The first approach entails a racemic transannular nitrone-olefin [3+2] cycloaddition from nitrone 238. Construction of the nitrone 238 began with aldehyde 241. Another key feature in this route involved a ring-closing metathesis for the formation of a 14-membered ring. The route ended upon formation of a key diol 237. The second approach incorporated the C14 methyl group at an early stage to probe its influence upon an intramolecular nitrone-olefin [3+2] cycloaddition. The approach began with dithiane 258. A key feature in the second plan involved asymmetric induction at the C5 carbon through a conjugate addition of an azide to an [alpha, beta]-unsaturated imide to form azide 290 with both excellent yield and stereocontrol for this addition. The route was advanced to oxaziridine 299. Conversion of 299 to the nitrone 300 and ultimately the intramolecular nitrone-olefin [3+2] cycloaddition was unsuccessful. The third and final approach involved using the basic plan of our first successful approach and employing the asymmetric induction of the second approach. This blending of strategies led to an asymmetric intermediate that links up with the first approach. The route began with dithiane 258. Again, the use of an azide addition to a [alpha, beta]-unsaturated imide led to azide 309. Upon formation of the 14-membered lactone, the azide function was first reduced and protected as carbamate 320 which in turn, increased the yield substantially over this same transformation from the first route. Amine 318 now stands ready to complete a formal synthesis of halichlorine and pinnaic acid.

Ce cap est franchi, depuis un certain nombre d'années, et le développement des recherches sur les toxines axées sur la thérapeutique est devenu foisonnant. Voyez telle ou telle toxine protéique ternaire, avec ses trois domaines, de fixation sur un récepteur membranaire, de translocation qui lui permet de franchir la membrane cellulaire, et d'activité enzymatique qui va alors pouvoir s'exercer dans la cellule. Qu'on parvienne à dissocier chacun de ces trois domaines en leur conservant leurs propriétés, et des possibilités nouvelles d'utilisation thérapeutique vont s'ouvrir, presque à l'infini semble-t-il. Car chacun d'entre eux pourra être couplé à une molécule active bien ciblée, et leur efficacité sera même améliorée pour peu qu'on parvienne à conjuguer leur action. Qu'un récepteur, qu'une molécule, propres au métabolisme ou à la physiologie d'une cellule cancéreuse soient identifiés, et des toxines conjuguées à un ligand de ce récepteur, de cette molécule (ce peut être un anticorps) bloqueront les canaux ioniques ou exerceront leur effet cytolytique sur cette seule cellule, sans nuire aux cellules saines. Nous voilà dégagés de l'utopie, le contenu de cet ouvrage le prouve abondamment. Et même si la concrétisation des promesses reste délicate, le champ d'action est si vaste initialement qu'il ne saurait rester improductif : il ne l'est d'ailleurs déjà plus.

Few individuals have so fundamentally influenced the science of organic chemistry as Albert Eschenmoser has. Over the years, a sizable fraction of Albert Eschenmoser′s work has been published in Helvetica Chimica Acta, hence the decision of the Editor to produce a special issue of the journal on the occasion of Albert Eschenmoser′s 75th birthday, celebrated on August 5, 2000. The highly impressive set of 67 original research papers submitted in response to the Editor′s invitation could not, for obvious technical reasons, be accommodated in a single issue and were thus divided over the August and September issues. The international reaction to this tribute has been so overwhelmingly enthusiastic that the two issues have been combined in this single volume for separate publication.
