Book excerpt: The modern era of synthetic chemistry can be characterized by increased understanding, development and application of transition metal complexes to traditionally organic transformations. While the field has focused largely on the chemistry of group 4-8 metals (and in particular, rhodium and palladium), there has been growing interest in the past decade in using so-called "coinage metals"--Gold, silver and copper--as catalysts for organic reactions. Gold(I) metal complexes have been shown to effect a range of organic transformations that proceed through [pi]-activation mechanisms, often with greater selectivity than other transition metal complexes. Additionally, in contrast to more established transition metal reactions, gold(I) processes can be conducted at low temperature and in the presence of water and air. Because of their mild conditions and high degree of control, gold(I) reactions are ideal for application to total synthesis. In the first chapter of this text, we discuss the current state of the art of gold(I)-catalysis in the context of complex molecule synthesis. The reactivity exhibited by gold(I) complexes is highly controlled by choice of catalyst ligand. In the second chapter, we discuss the development of ligand-controlled intramolecular reaction of dienes and allenes to afford selectively either [4+2]- or [4+3] cycloadducts. The purpose of this discussion is two-fold: first, to demonstrate the effect that stabilization of reactive intermediates has on the course of gold(I) reactions, and second, to develop methodologies aimed at constructing novel, complex ring structures. In the third chapter of this text, we take one of the reactions discovered in the course of our work in diene-allene cycloadditions and apply it to the total synthesis of antimalarial bisindole alkaloids flinderoles B and C. The key step, a gold(I)-catalyzed hydroarylation of an allene using a C2-indole nucleophile, simulatenously installs three of four unique structural motifs of this natural product.

The reactions of triazoles with metals lead to the synthesis of homoleptic complexes, coordination polymers and metal-organic frameworks (MOFs). These triazole compounds and materials have been employed for a variety of applications in chemistry ranging from nanomaterials to anion recognition. The synthesis of tris(1,2,3-triazolyl)borate completed and complemented the initial research began by Trofimenko in 1966. This ligand was used in metal reactions and found to form both homoleptic complexes and coordination polymers. The connectivity of the tris(1,2,3-triazolyl)borate varied dependent on the transition metal employed. The success of incorporating a triazole onto a borate ligand led to further work utilizing novel triazole derivatives. By combining a borane with a 1,2,3-triazole, a borate-nitrenium that has an overall neutral charge was synthesized. Nitrenium ligands are the nitrogen analog of Arduengo's N-heterocyclic carbene and have only recently been explored. Although initial attempts to synthesize a metal complex with the monodentate neutral nitrenium ligands were unsuccessful, these potential complexes could influence our understanding of transition metal bonds. Metal-organic frameworks were initially synthesized with carboxylates as the organic linkers connected to the metal centers of the scaffold. Recently, triazoles and tetrazoles have been utilized as an alternative for the formation of porous MOFs. The gate-opening effect of some breathing MOFs has been successfully shown with the inclusion of a bis-triazole linker with a rotating central moiety. By extending the central moiety from a phenyl ring to a biphenyl unit on the bis-triazole linker, we were able to demonstrate three separate states of rotation (open/open, open/closed, closed/closed) within three separate MOFs. These separate states were observed through single crystal X-ray diffraction and were the first example of an isostructurally designed family of gate-based breathing MOFs. Spin-crossover has been shown in iron MOFs by incorporating triazoles and tetrazoles within the organic linker. A family of mixed triazole/tetrazole as well as bis-tetrazole ligands were synthesized and tested with metal salts. A coordination polymer was synthesized with these ligands.

