Book excerpt: In a fast-moving field it is unlikely that articles written more than a year ago would be completely up to date. The purpose of this book is to bring to the nonspecialist an overall view as well as an update on the state of the art as it existed in the beginning of 1982, and to the specialist the opportunity to have a single source of information for how the other organisms do it, and also to enable him to find out the status of the various aspects of RNA processing with which he might not be to familiar. even if only some of these goals are achieved, all those who labored so diligently to bring about the publication of this book would be more than gratified.

Available in PDF, EPUB and Kindle. Book excerpt: The last few years have seen the rapid development of new methodology in the field of molecular biology. New techniques have been regularly introduced and the sensitivity of older techniques greatly improved upon. Developments in the field of genetic engineering in particular have contributed a wide range of new techniques. The purpose of this book therefore is to introduce the reader to a selection of the more advanced analytical and preparative techniques which the editors consider to be frequently used by research workers in the field of molecular biology. In choosing techniques for this book we have obviously had to be selective, and for the sake of brevity a knowledge of certain basic biochemical techniques and terminology has been assumed. However, since many areas of molecular biology are developing at a formidable rate and constantly generating new terminology, a glossary of terms has been included. The techniques chosen for this book are essentially based on those used in a series of workshops on 'techniques in molecular biology' that have been held at The Hatfield Polytechnic in recent years. In choosing these chapters we have taken into account many useful suggestions and observations made by participants at these workshops. Each chapter aims to describe both the theory and relevant practical details for a given technique, and to identify both the potential and limitations of the technique. Each chapter is written by authors who regularly use the technique in their own laboratories.

Available in PDF, EPUB and Kindle. Book excerpt: Methods in Virology, Volume VIII focuses on the methods used in virology, including microscopy, hybridization, viruses, and fingerprint analysis. The selection first offers information on the hybridization of viral nucleic acids; applications of oligonucleotide fingerprinting to the identification of viruses; and immunosorbent electron microscopy in plant virus studies. Discussions focus on the detection of double-stranded RNA, principles and mechanisms of fingerprint analysis, preparation of labeled nucleic acid probes, and basic methods of nucleic acid hybridization. The text then elaborates on quantitative transmission electron microscopy for the determination of mass-molecular weight of viruses and use of thin sectioning for visualization and identification of plant viruses. Topics include technical procedures for processing plant tissues, cytological modifications of diagnostic value, procedure and treatment of data to obtain the average mass of virus particles, and applications in virology. The book takes a look at the detection of genome-linked proteins of plant and animal viruses; methods for assay, purification, and characterization of prions; and the use of mosquitoes to detect and propagate viruses. The selection is a valuable source of information for researchers interested in the methods employed in virology.

Available in PDF, EPUB and Kindle. Book excerpt: This volume contains the papers presented at the international symposium on "Molecular Mechanisms in Protein Synthesis" held on September 26-27, 1983 at the Beyaz Ko§k in Emirgan, Bosphorus, Istanbul. The symposium aimed to create a medium for information exchange and discussions regarding the current developments in the area of protein syn thesis. To ensure an informal yet scientifically stimulating and productive atmosphere providing opportunity for relaxed and speculative discussions, the number of presentations was limited to twenty and that of attendants to about sixty. The emphasis in the symposium was laid on structure-function relations in the prokaryotic protein synthesizing systems and on the control mechanisms of eukaryotic protein synthesis, in particular, during chain initia tion. Other issues like evolutionary aspects of protein synthesis, translational components genes and proofreading were covered as well. The manuscripts represent the extended accounts of the oral presenta tions, and it has been aimed with the concluding remarks at the end of the volume to give a summarizing view of the presentations and the discussions.

Available in PDF, EPUB and Kindle. Book excerpt: Biochemical Actions of Hormones, Volume XII is a compendium of papers discussing aspects of molecular biology with regard to hormone action and to certain hormone receptors. One paper discusses the different approaches and strategies that are used in determining the amino acid sequence. The paper also describes the structural organization of polyprotein precursors such as the peptide chemistry approach or the nucleic acid approach. Another paper investigates the regulation of prolactin biosynthesis by peptide hormones as a model for peptidergic regulation of gene expression. The paper defines in detail which components of the biosynthetic machinery are being regulated by the polypeptide hormones. One paper also analyzes the molecular biology of the metallothioneins, focusing on the glucocorticoid regulation of metallothioneins genes. Experiments show that when the mouse MT-I gene is activated in lymphoid cells, or transferred to other cells, it still retains information needed in metal-regulated expression, though glucocorticoids can no longer regulate the same gene. Other papers discuss the purification and properties of the nerve growth factor receptor, and the thyrotropin receptor. Micro-biologists, biochemists, molecular biologists, cellular biologists, endocrinologists, and scientists involved in cell research will find this collection valuable.

Available in PDF, EPUB and Kindle. Book excerpt: The Discovery of Ribonuclease P and Enzymatic Activity of Its RNA Subunit Sydney Brenner and Francis H. C. Crick had a specific project in mind when they offered Sidney Altman a position in their group in 1969 to conduct postdoctoral research at the Medical Research Council Laboratory of Molecular Biology (LMB) in Cambridge, England. At the time, an intense international competition was on- ing in as many as a dozen labs to determine the three-dimensional structure of tRNA. At the LMB, Aaron Klug was attacking the structure by crystallographic analysis with Brian F. C. Clark providing large amounts of purified phenylalanine tRNA. (Eventually, Aaron announced his empirically determined 3-D structure of yeast phenylalanine tRNA, a structure that is generally common to tRNAs, due in part to several conserved, novel three-way nucleotide interactions. ) Concurrently, Michael Levitt, a Ph. D. student of Francis, was visually scrutinizing the cloverleaf secondary structure of the 14 tRNA sequences known at the time. Levitt was searching for nucleotide covariation in different parts of the molecules that were conserved in the 14 sequences known at the time. He identified a possible covariation of an apparent Watson-Crick pairing type between the residues at position 15 from the 5’ end of the tRNA and residue 48. This association implied these parts of the tRNA, namely the D loop containing residue 15 and the 5’ end of the T stem-adjoining residue 48, folded on one another in a tertiary structure shared by different tRNAs.

Book excerpt: The advent of human in vitro fertilization and developments in molecular science have made it possible to diagnose genetic disease in fetuses at a very early stage of development. This book presents the most recent advances and developments in this new area of applied research. Experts in the field describe and discuss the latest technology, from micromanipulation of single cells to sophisticated methods of DNA analysis. The book considers the implications of this new technology for fetal screening and the prevention of genetic disease, as well as for the control and manipulation of the normal sex ratio. It draws together current knowledge and discusses the new clinical opportunities for disease prevention. The book begins with an overview of the genetics of some common inherited diseases and the historical development of prenatal screening. It then draws together the most recent advances in preimplantation diagnostic techniques, including tissue assays for specific genetic disorders, and methods for typing and separating spermatozoa. The practical aspects of the new methodologies are considered, along with the potential for developing safer, more reliable and less invasive methods of diagnosis. The ethical and legal implications of this diagnostic approach are also discussed.

Available in PDF, EPUB and Kindle. Book excerpt: RNAs form complexes with proteins and other RNAs. The RNA‐infrastructure represents the spatiotemporal interaction of these proteins and RNAs in a cell‐wide network. RNA Infrastructure and Networks brings together these ideas to illustrate the scope of RNA‐based biology, and how connecting RNA mechanisms is a powerful tool to investigate regulatory pathways. This book is but a taste of the wide range of RNA‐based mechanisms that connect in the RNA infrastructure.
