Available in PDF, EPUB and Kindle. Book excerpt: RHIZOBIUM EST CAPABLE D'INDUIRE SUR LES RACINES DE LEGUMINEUSES DE VERITABLES ORGANES VEGETAUX AU SEIN DESQUELS LES BACTERIES REDUISENT L'AZOTE ATMOSPHERIQUE EN ION AMMONIUM GRACE AU COMPLEXE ENZYMATIQUE NITROGENASE. LORS DE CETTE SYMBIOSE, RHIZOBIUM ENVAHIT LE CYTOPLASME DES CELLULES HOTES ET SE DIFFERENCIE EN BACTEROIDE FIXATEUR D'AZOTE. EN ECHANGE DE SUBSTRATS CARBONES, CET ORGANELLE PRODUIT L'AMMONIAC, NORMALEMENT ASSIMILE PAR LES GLUTAMINE-SYNTHETASES NODULAIRES. L'HARMONIE DE CE SYSTEME REPOSE EN PARTICULIER SUR LA REGULATION DES FONCTIONS ANABOLIQUES DU BACTEROIDE QUI, LORS DE SA DIFFERENCIATION DE BACTERIE EN BACTEROIDE, PASSE D'UN ETAT D'ASSIMILATION DE L'AZOTE VERS UN ETAT D'EXPORT DE L'AZOTE. LE CONTROLE DU METABOLISME AZOTE CHEZ LES BACTERIES MET EN JEU UN REGULATEUR CENTRAL CODE PAR LE GENE GLNB, LA PROTEINE P#I#I. SELON LE MODELE QUI A ETE DEVELOPPE CHEZ LES ENTEROBACTERIES, LA PROTEINE P#I#I EST MODIFIEE PAR URIDYLYLATION EN FONCTION DU STATUT AZOTE DE LA CELLULE. CETTE PROTEINE CONTROLE L'ACTIVITE DE LA GLUTAMINE SYNTHETASE (GS) AU NIVEAU GENETIQUE ET METABOLIQUE. AU NIVEAU GENETIQUE, LA PROTEINE P#I#I REGULE L'EXPRESSION DU GENE DE LA GS, GLNA, PAR L'INTERMEDIAIRE DE L'ACTIVATEUR TRANSCRIPTIONNEL NTRC. AU NIVEAU METABOLIQUE, LA PROTEINE P#I#I REGULE L'ACTIVITE DE LA GS EN MODULANT SON ADENYLYLATION. RHIZOBIUM MELILOTI POSSEDE TROIS GLUTAMINE-SYNTHETASES, GSI, GSII ET GSIII. LA PROTEINE GSI HOMOLOGUE DE LA GS DES ENTEROBACTERIES EST LA SEULE GS DE RHIZOBIUM EXPRIMEE EN SYMBIOSE. AU COURS DE CETTE THESE, NOUS AVONS ETUDIE LA REGULATION DE L'ACTIVITE DE LA GSI ET LE ROLE DE LA PROTEINE P#I39I DANS LA SYMBIOSE ENTRE RHIZOBIUM MELILOTI ET LA LUZERNE. EN CULTURE PURE, LES MUTANTS GLNB SONT SEVEREMENT ALTERES AU NIVEAU DU METABOLISME AZOTE: DEREGULATION DE L'ADENYLYLATION DE LA GSI ET DE L'EXPRESSION DU GENE GLNII CODANT POUR LA GSII. DE PLUS, LES MUTANTS GLNB SONT FORTEMENT AFFECTES DANS LEUR PHENOTYPE SYMBIOTIQUE. ILS ENTRAINENT UN RETARD DE NODULATION ASSOCIE A UN DEFAUT D'INFECTION. BIEN QUE LES NODULES FORMES CONTIENNENT UN NIVEAU ELEVE DE NITROGENASE, LES PLANTES PRESENTENT UNE CARENCE AZOTEE MANIFESTE. CETTE CARENCE S'ACCOMPAGNE D'UNE FORTE ACCUMULATION D'AMIDON DANS LA ZONE DE FIXATION DU NODULE, SYMPTOME D'UNE PERTURBATION DE L'EQUILIBRE CARBONE/AZOTE DANS LES CELLULES VEGETALES. UNE CIBLE CONNUE DE LA PROTEINE P#I#I CHEZ LES ENTEROBACTERIES EST L'ADENYLYLATION DE LA GS. NOUS AVONS TESTE LE ROLE DE L'ADENYLYLATION DE LA GSI DE RHIZOBIUM MELILOTI EN SYMBIOSE, PAR LA CONSTRUCTION D'UNE SOUCHE DONT LA GSI N'EST PAS ADENYLYLABLE. NOUS PENSIONS QU'UN TEL MUTANT PERTURBERAIT LE FLUX D'AZOTE DU BACTEROIDE VERS LA PLANTE. CONTRAIREMENT A CETTE ATTENTE, CE MUTANT PRESENTE UN PHENOTYPE SYMBIOTIQUE NOD#+ FIX#+ DE TYPE SAUVAGE. CE PHENOTYPE S'EXPLIQUE PAR UNE FAIBLE ACTIVITE GSI DANS LES BACTEROIDES. L'EFFET DE LA PROTEINE P#I#I SUR LA SYMBIOSE EST DONC DU A D'AUTRES EFFETS. UNE HYPOTHESE SERAIT QUE LA PROTEINE P#I#I REGULE L'EXPRESSION OU L'ACTIVITE D'UN TRANSPORTEUR D'AMMONIAC BACTEROIDIEN DONT LA PRESENCE SERAIT INDISPENSABLE A LA NUTRITION AZOTEE DE LA PLANTE-HOTE

