Book excerpt: Abstract: Self-assembled monolayers (SAMs) are organized molecular assemblies that are formed by spontaneous adsorption of a compound in solution to a surface (e.g. alkanethiols on gold). The design, preparation, and characterization of several self-assembled monolayers and multilayers on surfaces (gold, indium tin oxide and quartz) are described. The systems were chosen based on their ability to form ordered films and to perform a given device function. SAMs were fabricated with selected functional groups at the air-monolayer interface, capable of complexing metal ions (e.g. dicarboxypyridine, dicarboxybenzene, imidazole, 4-hydroxypyridine) with the purpose of using these SAMs to construct multilayered films. Deposition of a second layer consisting of metal ions (e.g. Cu(II), Co(II) and Fe(III)), occurs by non-covalent metal ligand binding interactions between the metal ion layer and the different organic ligands on the surface. Deposition of subsequent layers was achieved by the incorporation of the appropriate organic ligands and metal ions. These monolayers and multilayered films were characterized by contact angle measurements, ellipsometry, grazing angle FT-IR, cyclic voltammetry and impedance spectroscopy following deposition of each layer on the film. Electrochemical analysis of the multilayered films shows alternating insulating/conducting behavior (cyclic voltammetry) and alternating changes in films capacitance (impedance spectroscopy) depending on the outermost layer of the film. Films capped with an organic layer show low conductivity, while films capped with a metal layer show conducting behavior. The electrochemical behavior of the films is related to the degree of "leakiness" or electrolyte solution permeation through the film, which is high for films with metal layers as the top layer and decreases once the film is capped with an organic layer. The alternating conducting/insulating behavior of the films allows for fabrication of multilayered thin films of variable thickness and tunable conducting properties. Ordered films were fabricated with up to seven layers of dicarboxypyridine and Cu(II), and 4-hydroxypyridine and Fe(III) metal-ligand units. The construction of these films provides an example of molecular films that could function as molecular wires or junctions due to their controllable electrochemical properties. Photocurrent generating films were fabricated by incorporation of chromophore groups (e.g. pyrene, porphyrins) into the multilayered structures. These films generate cathodic or anodic current upon photoexcitation of the chromophores. The monolayers functionalized with different organic ligands were also used to study lanthanide complexation on the surfaces. Successful deposition of different lanthanide ions was achieved from DMSO solutions. Monolayers of a bicyclic structure, 4, 7, 13, 16-tetraoxa-1,10,21-triaza-bicycle[8.8.5] tricosane-19,23-dione, attached to a hexadecanethiol molecule were used to study the ability of metal ion detection on the surface using electrochemical (cyclic voltammetry and impedance spectroscopy) techniques. The SAMs show higher complexation affinity for Li+ than for Na+ or K+. Preliminary studies were also carried out to investigate the ability of different SAMs to cell adhesion interactions. Future experiments will help elucidate a systematic relation of cell adherence and the bulk and molecular-level properties of the functionalized surfaces. The different multilayered films described in this dissertation served as preliminary models for different molecular scale device applications. Current work is focused in the design and preparation of more efficient photocurrent generating films, highly selective sensors for different types of ions, surfaces for cell adhesion and microbial interactions, and the study of other potential applications such as the design of micro and nanofluidic devices.

Self-assembled monolayers are organic molecules that can be deposited on a variety of surfaces, such as those of metals, metal-oxides, and semiconductors. Formation of SAMs on any inorganic material provides a ubiquitous way to impart desirable chemical and physical properties of organic and biological molecules to the inorganic substrate. It has been demonstrated that single molecules and their self-assembled monolayers can significantly alter the physical and electronic properties of inorganic conductors; moreover, studies have shown that the performance of many electrical devices can be transformed by modifying inorganic electrodes with organic SAMs. This is especially important for the development of next generation of ultra-compact electronic devices, in which the ability to control the interfacial charge-transport with a single monolayer of organic molecules would be ideal. We have developed different organic electronic architectures as test beds for studying the effect of monolayer properties, such as structural and geometrical parameters, on their electronic properties. By using a typical organic electronic device as a sensitive test platform, slight changes in a monolayer property, such as length, have been detected by studying the current- voltage characteristics (JV) of organic diodes functionalized with self-assembled monolayers (SAMs) of varying alkyl chain-length. Next, we describe the application of SAMs based on n-octylphosphonic acid (C8PA) and 1H,1H,2H,2H-perfluorooctanephosphonic acid (PFOPA) as anode buffer layers in C60-based organic photovoltaic (OPV) devices. We used the OPV platform to compare stabilities of organic monolayers exposed to ambient conditions with SAMs positioned inside working OPV devices. We found that the stabilities are different, suggesting the degradation mechanisms are distinct. The degradation of the OPV efficiency with respect to air exposure was significantly reduced with the perfluorinated PFOPA compared to the aliphatic C8PA. We attributed the OPV degradation to moisture diffusion from the top aluminum electrode and we discuss that the lowering of the anode work function is the result of hydrolysis of the SAM buffer layer. Next, we demonstrated the dependence of molecular electronic properties on the functional group substitution and that the changes in these properties can be measured using the organic light-emitting (OLED) platform. Specifically, we compared bilayered organic monomolecular systems immobilized on an inorganic electrode as the charge-injecting components of the organic light emitting diodes (OLEDs). Our bilayered interfaces comprise ordered inert primary and functional reactive layers, and they differ in only one parameter: the molecular structure of the terminal functional group. We demonstrate that we can visualize the differences in the charge transfer dynamics of two bilayered systems via patterned electroluminescence. In addition, we describe a new protocol for the preparation of shape-controlled multicomponent particles comprising metallic (Au and Ti), magnetic (Ni), and oxide (SiO2, TiO2) layers. First, we discuss the application and attractiveness of the colloidal structures, Janus Particles (JPs), that possess two different surfaces, varying either in polarity, hydrophilicity, etc. Next, we present our method for specifically controlling the composition, shape, and size of the micro-JPs. We demonstrate how this protocol permits fabrication of non-symmetrical particles by orthogonally functionalizing their opposite sides using well-established organosilanes and thiol chemistries (based on SAMs). We propose that these colloids may be used as convenient materials for studying non-symmetrical self-assembly at the meso- and micro-scales, due to their unique geometries and surface chemistries"--Pages viii-x.