Book excerpt: The formation of saturated carbon-carbon bonds in a precise and controlled manner is arguably the principal objective of organic synthesis. Carbocyclic ring systems comprise the underlying structure for the preponderance of natural products and pharmaceutical agents. Therefore, synthetic methods capable of selectively initiating polycyclization processes in the presence of spectating functionality are of significant value, particularly so when multiple stereocenters are formed enantioselectively. The emergence of phosphine gold(I) catalysis over the past decade has opened up new avenues to carbocycle formation via pi activation of alkynes occurring under exceedingly mild conditions and with excellent chemoselectivity. The research described herein describes the use of homogenous gold(I) complexes to initiate electrophilic cyclization cascades. Through rational substrate design, carbocationic centers may be generated in a predictable manner and employed in subsequent intramolecular cyclization processes. Chapter 1 introduces the unique reactivity observed in complexes of gold imparted by its relativistically accelerated valence electrons. One consequence of this perturbation is the linear geometry maintained by gold(I) complexes, minimizing the influence of ligand-based chirality on reactions occurring at coordinated alkynes. In spite of this challenge, moderate levels of enantioselectivity were achieved in the desymmetrization of dienynes by cycloisomerization using chiral bisphosphite gold(I) catalysts. Ultimately, we were able to achieve selectivities up to 98% ee using hindered chiral bisphosphine gold(I) catalysts during the evaluation of another enyne cycloisomerization reaction, described in chapter 2. In this process, an initial regioselective cyclization was used to generate a carbocationic species poised to undergo intramolecular trapping. Consistently high enantioselectivity was maintained using various pendant oxygen, carbon and nitrogen nucleophiles. The diastereomerically pure bi- and tricyclization products obtained provided support for a concerted polyene cyclization mechanism as predicted by the Stork- Eschenmoser postulate. Chapter 3 describes another tandem process exploiting the transient cationic species arising from gold(I)-promoted enyne cycloisomerization. In this case, a gold(I)-initiated tandem cyclopentannulation reaction was employed in the total synthesis of the novel triquinane ventricosene. A cyclopropanol unit embedded in the enyne substrate underwent a semipinacol rearrangement in response to the carbocation, leading cleanly to bicyclo[3.2.0]heptan-6-one products. For cyclopentenyl substrates, the hindered all-carbon quaternary center and all of the ring fusions of the angular triquinane ring system were formed at once. The choice of a hydrocarbon target highlighted the utility of gold(I) catalysts as selective activators of carbon unsaturation; throughout the synthesis only a single heteroatom was present. This work concludes by extending the scope gold(I)-catalyzed carbocyclization reactions which generate useful cationic intermediates. The gold(I)-catalyzed Rautenstrauch rearrangement forms a cyclopentene-based cationic species which was shown to undergo efficient trapping by pendant arenes to give a saturated 5,6-ring fusion comprising a chiral benzylic quaternary center. The chirality transfer observed in the parent process was found to be conserved in the tandem process. Interestingly, cyclization of racemic substrates by chiral bisphosphine digold catalysts was found to proceed with moderate enantioselectivity, suggesting a competing mechanism is in effect which proceeds through an achiral intermediate.

New chiral acyclic diaminocarbenes were also developed and tested along side new C3-symmetric phosphite ligands in an asymmetric intramolecular hydroamination of allenes. Structure activity correlations were developed for the potential use in further rational ligand design. The synthesis of 6a,7-dihydro-5-amino-dibenzo[c,g]chromene derivatives via a gold-catalyzed domino reaction of alkynylbenzaldehydes in the presence of secondary amines was developed. These were sent to be screened for biological activity.

Book excerpt: In modern synthetic chemistry, the formation of multiple C-C bonds in a limited number of steps to access both diversity and complexity has become an important challenge. In recent years, significant research has been devoted to the synthesis of 1,2,3-triazole containing compounds in a multimetal-catalyzed domino sequence due to the remarkable biological activity and pharmaceutical potency of these compounds. Considering the increasing significance of 1,2,3-triazoles in synthetic chemistry, mixing all of the starting components in a one-pot, one-step process to create the final triazole product will be highly efficient with respect to time, cost and simplicity of performance. A number of published papers illustrated the diverse applications of 1,2,3-triazoles (e.g., palladium-mediated couplings and direct C-H bond functionalization). Similarly, this work demonstrates the development of a highly efficient and practical approach to the synthesis of these versatile triazole-containing motifs via a one-pot, one-step cascade process that involves Sonogashira, Heck and carbopalladation/annulation reactions.