Available in PDF, EPUB and Kindle. Book excerpt: During the past three decades there has been a large amount of research on biological nitrogen fixation, in part stimulated by increasing world prices of nitrogen-containing fertilizers and environmental concerns. In the last several years, research on plant--microbe interactions, and symbiotic and asymbiotic nitrogen fixation has become truly interdisciplinary in nature, stimulated to some degree by the use of modern genetic techniques. These methodologies have allowed us to make detailed analyses of plant and bacterial genes involved in symbiotic processes and to follow the growth and persistence of the root-nodule bacteria and free-living nitrogen-fixing bacteria in soils. Through the efforts of a large number of researchers we now have a better understanding of the ecology of rhizobia, environmental parameters affecting the infection and nodulation process, the nature of specificity, the biochemistry of host plants and microsymbionts, and chemical signalling between symbiotic partners. This volume gives a summary of current research efforts and knowledge in the field of biological nitrogen fixation. Since the research field is diverse in nature, this book presents a collection of papers in the major research area of physiology and metabolism, genetics, evolution, taxonomy, ecology, and international programs.

Book excerpt: Rhizobium species involved in root nodule formation on legume plants are one of the best known groups of micro organisms. The Rhizobium legume symbiosis continues to be of strategic importance particularly in the context of food production. As the world population grows, it is also neces sary to have new developments taking place in crop improve ment. The development and application of new technologies in biological sciences over the past number of years have made the entire area of plant-microbial interaction an exciting and challenging research area to be involved in. In view of the importance of symbiotic nitrogen fixation, it is not surpris ing that it still represents one of the priority areas for commercial development in agricultural biotechnology. Since this symbiosis involves an association between procaryotic and eucaryotic partners, it requires of necessity a co-ordinated and interdisciplinary approach. It was in this spirit that this international conference was organised. The scientific programme was designed to focus on physio logical limitations affecting symbiotic nitrogen fixation and the potential for overcoming such limitations using genetic technologies. Participants were drawn from contractants of the EEC DGVI "Energy in Agriculture" nitrogen fixation prog ramme. The scientific programme was also supplemented with invited scientists from Europe and North America to provide appropriate expertise on the various conference topics.

Book excerpt: The 14th International Nitrogen Fixation Congress was held in Beijing, China from October 27th through November 1st, 2004. This volume constitutes the proceedings of the Congress and represents a compilation of the presentations by scientists from more than 30 countries around the World who came to Beijing to discuss the progress made since the last Congress and to exchange ideas and information. This year marked the 30th anniversary of the first Congress held in Pullman, Washington, USA, in 1974. Since then, this series of Congresses has met five times in North America (three in the United States and once each in Canada and Mexico), once in South America (Brazil), four times in Western Europe (once each in Spain, The Netherlands, Germany and France), once in Eastern Europe (Russia), and once in Australia; and now for the first time in Asia. China was a most appropriate choice because China is a big country with the largest population in the World, about 1. 3 billion people, which is about 22% of the World’s population. It is traditionally an agricultural country, even though China has only 7% of the available farming land. This situation explains why agriculture and its productivity are major issues for the Chinese people, its government and the scientists in the field.

Book excerpt: For 31 years, the North American Symbiotic Nitrogen Fixation Conference (for merly Rhizobium Conference) has been a forum for scientists and graduate students to discuss their research advances, extending from basic aspects to agricultural appli cations, and dealing with topics ranging from bacterial genetics and metabolism to plant genetics and physiology. Nitrogen fixation, being a major life-supporting process on this planet, has attracted the interest of researchers for more than one century. Nitrogen fixation is responsible for the conversion of "inert" dinitrogen (N ) gas from the atmos z phere into usable ammonia, replacing the fixed nitrogen constantly being lost to the atmosphere by the denitrification process. Worldwide agricultural productivity is deter mined by the availability of fixed nitrogen in all its forms, which the continually increas ing human population depends upon for survival. An international momentum is developing in the use of biofertilizers to improve legume and non-legume crop yields and in the study of genomics in this area. New studies are being undertaken in several laboratories to study novel nitrogen-fixing systems, including non-legume crops. The North American Conferences on Symbiotic Nitrogen Fixation have periodically chronicled the advances in our knowledge of this area. For the first time, this conference was held in Mexico. Thus, all three North Amer ican countries have been hosts for this event. This year conference brought nearly 200 scientists from 18 different countries presenting lectures and over 80 posters.

Several types of nitrogen fixing bacteria are recognized: the free-living bacteria in soil or water; symbiotic bacteria making root nodules in legumes or non-legumes; associative nitrogen fixing bacteria that resides outside the plant roots and provides fixed nitrogen to the plants; endophytic nitrogen fixing bacteria living in the roots, stems and leaves of plants. In this book there are 11 chapters related to biological nitrogen fixation, regulation of legume-rhizobium symbiosis, and agriculture and ecology of biological nitrogen fixation, including new models for autoregulation of nodulation in legumes, endophytic nitrogen fixation in sugarcane or forest trees, etc. Hopefully, this book will contribute to biological, ecological, and agricultural sciences.
