Book excerpt: Viral vectors are superior tools for gene therapy and as a genetic vaccine platform because viruses have evolved to efficiently infect and transfer their genomes to cells. Several impressive successes in viral vector-based gene therapies have been reported in humans, including restoration of vision in patients with Leber’s congenital amaurosis by retinal gene transfer and cures for severe immune deficiencies by gene transfer to hematopoietic stem cells. However, the mammalian immune system has evolved in parallel to fend off invading pathogens such as viruses. Innate and antigen-specific adaptive immune responses against viral vectors and therapeutic transgene products pose serious hurdles for successful gene therapy. Pre-existing immunity in humans, resulting from prior exposure to the parent virus that forms the basis for the gene transfer vehicle may be derived from, often prevents efficient gene transfer. This problem also reduces our ability to use certain vectors for genetic vaccination or in anti-cancer therapy. For these reasons, the gene transfer community has been extensively studying the mechanisms of immune responses against viral vectors and has started to develop strategies and protocols to block or circumvent such responses. Choice, design and engineering of a vector as well as the route of administration/target tissue can be optimized/ altered to minimize immune responses or evade pre-existing immunity. Immune suppression and modulation strategies are being developed in order to minimize inflammation, prevent antibody or T cell responses against vectors, and to promote tolerance to therapeutic gene products. Combinations of these approaches will likely facilitate clinical applications of gene therapy for many target diseases and also aid in vaccine development.

Available in PDF, EPUB and Kindle. Book excerpt: It has often been said that generals prepare for the next war by re-fighting the last. The Deepwater Horizon (DWH) oil spill was unlike any previous – an underwater well blowout 1,500 meters deep. Much has been learned in the wake of DWH and these lessons should in turn be applied to both similar oil spill scenarios and those arising from “frontier” explorations by the marine oil industry. The next deep oil well blowout may be at 3,000 meters or even deeper. This volume summarizes regional (Gulf of Mexico) and global megatrends in marine oil exploration and production. Research in a number of key areas including the behavior of oil and gas under extreme pressure, impacts on biological resources of the deep sea, and the fate of oil and gas released in spills is synthesized. A number of deep oil spills are simulated with detailed computer models, and the likely effects of the spills and potential mitigation measures used to combat them are compared. Recommended changes in policies governing marine oil exploration and development are proposed, as well as additional research to close critical and emerging knowledge gaps. This volume synthesizes state-of-the-art research in deep oil spill behavior and response. It is thus relevant for government and industry oil spill responders, policy formulators and implementers, and academics and students desiring an in-depth and balanced overview of key issues and uncertainties surrounding the quest for deep oil and potential impacts on the environment.

Available in PDF, EPUB and Kindle. Book excerpt: Stimulus-response compatibility refers to the finding that certain mappings of stimuli to responses produce faster and more accurate responding than do others. The present volume surveys compatibility research which falls into four broad categories: (a) mental representation and coding (b) neurophysiological mechanisms (c) motor performance (d) human factors applications. The major findings and models within each of the categories are summarized, and an integrated perspective is provided. The research indicates that compatibility effects reflect basic cognitive processes that bear on a range of issues in cognitive science and that have applied implications for human factors specialists.

Available in PDF, EPUB and Kindle. Book excerpt: The type 2 immune response that develops during infectious disease has undergone major paradigm shifts in the last several years as new cell types and pathways have been identified. It is now clear that the type 2 immune response, characterized by elevations in specific cytokines, including IL-4, IL-5 and IL-13, is associated with helminth infections in both humans and mice. This response is complex and includes effector functions that mediate resistance, contributing to expulsion and in some cases destruction, of the parasite. But just as importantly, the type 2 immune response can also mediate tolerance mechanisms, which can mitigate tissue injury as these large multicellular parasites transit through vital organs. The tolerance mechanisms include both tissue repair and immune regulatory effects. These latter aspects of the helminth-induced type 2 immune response are increasingly recognized as a potential resource that can be mined for the development of novel immunotherapies that may enhance wound healing, control of autoimmune and inflammatory diseases and regulation of metabolic homeostasis. In this book, leading researchers in this exciting and dynamic field discuss the latest findings and emerging concepts, providing an intellectual framework that can be used as a basis for new discoveries and potentially new treatments for diseases associated with inflammation.

Book excerpt: The two major subsets of CD4+ helper T cells, designated Th1 and Th2, have quite different patterns of cytokine production and, as a consequence, have very different roles in immune responses. The articles in this volume review both basic and clinical studies of T cell heterogeneity, including: the mechanisms by which Th1 and Th2 cells develop and maintain their differences in cytokine production; the different roles of Th1 and Th2 cells in allergy, autoimmunity and infectious diseases; the prospects and strategies for therapeutic manipulation of Th1 and Th2 cells; and the control of Th1 and Th2 responses by regulatory T cell subsets. The volume should give the reader a view of the development and function of Th1 and Th2 cells and the attempts to treat immunological diseases with therapies directed towards altering the Th1/Th2 balance.

Available in PDF, EPUB and Kindle. Book excerpt: This book analyzes compliments and compliment responses in naturally occurring talk-in-interaction in German. Using Conversation Analytic methodology, it views complimenting and responding to compliments as social actions which are co-produced and negotiated among interactants. This study is the first to analyze the entire complimenting sequence within the larger interactional context, thereby demonstrating the interconnectedness of sequence organization, turn-design, and (varying) function(s) of a turn. In this regard, the present study makes a novel contribution to the study of talk-in-interaction beyond German. The book adds to existing work on interaction and grammar by closely analyzing the functions of linguistic resources used to design compliment turns and compliment responses. Here, the study extends previous Conversation Analytic work on person reference by including an analysis of inanimate object reference. Lastly, the book discusses the use and function of various particles and demonstrates how speaker alignments and misalignments are accomplished through various grammatical forms.
