Book excerpt: Bromochloroacetic acid is a water disinfection by-product. Bromochloroacetic acid was nominated for toxicity and carcinogenicity studies in rats and mice because of widespread human exposure and because a related dihaloacetate, dichloroacetate, was found to be carcinogenic to the liver of rats and mice. Drinking water was selected as the route of exposure to mimic human exposure to this chemical. Male and female F344/N rats and B6C3F1 mice were exposed to bromochloroacetic acid (greater than 95% pure) in drinking water for 2 weeks, 3 months, or 2 years. Genetic toxicology studies were conducted in ¿Salmonella typhimurium,¿ ¿Escherichia coli,¿ and peripheral blood erythrocytes of exposed mice. Illustrations.

Available in PDF, EPUB and Kindle. Book excerpt: Disinfection By-Products and Human Health is based on contributions from speakers who participated in May 2011 workshops on Disinfection By-Products (DBPs) and Human Health at Ozwater 11 in Adelaide, Australia or at an AWA sponsored workshop at the Curtin Water Quality Research Centre, Perth, Australia. The contributions are prepared to facilitate communication with practitioners, rather than researchers, making use of overview illustrations rather than dense text or data tables. Each chapter concludes with up to 5 key findings that are take-home messages for practitioners. Disinfection By-Products and Human Health is aimed specifically at drinking water professionals (engineers, chemists and public health professionals) working on the front lines of drinking water issues where they must encounter actual day-to-day issues of risk management concerning DBPs in relation to all the other regulatory and water quality issues they must manage. Although a topic this complex is certainly not amenable to simplistic explanations, this book aims to provide drinking water professionals with a pragmatic assessment of the current evidence and emerging issues concerning DBPs and public health. Disinfection By-Products and Human Health is an essential, practical and accessible guide for drinking water professionals, engineers, chemists and public health professionals. Editors: Steve E. Hrudey, Professor Emeritus, Analytical & Environmental Toxicology, University of Alberta, Canada, Jeffrey W.A. Charrois, Director and Associate Professor, Curtin Water Quality Research Centre, Curtin University of Technology, Australia, Steve Hrudey is professor emeritus in analytical and environmental toxicology in the University of Alberta's Faculty of Medicine & Dentistry. He spent 13 years as a cabinet-appointed member of the Alberta Environmental Appeals Board, the last four as chair, and was the first non-lawyer to hold this position. During this period, he served on 36 public hearing panels, 19 as chair of the panel. In addition he has testified before senate committees in Canada and the Legislative Council in Western Australia. Hrudey has served on a number of high-profile expert panels, including the Research Advisory Panel to the Walkerton Inquiry (2000-2002), the Expert Panel on Safe Drinking Water for First Nations (2006), the Technical Advisory Committee to the B.C. Minister of Health on turbidity and microbial risk in drinking water (2007-2008, as chair), the Expert Advisory Panel on Water Quality for Washington, D.C., to the U.S. Army Corps of Engineers (2009-2011) and chair of the Royal Society of Canada Expert Panel on Environmental and Health Impacts of Canada's Oil Sands Industry (2009-2010). He has also co-authored or edited nine books, including the widely acclaimed book inspired by the Walkerton tragedy: Safe Drinking Water - Lessons from Recent Outbreaks in Affluent Nations (IWA Publishing, 2004). He has written 26 book chapters, 19 expert panel reports, 163 refereed journal articles, 15 science discussions, six media op-eds and 73 conference proceeding papers. Hrudey is the 2012 winner of the American Water Works Association A.P. Black Research Award for contributions to water science and water supply. The requirements for supplying water to a home, a city or a factory can be very different. Experts in these fields explain the nuances of the details involved in maintaining adequate quantity and quality for these different consumers. Waste water management can be of even greater concern, yet its management can follow similar paths when compared to sophisticated water supply treatment. Both the physics and chemistry of these fields are fully covered. Volume 2 deals with the big picture of regional water supplies, how they become contaminated, how they can be protected and how they can best serve the surrounding populations and industries. Significant focus is placed upon the natural chemistry of available water supplies and its biological impacts. Case studies from regions around the world offer an excellent picture of the world's water resources.

Specific sections examine this from both a toxicological and analytical perspective. Within these sections, the book defines appropriate strategies to both assess and ultimately control genotoxic impurities, thus aiding the reader to develop effective control measures. An opening section covers the development of guidelines and the threshold of toxicological concern (TTC) and is followed by a section on safety aspects, including safety tests in vivo and vitro, and data interpretation. The second section addresses the risk posed by genotoxic impurities from outside sources and from mutagens within DNA. In the final section, the book deals with the quality perspective of genotoxic impurities focused on two critical aspects, the first being the analysis and the second how to practically evaluate the impurities.

Available in PDF, EPUB and Kindle. Book excerpt: This volume will provide a contemporary account of advances in chemical carcinogenesis. It will promote the view that it is chemical alteration of the DNA that is a route cause of many cancers. The multi-stage model of chemical carcinogenesis, exposure to major classes of human carcinogens and their mode-of-action will be a focal point. The balance between metabolic activation to form biological reactive intermediates and their detoxification, ensuing DNA-lesions and their repair will be profiled. It will describe the chemical changes that occur in DNA that result from endogenous insults including epigenetic changes that lead to gene silencing. It will describe major mechanisms of mutagenesis, affects on tumor suppressor genes and proto-oncogenes, and how cell-cycle check points can be by-passed by the "stealth-like" properties of chemical carcinogens. Environmental agents that can promote tumor formation will be discussed. The monograph will have wide appeal as a knowledge base for graduate students, post-doctoral fellows and faculty interested in this aspect of cancer causation and research.

Available in PDF, EPUB and Kindle. Book excerpt: Atlas of Histology of the Juvenile Rat should be of interest to toxicologic pathologists, toxicologists, and other biological scientists who are interested in the histomorphology of juvenile rats. For several decades the laboratory rat has been used extensively in nonclinical toxicology studies designed to detect potential human toxicity of drugs, agrochemicals, industrial chemicals, and environmental hazards. These studies traditionally have involved young adult rats that are 8-10 weeks of age as studies are started. It is becoming increasingly apparent that children and young animals may have different responses to drug/chemical exposures, therefore, regulatory agencies are emphasizing toxicology studies in juvenile animals. While the histologic features of organs from young adult and aged laboratory rats are well known, less is known about the histologic features of organs from juvenile rats. Final histologic maturity of many organs is achieved postnatally, thus immature histologic features must be distinguished from chemical- or drug-related effects. While this postnatal organ development is known to exist as a general concept, detailed information regarding postnatal histologic development is not readily available. The Atlas includes organs that are typically sampled in nonclinical toxicology studies and presents the histologic features at weekly intervals, starting at birth and extending through postnatal day 42. - Written and edited by highly experienced, board-certified toxicologic pathologists - Includes more than 700 high-resolution microscopic images from organs that are typically examined in safety assessment toxicology studies - Detailed figure legends and chapter narratives present the salient features of each organ at each time interval - Figures are available for further study via Elsevier's Virtual Microscope, which allows viewing of microscopic images at higher magnification - Valuable resource for toxicologic pathologists who are confronted with interpretation of lesions in juvenile rats in situations where age-matched concurrent controls are not available for comparison, e.g., with unscheduled decedents - Figures are available for further study on ScienceDirect with Virtual Microscope, which allows viewing of microscopic images at higher magnification