Book excerpt: The subsequent work describes advances in modifying the chemical properties of various substrates to tailor the surface properties for specific applications. This is achieved by making use of a molecular assembly known as self-assembled monolayers, or SAMs. SAMs are composed of tightly packed organic molecules that form a well-ordered structure on a substrate. Typically, the head group of the monomer is covalently anchored to the substrate, and monolayer order and self-assembly is achieved through van der Waals interactions between the long alkyl chains of the monomer's tail group. Monolayers containing head groups consisting of thiols, siloxanes, and phosphonates have been demonstrated on gold, glass, and metal oxides, respectively. We have expanded upon existing monolayer technology and designed monolayers with either new head group or new tail group functionalities. The resulting surfaces have been characterized by a variety of techniques including infrared spectroscopy, contact angle analysis, quartz crystal microbalance analysis, surface plasmon resonance imaging, and atomic force microscopy. We have also explored applications for these functionalized surfaces in areas ranging from microelectromechanical systems (MEMS) lubrication to platforms for studying neuronal development in vitro. In the area of MEMS lubrication, the development of new surface coatings is critical for combating wear and increasing the device lifetime. We reported a class of arsonic acid SAMs that form readily on oxide substrates including silicon oxide, borosilicate glass, and titanium oxide. The monolayers are easily prepared using a straightforward soaking technique, which is amenable to large-scale commercial applications. We have characterized monolayer formation on borosilicate glass and titanium oxide using infrared spectroscopy. Monolayers on borosilicate glass, native silicon oxide and titanium oxide were also evaluated with contact angle measurements, and as wear measurements using nanoscratching experiments. On titanium oxide and borosilicate glass, monolayers prepared from hexadecylarsonic acid provide significantly greater surface protection than surfaces reacted under similar conditions with hexadecylphosphonic acid, a common modifying agent for oxide substrates. To develop a platform for in vitro studies of neuronal development, we have utilized mixed-monolayers incorporating low densities of cell-adhesive peptides. The monomers feature a tetraethylene glycol moiety in the tail group to prevent the non-specific adsorption of proteins, and a low density of monomers were terminated with an azide moiety to specifically attach a laminin-derived peptide (IKVAV) terminated with an alkyne group via the copper-mediated azide-alkyne cycloaddition (CuAAC) reaction. To achieve this, a pentynoic acid molecule was appended to the N-terminus of the peptide during solid phase synthesis. Surfaces containing 0.01% and 0.1% azide-coupled peptide were determined to be resistant to the non-specific adsorption of proteins. Hippocampal neurons dissected from embryonic mice were cultured on these surfaces and the effects of the peptides on neurite outgrowth were observed. Similar neurite numbers per cell were observed on both substrates, but longer neurites were measured on the 0.1% azide-coupled peptide substrate. Unfortunately, further studies revealed that aldehyde fixation methods for immunohistochemistry did not successfully attach neuronal cells to the surface due to limited attachment points on the surface. Many developmental cell biology experiments require downstream immunohistochemical analysis. As such, to overcome this limitation and to simplify the surface preparation, a protein-resistant intermolecular zwitterionic monolayer, which supports cell fixation, was utilized. We have shown that the intermolecular zwitterionic monolayer has well-defined, non-receptor mediated cellular attachment provided by cell-surface sugar interactions. Exploiting these properties, we have developed a monolayer stripe assay, where the interactions between neurons (cell bodies and neurites) and extracellular matrix (ECM) proteins or guidance cues can be observed and quantified. This system goes beyond current technologies and is capable of evaluating neuronal response to the extracellular matrix protein, laminin, which has previously been considered a control molecule in neuronal stripe assays. Taken together, this work highlights advancements in the field of self-assembled monolayer chemistry with practical applications. In particular, we have focused on the functionalization of glass and oxides surfaces for applications in device lubrication. As well, we have developed two alkanethiol self-assembled monolayer approaches for generating surfaces that are both protein resistant and cell permissive, advancing the tools available for studying neuronal development in vitro.
