Book excerpt: Since the last International Bile Acid Meeting in San Diego in 1994, new advances have been made in the understanding of bile acid metabolism and transport. The cytotoxic as well as the therapeutic effects of specific bile acids have been further explored and the beneficial effects of ursodeoxycholic acid in various chronic cholestatic conditions have been substantiated. Much basic and clinical research has been conducted to unravel the underlying mechanisms of action. This volume, the proceedings of Falk Symposium No. 93 (XIV International Bile Acid Meeting), held in Freiburg-im-Breisgau, Germany, October 22-24, 1996, is dedicated to both basic and clinical aspects of bile acid research related to the role of bile acids in hepatobiliary diseases.

Available in PDF, EPUB and Kindle. Book excerpt: It has been recognized for almost 200 years that certain families seem to inherit cancer. It is only in the past decade, however, that molecular genetics and epidemiology have combined to define the role of inheritance in cancer more clearly, and to identify some of the genes involved. The causative genes can be tracked through cancer-prone families via genetic linkage and positional cloning. Several of the genes discovered have subsequently been proved to play critical roles in normal growth and development. There are also implications for the families themselves in terms of genetic testing with its attendant dilemmas, if it is not clear that useful action will result. The chapters in The Genetics of Cancer illustrate what has already been achieved and take a critical look at the future directions of this research and its potential clinical applications.

The median survival after the initial diagnosis of glioblastoma (GBM) is only 15 months, and less than 10% of patients survive three years post-diagnosis. Surgical treatment followed by adjuvant therapies such as radiotherapy and chemotherapy represents the classical strategy in glioma management. The revised WHO 2016 classification now distinguishes the oligodendrogliomas with 1p19q codeletion and IDH mutation from the astrocytomas with or without IDH mutations, thereby creating homogenous and pathologically distinct subgroups. While the status of gene expression and mutations define components of GBM subtypes, it was also found that response to therapies was different for each subtype, suggesting that personalized treatment based on genomic alterations could lead to a more favorable outcome for this disease.

Book excerpt: The discovery of microRNA (miRNA) involvement in cancer a decade ago, and the more recent findings of long non-coding RNAs in human diseases, challenged the long-standing view that RNAs without protein-coding potential are simply “junk” transcription within the human genome. These findings evidently changed the dogma that “DNA makes RNA makes protein” by showing that RNAs themselves can be essential regulators of cellular function and play key roles in cancer development. MiRNAs are evolutionarily conserved short single-stranded transcripts of 19–24 nucleotides in length. They do not code for proteins, but change the final output of protein-coding genes by regulating their transcriptional and/or translation process. Ultraconserved genes (UCGs) are non-coding RNAs with longer length (>200bp) that are transcribed from the ultraconserved genomic region. Both miRNAs and UCGs are located within cancer-associated genomic regions (CAGRs) and can act as tumor suppressors or oncogenes. In this chapter, we present principles and concepts that have been identified over the last decade with respect to our understanding of the function of non-coding RNAs, and summarize recent findings on the role of miRNAs and UCGs in cancer development. Finally, we will conclude by discussing the translational potential of this knowledge into clinical settings such as cancer diagnosis, prognosis and treatment.

This book was released on 2010 with total page 728 pages. Available in PDF, EPUB and Kindle. Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.
