of the Laboratories for Reproductive Biology. Our very special thanks to Mrs. Carolyn Jaros for her help in handling the local arrangements. Mrs. Martha Byrd and Mrs. Linda Rollins typed the manuscripts. Miss Leslie Wells and Mr. Albert Smith kindly assisted in proof reading, and Dr. Elizabeth Wilson gave much help with the final editing process. To all of these people, we are most grateful.
