Among these bioactive compounds, several A wide variety of plant-derived compounds, including polyphenols, are present in the human studies revealed that resveratrol, a trans-3,4,5-trihydroxystilbene, found in various natural products may be one of the main wine micro-components responsible for health benefits. Indeed, this polyphenol possesses a myriad of beneficial effects and can act at multiple levels, such as cellular signalling, enzymatic pathways and gene expression. Many laboratories have studied the molecular and cellular mechanisms of resveratrol in various models to get insight into its mechanisms of action in various disease processes.The increase of resveratrol interest is both reflected by the number of publications in databases, approximately 4,520 in Medline, and by the large number of patents copyrighted by biotechnology companies with new applications in cosmetic fields, central nervous system, food complement, preparations, to better understand the origins of resveratrol, its cellular and molecular mechanisms in plants and humans, this book on resveratrol has been divided into sections ranging from natural and industrial production to the latest scientific advances and clinical trials through the participation of leading scientists in each area covered in this book. Section 1 describes the sources and new engineering processes for custom and commercial bioreactors production of resveratrol. Section 2 is devoted to the common and new methods of resveratrol analysis. The following sections concern the health benefits of resveratrol. Section 3 shows the anti-oxidant properties of resveratrol in various physiopathological states. Section 4 focuses on the cardioprotective effects of resveratrol and the improvement of the endothelial function.Section 5 deals with metabolic disorders and the potential use of resveratrol in the prevention and treatment of obesity and osteoporosis. Section 6 is dedicated to health benefits of resveratrol against inflammation and associated diseases such as rheumatoid arthritis. Section 8 reviews the potential use of resveratrol in neuroprotection such as neurovascular system and neuroglial plasticity, and in the prevention and treatment of neurodegenerative diseases such Alzheimers and Parkinsons diseases. Besides the prevention of a number of diseases, other aspects such as dietary intake and bioavailability are important for the comprehension of resveratrol mechanisms of action. In this area, section 9 provides an overview of the metabolism of resveratrol, its pharmacokinetics and the recent advances in resveratrol delivery.The last part of this book, section 10, is dedicated to the clinical development of resveratrol and to the future development of resveratrol derivatives to provide new pharmacological tools for treatment of various diseases. Book excerpt: Resveratrol (3,5,4'-trihydroxystilbene) is a polyphenolic phytoalexin found in high quantities in various dietary sources, such as grapes, red wine, berries and peanuts. Various reports have demonstrated a significant role of resveratrol in the management of several old age diseases including cancer. The efficacy of resveratrol as an anti-cancer agent resides in its ability to interfere with cell proliferation and metastasis and enhancement of apoptosis. Resveratrol has been shown to act on several intracellular targets to exert these effects. However, the exact mechanism by which resveratrol mediates its beneficial cancer chemotherapeutic actions are not clear and is the focus of this study. Based on the reported data, we hypothesized that resveratrol mediates its anti-cancer action against metastatic prostate cancer by inhibiting the signaling pathway which involves miR-21 expression and function. To address this hypothesis, we show that resveratrol decreased cell viability, migration and invasiveness of androgen-receptor negative and highly aggressive human prostate cancer cells, PC-3M-MM2. These effects of resveratrol were associated with the inhibition of miR-21 , since over-expression of miR- 21 with pre-miR-21 oligonucleotides attenuated resveratrol's effect on these cells. Additionally, resveratrol increased the expression of tumor suppressors, PDCD4 and maspin, which are negatively regulated by miR-21 and knockdown of PDCD4 by short interfering (si) RNA reversed the resveratrol's effect on prostate cancer cells. PC-3M-MM2 cells also exhibits high levels of phospho-Akt (pAkt), which were reduced by both resveratrol and LY294002, a known PI3-kinase inhibitor. MiR-21 expression in these cells appears to be dependent on Akt, as LY294002 reduced the levels of miR-21 along with a concurrent increase in PDCD4 expression. These in vitro findings were further corroborated in a severe combined immunodeficient (SCID) mouse xenograft model of prostate cancer. Oral administration of resveratrol not only inhibits the tumor growth but also decreased the incidence and number of metastatic lung lesions. These tumor- and metastatic-suppressive effects of resveratrol were associated with reduced miR-21 and pAkt, and elevated PDCD4 levels. Future investigation into the molecular mechanisms revealed that resveratrol suppressed prostate cancer growth by decreasing the levels of insulin-like growth factor-1 (IGF-1) and its receptor (R). Previous studies had associated elevated levels of serum IGF-1 with high risk of prostate cancer. IGF-1, after binding to its receptors, acts as a potent mitogen which stimulates cancer cell growth and proliferation mainly by activating Akt signaling pathway. Interestingly, this effect of resveratrol on IGF-1/IGF-1R was independent of its effect on miR-21 . In summary, our data show that resveratrol exerts its anti-cancer effect on metastatic prostate cancer cells, at least in part, by targeting Akt/ miR-21 pathway. These data highlight a potential molecular mechanism for resveratrol's anti-cancer action for the treatment of metastatic prostate cancer and suggest that inhibition of the IGF-1/Akt/miR-21 pathway is a rationale approach for the treatment prostate cancer metastasis. Book excerpt: Current research suggests resveratrol, a phytoalexin found predominately in grapes, may function as a chemopreventive and/or chemotherapeutic agent for various cancers, including colorectal cancer. However, the underlying mechanism(s) involved in these activities remain elusive. Thus, the objective of the studies discussed here sought to investigate the effect of resveratrol treatment on gene modulation in human colorectal cancer cells in order to identify and characterize novel molecular targets that contribute to the observed anticancer activities of resveratrol. Here, we identify activating transcription factor 3 (ATF3) and early growth response-1 (Egr-1) as novel targets of resveratrol and provide data to elucidate the mechanism(s) of regulation and how each target contributes to the anticancer effect of resveratrol in colorectal cancer cells. We demonstrate the involvement of resveratrol in ATF3 transcriptional regulation, which is facilitated by Egr-1 and Krüppel-like factor 4 interactions, and show that ATF3 contributes, at least partially, to resveratrol-induced apoptosis (Chapter 3). Moreover, we suggest that increased Egr-1 transcriptional activity by resveratrol requires posttranslational acetylation of Egr-1 in a SIRT1-independent manner. This acetylation by resveratrol may contribute to Egr-1-mediated expression of the pro-apoptotic protein nonsteroidal anti-inflammatory drug (NSAID)-activated gene-1 (NAG-1) induced by the phytoalexin (Chapter 4). Taken together, the work presented here provide (1) novel mechanisms by which resveratrol induces ATF3 and Egr-1 expression and (2) represent additional explanations for the anti-tumorigenic/anti-carcinogenic effects of resveratrol in human colorectal cancer cells.
