Book excerpt: Epidermal growth factor (EGF) and transforming growth factor a (TGFalpha) acting through EGF receptors (EGFR) regulate the development of normal mammary epithelial cells (MEC) and breast cancer. Primary culture studies examined the effects of EGF and TGFalpha in MEC. PD158780, a selective inhibitor of the tyrosine kinase domain of EGFR, helped demonstrate that EGFR signaling was required for the proliferation and functional differentiation of immature MEC from days 0-7 of culture, and survival of terminally differentiated MEC from days 17.5-21 of the study. EGFR levels were expressed at high levels in non-functional MEC and MEC undergoing apoptosis whereas functionally differentiated MEC expressed relatively low levels of EGFR. In EGF medium, cultured MEC expressed peak levels of erbB2 and erbB3 from day 7-14 of culture, whereas erbB4 levels were only detected within MEC cultured for 7 and 10.5 days. MEC isolated from mammary glands expressed high levels of erbB2 and erbB3 in virgin rats and rats during pregnancy and involution, and relatively low levels during lactation, whereas erbB4 appeared to be uniformly expressed throughout the developmental stages analyzed. These findings will help in the development of therapies to treat aggressive human breast cancers that often overexpress EGFR and/or other erbB receptors.

They occur during mammary cell proliferation in normal virgin, pregnant and lactating mouse mammary glands, and in early embryo cells. To test the roles of Cr-1, we infected normal mouse mammary cells with retroviruses over-expressing Cr-1, or an antisense version, in order to inhibit the production of this polypeptide. Cr-1 stimulated proliferation and was inhibitory to differentiation (lactation). Reduction of Cr-1 did not affect differentiation but decreased growth rate and increased apoptosis. Cr-1 overexpression transforms cells but appears not to be tumorigenic. We completely inactivated the Cr-1 genes in order to produce mice that expressed no Cr-1 in mammary glands as well as in all other tissues. To this end, we inactivated Cr-1 in embryo stem (ES) cells by homologous recombination and produced Cr-1 gene-targeted mice. Studies on these Cr-1 ( -/- ) cells showed that ES cell differentiation to cardiac myocytes was prevented. The "knockout" animals demonstrate that Cr-1 is essential for axis formation and embryonic development. Cr-1 absence causes aberrant proliferation of mesoderm cells, loss of migratory signals and absence of heart development.

Over decades of study, the EGF receptor structure, its ligand binding domains, the physical biochemistry underlying its intrinsic tyrosine kinase catalytic function and the modular interactions with SH2, PTB, and SH3 domain containing signaling adaptor p- teins required for signal transduction, have been extensively dissected. Not only is the EGF receptor the nexus of many streams of information, but it also forms one part of a calcul- ing device by forming dimers and oligomers with the other three receptors in its family in response to at least eleven ligands (some of which are expressed in multiple forms with overlapping or quite distinct functions). This phenomenon, while recruiting to the inner surface of the cell membrane and activating multiple second messenger proteins, also allows the possibility of cross talk between these systems, permitting a further layer of information to be exchanged. Less well described are the cross re gulation of the EGF receptor and other anti-apoptotic, mitogenic and metabolic signaling systems. The study of these systems has yielded new surprises. One hurdle in these efforts has been that signal transduction pathways have frequently been defined in the generic absence of their tissue-specific or cell-interaction specific context.

Beyond cell proliferation, activation of EGF receptor by its specific ligands controls important physiological processes, such as cell differentiation, apoptosis, cell migration, and cell shape. Activation of autocrine growth loops, consisting in solid human tumors of upregulated expression of EGFR together with increased production of ligands suggested its crucial role in autonomous tumor growth.

Determining the specific role of HGF/c-Met signaling in breast cancer has been compounded by the fact that c-Met communicates with a number other receptor pathways that lead to various pathological outcomes. In the present study we show that the epidermal growth factor receptor (EGFR) plays a direct role in HGF/c-Met mediated biological activities. We used the clinically relevant EGFR inhibitors gefitinib and erlotinib to determine the role of EGFR in mediating HGF signaling. Inhibition of EGFR through these small molecule inhibitors significantly reduces the ability of HGF to induce proliferation, cell motility and invasion as well as cell scattering in various mammary epithelial cells. Furthermore, our data indicate that gefitinib and erlotinib negatively regulate the ability of HGF to activate its cognate receptor c-Met and that this action is EGFR-dependent. Our results provide a novel mechanism of action for EGFR as a mediator of HGF signaling thereby linking EGFR to the oncogenic potential of c-Met in mammary carcinomas cells.

Its role in breast cancer, however, is unclear. We have studied the mechanism of induction of HB-EGF(by EGF(in the spontaneously immortalized mammary epithelial cell line MCF-10A and in MCF-10A cells that have been transformed with oncogenic Ha-ras. The levels of EGFR, erbB-2 and erbB-3 were very similar in the nontransformed and in the transformed MCF-10A cells. However, the levels of mitogen- activated protein kinase (MAPK) were elevated in the MCF-10A cells transformed with the Ha-ras oncogene. Neutralizing antibodies and inhibitors against the EGFR were able to inhibit the induction of HE-EUF mRNA levels in the nontransformed MCF-10A cells but not in the MCF- lOA cells transfected with oncogenic Ha-ras. However, an inhibitor of MAPK kinase (MEK) completely blocked the EOF induction of HB-EOF mRNA levels in the parental MCF-10A cells as well as in transformed MCF-10A cells, which suggests that EUF may be operating through other erbB receptors such as erbB-2/erbB-3 heterodimers and that MAPK is involved in the i, of induction in nontransformed and transformed human mammary.
