A mammary cell-free system proved impractical due an inhibitor of translational initiation and a mammary- specific RNAse. However, using RNAse inhibitors, intact polyribosomes were prepared from mammary tissue. Both ribosome concentration and the proportion of palysomal ribosomes increased from late pregnancy to peak lactation. In contrast, the concentration of ribosomes and the proportion in polysomes both decreased when serum prolactin was experimentally depressed during lactation. Therefore, control of translation in vivo involves changes in both ribosome concentration and recruitment into polysomes. In mammary expiants, differences in protein synthesis per ribosome were measured after culture with insulin and cortisol, with or without prolactin. Translational responses to prolactin, were variable between different rats and different times of year. Responses to various hormone combinations were greater when expressed per unit DNA than per RNA suggesting that they involved both increased ribosome concentration and more efficient translation. Polysome analysis confirmed that the concentration of all ribosome species in expiants cultured with hormones exceeded that in expiants cultured without hormones but suggested no difference in the proportion of polysomal ribosomes. Therefore, alterations in translational efficiency in vitro were due to concomitant changes in translational initiation, elongation and termination. Insulin-like growth factor-1 and epidermal growth factor (IGF-1 and EGF) may modulate mammary proliferation and differentiation. In the presence of cortisol and prolactin, the response of translational efficiency to IGF-1 was, like that to insulin, dose dependent over a wide concentration range with a probable minimum effective concentration of lOng/mL. This suggested that each hormone acted through its own receptor. In contrast, EGF stimulated translational efficiency independently of accompanying hormones or concentration between 1 and 20ng/mL Results indicate that both growth factors affect translational efficiency in expiants.

Book excerpt: Study was conducted to find out the effect of boiler liver extract (BLEX) from rats at different physiological states like early mid late pregnancy and at o,5th,10th and 20th days of lactation to investigate the characteristic of blese in the tissues in relation to its stimulation of amino acids incarportation by the mamary homogenate and also other propesties of the BLEX The invitro protein synthesis by the rat mammary gland was stimulated by the boild liver extract from the lactating rat. The stimulation was 0.9 fold and 0.3 fold in lactating and non lactating mammary homogenates respectively. Such stimulatary activity could not be noticed in blex from non lactating rat s. Dialysed BLEX from lactating rat gave 1.0 fold and 0.4 fold increase in incarporation of amino acids by lactating and non lactation mammary homogenate from non lactating liver the stimulation was less pronounced. The 0-25% ammonium sulphate fraction (Fri) of the BLEX from lactating rat liver exhibited more pronounced stimulation in incarporation of amino acids namely by 4 folds in lactating and 2. folds in non lactating mammary homogenates. The BLEX, dialysed BLEX and other fractions from rats at different stages of pregnancy and lactation revealed that the peak activity resides in the liver tissues of rats at 0 day of lactation. The stimulatory progress in pregnancy and declined with involuation.

Available in PDF, EPUB and Kindle. Book excerpt: Lactation: A Comprehensive Treatise, Volume II, Biosynthesis and Secretion of Milk/Diseases, is part of a three-volume treatise containing a total of 28 chapters. The present volume is devoted to the functioning of the mammary gland in the synthesis and secretion of milk. Emphasis has been placed on studies which have described on a biochemical and molecular basis the operation of the pathways and structures involved in the biosynthesis of milk in the functional secretory cells of the mammary gland and the diseases of lactation that can affect them. The volume is organized into two parts. Part I on the biosynthesis of milk covers topics such as the general metabolism associated with the synthesis of milk; biosynthesis of milk fat and milk proteins; biochemistry of lactose and related carbohydrates; and cell structure relating to the formation and secretion of milk. Part II on diseases of the mammary gland and lactation includes studies on the clinical endocrinology of prolactin, microbial diseases of the mammary gland, ketosis, and mouse mammary tumors.

Available in PDF, EPUB and Kindle. Book excerpt: Lactation: A Comprehensive Treatise, Volume IV, The Mammary Gland/Human Lactation/Milk Synthesis, forms the fourth in a series. When the first three volumes were published in 1974, publication of future volumes was not contemplated. However, the gratifying acceptance by the scientific community and the continuation of rapid advances in lactation, have provided the impetus for the continuation of the series. The present volume is concerned with general aspects of the mammary gland, human lactation, and mechanisms of milk synthesis. The volume is divided into three main parts. Part I covers some aspects of the mammary gland and lactation not discussed in the first three volumes and expands on others. Topics discussed include the development of the mammary apparatus and neuroendocrine control of lactation. Part II is devoted to more specific consideration of human lactation as a whole. It includes studies on breast feeding and the breast cancer process. Part III on milk synthesis deals with the mechanisms of milk synthesis; enzymology and control of lactose biosynthesis; molecular aspects of milk protein biosynthesis; and ion and water transport in the mammary gland.
