Book excerpt: Stilbenic compounds are natural phytoalexins in some plant species, including grapevine, that play a role in restricting the growth of microbial pathogens. Stilbene synthase (STS) mediates the biosynthesis of stilbenic compounds. The grapevine genome contains a large family of 48 preliminarily annotated STS genes. Therefore, the regulation and function of each STS gene needs to be understood to ascertain their roles in grapevine’s defense against fungal pathogens. In our previous work, we found that nine STS genes are regulated differentially in response to the inoculation of powdery mildew fungus (PM), Erysiphe necator (Schw.) Burr. In this project, the promoter and coding regions of STS22 and STS7 were isolated from Norton and Cabernet Sauvignon grapevines and were introduced into Arabidopsis plants. The expression of each STS transgene was directed by its native promoter or by the constitutive Cauliflower mosaic virus 35S promoter. The results showed that the promoter and intron might work together to influence the expression of STS22/7. Using high-performance liquid chromatography (HPLC) and liquid chromatography/tandem mass spectrometry (LC/MS/MS) to quantify the new compounds, up to 39.57 [mu]g g−1FW of trans-piceid, 12.57 [mu]g g−1FW of cis-piceid and 12.08 [mu]g g−1FW of trans-resveratrol were detected in transgenic Arabidopsis plants. The anti-fungal activity of stilbenes accumulated in transgenic Arabidopsis was investigated. The transgenic lines with high stilbene content showed less susceptibility to Botrytis cinerea. There was no significantly increased resistance to powdery mildew, presumably because of the low stilbene compound level. Therefore, there was a strong positive correlation between the concentration of stilbenes and resistance to pathogen in plants.
