L'un des problèmes sanitaires émergeant est la libération des bactéries pathogènes et résistantes aux antibiotiques dans l'environnement aquatique. Les décharges plasma à la pression atmosphérique sont d'un grand intérêt pour de nombreuses applications. L'une de ces applications est la décontamination microbiologie des liquides. Les travaux développés dans cette thèse participent à mieux appréhender l'intérêt du plasma pour la décontamination microbiologie de liquides et pour la dégradation des gènes de résistance aux antibiotiques. La source utilisée est un multi-jets plasma généré à partir d'un mélange hélium-oxygène à la pression atmosphérique. Différents diagnostics ont été réalisés pour la caractérisation de la décharge plasma. Les mesures de courant et de tension réalisées nous ont permis de déterminer l'énergie du plasma. La spectroscopie d'émission optique mise en oeuvre nous a permis de mettre en évidence et d'identifier les espèces réactives de l'oxygène et de l'azote (RONS) dans le plasma. La création des RONS tels que le peroxyde d'hydrogène, les nitrites et les nitrates dans le liquide modifie la chimie du liquide. Dans ce contexte nous avons quantifié la production de ces espèces dans le liquide. L'interaction plasma-bactérie pour la décontamination microbiologie des liquides a été évaluée à travers trois études distinctes. La première consiste à comprendre l'influence de la phase de croissance des bactéries sur l'efficacité du procédé de décontamination par plasma. Nous avons montré que les bactéries en phase exponentielle sont plus sensibles au traitement plasma que les bactéries issues de la phase stationnaire. De plus, les cellules en phase exponentielle sont plus susceptibles de réduire leur activité métabolique en passant à l'état de bactéries viables mais non-cultivables (VBNC) après un court temps d'exposition au plasma tandis que celles en phase stationnaire acceptent un temps plus long d'exposition au plasma avant d'entrer dans cet état. Pour mieux appréhender les raisons qui peuvent expliquer ces différences, nous avons évalué l'effet du plasma sur l'intégrité de la membrane cellulaire et de l'ADN. Dans la seconde étude, nous avons évalué l'effet de la présence du plasmide sur la sensibilité des bactéries au traitement par plasma. Cette étude a révélé que les bactéries portant un plasmide sont plus sensibles au plasma que celles qui en sont dépourvues. Les bactéries soumises à une intégration de plasmide ont subi plus de dommages membranaires et un taux de ROS intracellulaires plus élevé après exposition au plasma. Nous suggérons que la présence de plasmides présente des inconvénients pour les bactéries en diminuant leur croissance et en imposant un état d'affaiblissement. La dernière étude consiste à évaluer l'effet du plasma sur la dégradation des gènes de résistance aux antibiotiques et sur la réduction des mécanismes de transfert horizontal des gènes (HGT) entre les bactéries donneuses traitées par plasma et celles réceptrices non traitées par plasma. Nous avons démontré que le plasma réduit le nombre de cellules donneuses, ce qui par conséquent diminue la fréquence de transfert conjugatif. Nous avons également conclu que le transfert de plasmides pouvait se produire chez les bactéries VBNC. Toutefois, la prolongation du temps d'exposition entraîne une inhibition de HGT. Bien que le plasma semble être une solution pour la décontamination des liquides et la dégradation des gènes de résistance aux antibiotiques, il serait pertinent de réaliser des études intégrant des conditions qui se rapprochent de l'environnement réel avec la présence de matière organique et en augmentant le volume du liquide traitement.

