Book excerpt: The effectiveness of image restoration techniques for use in high-pressure fluorescence microscopy are investigated and characterized. A quartz capillary, which acts as both a mechanical support and an optical window, is utilized as a high-pressure imaging chamber. We apply point-spread-function deconvolution using both a capillary and a flat slide control to analyze the relative contrast and resolution enhancement in fluorescent images. The asymmetry in the lateral and axial directions is examined for all point spread functions. Spatial deconvolution is shown to be an effective method at high pressures to reduce out-of-focus fluorescence in microscope images by reversing the effects of optical aberrations. The results confirm there is significant image improvement using either low- or high-numerical aperture objectives. Lateral resolution is improved over raw images by applying calculated and measured point spread functions, with both processes yielding comparable results.
