Principles of Endocrine Pharmacology concerns itself with the ther apeutic use of hormones, and hormone like substances, or drugs that can act either by suppressing or enhancing the metabolism of certain glands of internal secretion. Other drugs used for nonendocrine ther apies can likewise affect the endocrine system. Endocrine pharmacology emerged in the early 1900s with the use of crude pituitary extracts. By the mid-1900s several investigators had isolated and begun to synthesize hormones or hormonelike substances. Recognizing the limited supply of hormones that could be obtained both from animal sources and human autopsy material, the search for so called hormone substitutes also began early in the 1900s. Recently, re combinant DNA technologies have been used to provide alternative therapeutic sources of human insulin and human growth hormone. Aside from insulin, perhaps no other use of hormonally-active sub stance is better exemplified by those drugs which affect fertility. The synthesis of an orally-effective steroid represented one of the first major breakthroughs in the chemical suppression of ovulation. Since the orally active 19-norsteroids were introduced in the 1950s, several oral contra ceptive steroid preparations have been marketed. Indeed, the advent of oral contraceptives for birth control has led to a renewed interest in endocrine pharmacology.

Book excerpt: It can be concluded (under the specific experimental procedures em ployed) that:- 1) HCG labelled with 1 - 2 atoms of radioactive iodine did not differ sig nificantly from the unlabelled hormone; 2) The ovary alone exhibited a capacity to affix specifically HCG; 3) The amount of radioactive material in the ovary was directly proport ional to the quantity of labelled HCG injected; 4) When the HCG present in the circulation is bound to an antiserum to HCG, the antigen-antibody complex is not concentrated by the ovary; 5) Circulating labelled HCG decreased to 50% within 30 minutes following a single intravenous injection; 6) There are four different phases of ovarian uptake of HCG, namely: the first phase, when there is only an inflow from the circulation and stor age mainly in the follicular envelopes; the second phase, when there is a greater inflow than outflow; the third phase, when the inflow is equal to the outflow; and the fourth period, when the outflow is bigger than the in flow. Book excerpt: Somatostatin was discovered in 1971, by Guillemin and his colleagues during their search for the hypothalamic growth hormone-releasing factor. A peptide was found in ovine hypothalamus which inhibited the release of growth hormone from cultured anterior pituitary cells. 1,2 Determination 3 of its amino acid sequence indicated that it was a tetradecapeptide with a molecular weight of 1639 (Figure lJ. An identical peptide was later isolated from porcine hypothalamus by Schally and his coworkers. 4 The peptide was named somatostatin in the belief that it was a hypothalamic releasing factor whose sole function was to inhibit the secretion of growth hormone. It soon became evident, however, that a peptide with identical immunologic characteristics and biologic activity was present in the D cells of pancreatic islets, in D-like cells of the gastrointestinal tract, in parafollicular cells of the thyroid gland, and in extrahypo thalamic neurons of both the central and peripheral nervous system in various species, including man.