Book excerpt: Low water activity (aw) and dried foods such as dried dairy and meat products, grain-based and dried ready-to-eat cereal products, powdered infant formula, peanut and nut pastes, as well as flours and meals have increasingly been associated with product recalls and foodborne outbreaks due to contamination by pathogens such as Salmonella spp. and enterohemorrhagic E. coli. In particular, recent foodborne outbreaks and product recalls related to Salmonella-contaminated spices have raised the level of public health concern for spices as agents of foodborne illnesses. Presently, most spices are grown outside the U.S., mainly in 8 countries: India, Indonesia, China, Brazil, Peru, Madagascar, Mexico and Vietnam. Many of these countries are under-developed and spices are harvested and stored with little heed to sanitation. The FDA has regulatory oversight of spices in the United States; however, the agency’s control is largely limited to enforcing regulatory compliance through sampling and testing only after imported foodstuffs have crossed the U.S. border. Unfortunately, statistical sampling plans are inefficient tools for ensuring total food safety. As a result, the development and use of decontamination treatments is key. This book provides an understanding of the microbial challenges to the safety of low aw foods, and a historic backdrop to the paradigm shift now highlighting low aw foods as vehicles for foodborne pathogens. Up-to-date facts and figures of foodborne illness outbreaks and product recalls are included. Special attention is given to the uncanny ability of Salmonella to persist under dry conditions in food processing plants and foods. A section is dedicated specifically to processing plant investigations, providing practical approaches to determining sources of persistent bacterial strains in the industrial food processing environment. The volume marks the first IARC evaluation of nucleoside analogs that act as antiviral agents. The evaluation responds in part to recent findings that zidovudine (AZT) an effective antiretroviral agent now being given to pregnant HIV-infected women to prevent maternal-to-fetal transmission of the virus is a transplacental carcinogen in mice. The opening monograph evaluates the carcinogenicity to humans of the antiretroviral nucleoside analogs zidovudine (AZT) zalcitabine (ddC) and didanosine (ddI) and the antiherpesvirus drug aciclovir. Of these aciclovir and didanosine could not be classified on the basis of available data. For zidovudine transplacental administration to mice resulted in an increased incidence and multiplicity of lung and liver tumours and in an increased incidence of female reproductive tract tumours in one study but not in another involving treatment at a lower dose. Despite observation of toxic effects in some studies of humans human carcinogenicity data were judged to provide inadequate evidence of carcinogenicity in humans. Zidovudine was classified as possibly carcinogenic to humans. Similar weaknesses in human carcinogenicity data for zalcitabine which consistently induces thymic lymphomas in mice resulted in its classification as possibly carcinogenic to humans. The second monograph evaluates four DNA topoisomerase II inhibitors: etoposide teniposide mitoxantrone and amsacrine. Of these etoposide - one of the most widely used and effective cytotoxic drugs in combination therapy - was classified as probably carcinogenic to humans and etoposide in combination with cisplatin and bleomycin was judged to be carcinogenic to humans. Teniposide was classified as probably carcinogenic to humans and mitoxantrone and amsacrine were classified as possibly carcinogenic to humans. Of the three pharmaceutical agents evaluated in the final monograph hydroxyurea which is widely used in cancer treatment and increasingly in combination with didanosine in HIV infection could not be classified. Phenolphthalein a widely used laxative now being withdrawn from the market in many countries because of toxicological concerns was classified as possibly carcinogenic. Vitamin K substances could not be classified on the basis of available evidence.

Book excerpt: Non-thermal (cold) plasmas at atmospheric pressure have recently found many breakthrough applications in biology, medicine, and food security. Plasmas can efficiently kill bacteria, yeasts, moulds, spores, biofilms and other hazardous microorganisms, including potential bio-terrorism agents. They can be employed for bio-decontamination and sterilization of surfaces, medical instruments, water, air, food, even of living tissues without causing their damage. Direct or indirect plasma interaction with living cells of microorganisms or even humans enables novel bio-medical applications, e.g. treatment of skin diseases and ulcers. Plasma-enhanced blood coagulation coupled with its antiseptic properties proved success in wound healing and opens new possibilities in surgery, emergency medicine and military applications. Plasma treatment allows cell manipulations, their removal and targeted transfer into the injured area, which can accelerate wound healing. Plasma induced apoptosis (programmed cell death) of tumor cells brings forth a great potential for cancer treatment. Besides, plasma enables painless treatment of dental caries, root canal disinfection, and other dentistry applications. This book is a selection of reviewed manuscripts issuing from the NATO Advanced Research Workshop Plasma for bio-decontamination, medicine and food security held in Jasná, Slovakia, on 15-18 March 2011. It provides a comprehensive overview of the current knowledge and research activities focused at the plasma applications in areas such as bio-decontamination, water chemistry, effects on cells; biofilm inactivation, UV sterilization, and medicine, especially tissue treatment and wound healing, as well as dentistry and food security.
