Available in PDF, EPUB and Kindle. Book excerpt: During the past two decades research on the testis has created a vast number of publications. New techniques, such as cell and gene clon ing, germ cell manipulation and transplantation, as well as gene ex pression, have surpassed the classical approaches that involved studies on receptor activation and endocrine, paracrine and autocrine regula tory mechanisms. Due to the expansion of this field of research it has become increasingly difficult to integrate the large number of observa tions and relate them to the physiology and pathophysiology of the tes tis. More emphasis must, therefore, be put on the integration of isolated findings and the construction of the total picture. In order to stimulate this specific mental activity we have named this book Testicular Tan gram. We hope that the chapters in this book will be used as pieces of the testicular puzzle and that the reader enjoys composing new shapes. The chapters of this book are representative of the plenary lectures presented at the I ih European Workshop on Molecular and Cellular Endocrinology of the Testis held in Doorwerth The Netherlands, from April 6 - I 0, 2002. Publication of this book has been made possible by the generous support of the Ernst Schering Research Foundation. We would also like to thank the distinguished scientists, who have pro vided us with their excellent manuscripts well before the start of the Workshop. We acknowledge the help of Dr. U. -F. Book excerpt: Combined oral contraceptives are the most convenient and accepted method of hormonal contraception. Nevertheless, the medical community and consumers constantly demand innovation, additional benefits during use and lower hormonal load despite the high safety profile of available products. At the Ernst Schering Research Foundation Workshop 52, new perspectives and mechanisms for tissue-selective, estrogen-free contraception were discussed. The aim of the workshop was to bring together experts in the field of molecular and pharmacodynamic action of progestins with clinicians and medical experts to discuss potential medical endpoints, physiological reactions and (bio)marker useful describing the tissue selectivity and the contraceptive action of new progestins in different target organs. A major factor for successful realization of these new concepts is a deeper understanding of local pharmacological responses to progestins in general and to new progestins in particular.

Available in PDF, EPUB and Kindle. Book excerpt: Personalized medicine employing patient-based tailor-made therapeutic drugs is taking over treatment paradigms in a variety of ?elds in oncology and the central nervous system. The success of such therapies is mainly dependent on ef?cacious therapeutic drugs and a selective imaging probe for identi?cation of potential responders as well as therapy monitoring for an early bene?t assessment. Molecular imaging (MI) is based on the selective and speci?c interaction of a molecular probe with a biological target which is visualized through nuclear, magnetic resonance, near infrared or other methods. Therefore it is the method of choice for patient selection and therapy monitoring as well as for speci?c e- point monitoring in modern drug development. PET (positron emitting tomography), a nuclear medical imaging modality, is ideally suited to produce three-dimensional images of various targets or processes. The rapidly increasing demand for highly selective probes for MI strongly pushes the development of new PET tracers and PET chemistry. ‘PET chemistry’ can be de?ned as the study of positron-emitting compounds regarding their synthesis, structure, composition, reactivity, nuclear properties and processes and their properties in natural and - natural environments. In practice PET chemistry is strongly in?uenced by the unique properties of the radioisotopes used (e. g. , half-life, che- cal reactivity, etc. ) and integrates scienti?c aspects of nuclear-, organic-, inorganic- and biochemistry.

Book excerpt: The current progress in molecular medicine allows the identification of a plethora of new and often human-specific drug targets. An early in vivo validation of specific ligands binding to these targets in humans is needed to assess their potential for targeted imaging and radiotherapy. Radiopharmaceuticals are uniquely suitable for such target validation studies. The purpose of the Ernst Schering Research Foundation Workshop 48 was to offer a forum for an open exchange on the state of the art in the early development of such radiopharmaceuticals. Experts from academia, industry and regulatory authorities provided contributions covering the identification of targets, the necessary preclinical studies on the safety of ligands as well as their validation in human clinical trials.

Available in PDF, EPUB and Kindle. Book excerpt: The successful treatment of acute stroke remains one of the major challenges in clinical medicine. Over the last decades, the understanding of stroke pathophysiology has greatly improved, while the therapeutic options in stroke therapy remain very limited. Today, hyperacute mechanisms of damage, such as excitotoxicity, can be discriminated from delayed ones, such as inflammation and apoptosis. Targeting of inflammation has already been successfully applied in various stroke models, but translation into a clinically efficacious strategy has not been achieved so far. In this book, leading experts in basic cerebrovascular research as well as stroke treatment review the current evidence for and against an important role for inflammation in stroke, and explore the potential of treating or modulating inflammation in stroke therapy.

Available in PDF, EPUB and Kindle. Book excerpt: An essential component of inflammation is the migration of circulating leukocytes from blood into tissues. This process is characterized by a multistep paradigm of sequential cell adhesion and activation events that lead to the extravasation of specific leukocyte subsets to different tissues in health and disease. The first step of leukocyte extravasation, the rolling of leukocytes, is primarily mediated by the interactions of selectins and their ligands. It has recently become evident that fucosyltransferases are crucial for selectin ligand synthesis, inflammation, and skin homing. This book provides an in-depth overview of the mechanisms of leukocyte trafficking and of the molecular mechanisms of selectin/selectin ligand interactions and discusses options for pharmacological intervention to treat inflammatory diseases.

Available in PDF, EPUB and Kindle. Book excerpt: Methylation of DNA at cytosine residues as well as post-translational modifications of histones, including phosphorylation, acetylation, methylation and ubiquitylation, contribute to the epigenetic information carried by chromatin. These changes play an important role in the regulation of gene expression by modulating the access of regulatory factors to the DNA. The use of a combination of biochemical, genetic and structural approaches has allowed demonstration of the role of chromatin structure in transcriptional control. The structure of nucleosomes has been elucidated and enzymes involved in DNA or histone modifications have been extensively characterized. Since deregulation of epigenetic marks has been reported in many cancers, a better understanding of the underlying molecular mechanisms bears the promise that new drug targets may soon be found. The newest developments in this quickly developing field are presented in this book.

Book excerpt: Since the first successful transfer of an in vitro fertilised human egg in 1976, modern endocrinology, genetics, and assisted reproductive technologies have opened new frontiers of research with the aim to treat infertile women. In this workshop we set out to promote an interdisciplinary discussion between experts from various fields of basic, company-based and clinical research related to folliculogenesis and oocyte development. The aim of this workshop was to present, discuss and assess novel approaches in mammalian folliculogenesis and oocyte development that may have an impact on fertility/ infertility in the near or distant future. Key issues were the understanding of new modulators of folliculogenesis and regulators of cytoplasmic as well as meiotic oocyte maturation, modern technologies, the aging oocyte and pathogenetic mechanisms of infertility.

Book excerpt: Pharmaceutical companies are spending increasing amounts of money on drug discovery and development. Nevertheless, attrition rates in clinical development are still very high, and up to 90% of new compounds fail in clinical phase I - III trials, which is partially due to lack of clinical efficacy. This indicates a strong need for highly predictive in vitro and in vivo models. The "50th International Workshop of the Ernst Schering Research Foundation" focussed on "Animal Models of T Cell-Mediated Skin Diseases". Such animal models should have impact not only on inflammatory dermatoses but also on other inflammatory disorders due to their model character. The current volume summarises recent advances in animal research that are important for anti-inflammatory drug discovery.
