Book excerpt: Phytoplankton and zooplankton are important components of marine ecosystems, and play a major role in the biological pump, affecting carbon transport in the global oceans. Their dynamic heterogeneous spatial and temporal distributions require special tools for observing them at the ecological scales relevant to the individual organisms. In this work, I used optic and acoustic methods to study plankton organisms at spatial scales of meters and temporal scales ranging from minutes to months. Using two in situ microscopes I described the fine-scale vertical distribution of phytoplankton and several zooplankton taxa in a coastal location in the Southern California Bight. Highly resolved spatial observations revealed cryptic maxima of fluorescent particles not observed with traditional fluorometers. Furthermore, this high sampling resolution revealed that water density, and not depth, regulated the vertical position, and interactions between observed phytoplankton and zooplankton distributions. Underwater acoustic echosounders can be powerful tools to observe in situ plankton distributions. Interpreting the acoustic echoes, however, requires highly calibrated instruments and ground-truthing experiments to identify the source of acoustic signals. This work presents the description of a novel combination of a broadband, high-frequency (1.5-2.5 MHz) echosounder and a stereoscopic camera --combined, these systems can localize the echo produced by an individual target while simultaneously providing visual identification of the target. This work has provided one of the first comparisons of in situ measured broadband target strength (BTS) and the expected signal using a physical model. The results of this experiment revealed unexpected, important differences between measured and modeled BTS. This system was also used to make in situ observations of individual fragile gelatinous organisms, marine snow particles and phytoplankton, providing evidence of their significant acoustic reflectivity. Finally, using a moored in situ microscope (Scripps Plankton Camera) similar in design to the O-Cam helped identifying a parasite-host interaction over a period of a few months. This is the first reported observation of Paradinium poucheti parasitizing Oithona similis in the North Pacific Ocean. The short time-series revealed that the prevalence of this parasite is higher than previously observed in other ocean basins.

We have attempted to provide a mix of contributions, composed of reviews of the state-of-the-art, reports on current research activi ties, summaries of the design and testing of a new generation of innovative instruments for biological and chemical sampling and sorting, and some imaginative ideas for future experiments on stimulated mixing in continental shelf seas. We encouraged the contributors to present critical and thought provoking assessments of current wisdom specifying the sorts of techniques and observational strategies needed to validate the various hypotheses linking physical structure, mixing and circulation to plankton biomass and production. We hope this volume will appeal to incoming research students and established scholars alike. We certainly have enjoyed working with all the authors in compiling this book. Book excerpt: The application of mesocosms, defined in this report as artificial 3 3 experimental enclosures ranging in size from 1 m to 10m , to address various problems in the marine sciences has been a relatively recent development. The application of the technology was dictated by the realization that many important ocean processes and interactions cannot be fully understood from observations in the natural environment or in smaller enclosures. Such studies involve, for example, determining the interactions between, and energy transfer from, one trophic level to another, the biogeochemical cycling of elements and compounds, etc. These and similar interactions and rate processes cannot normally be established in situations (nature) where the detection and quantification of rate processes are confused by advection and/or the inability to study the same populations over time. In the case of microcosms, mixed populations of primary producers, consumers, and carnivores cannot be maintained, in balance, for a sufficient length of time to determine normal interactions between the various components of these trophic levels. This report, prepared by SCOR Working Group 85, critically examines past applications of mesocosms to ocean research, though there is no attempt to comprehensively review all literature relevant to the subject. Further, the report outlines some important advances emanating from their use and provides recommendations for future applications. It constitutes the first of two reports from the Working Group (see Introduction).

Techniques developed by molecular biologists have now been adopted by a wide range of scientific disciplines. The aim of this ASI was to assess progress in this new field of research, to compare and describe techniques and experimental approaches, and to foster communication between disciplines. The ASI offered an excellent opportunity to bring together aquatic ecologists with molecular biologists and to encourage efficient technology transfer. The meeting of information on the status provided a forum for detailed and broad exchange and trends of aquatic molecular ecology and to assess how emerging molecular techniques might solve some important problems in ecology which have prove intractable because of lack of appropriate methodologies.
