The coverage starts in the early 1950s when the subject of inference for - stricted parameter spaces began to be studied and ends around the middle of 2004. It presents known, and also some new, results on (in)admissibility and minimaxity for nonsequential point estimation problems in restricted ?ni- dimensional parameter spaces. Relationships between various results are d- cussed and open problems are pointed out. Few complete proofs are given, but outlines of proofs are often supplied. The reader is always referred to the published papers and often results are clari?ed by presenting examples of the kind of problems an author solves, or of problems that cannot be solved by a particular result. The monograph does not touch on the subject of testing hypotheses in - stricted parameter spaces. The latest books on that subject are by Robertson, Wright and Dykstra (1988) and Akkerboom (1990), but many new results in that area have been obtained since. The monograph does have a chapter in which questions about the existence of maximum likelihood estimators are discussed. Some of their properties are also given there as well as some algorithms for computing them. Most of these results cannot be found in the Robertson, Wright, Dykstra book.

The potential sources of gravitational waves in globular clusters are analyzed using sophisticated dynamics simulations involving intermediate mass black holes and including, for the first time, high-order post-Newtonian corrections to the equations of motion. The thesis further demonstrates our ability to accurately measure the parameters of the sources involved in intermediate-mass-ratio inspirals of stellar-mass compact objects into hundred-solar-mass black holes. Lastly, it proposes new techniques for the computationally efficient inference on gravitational waves. On 14 September 2015, the LIGO observatory reported the first direct detection of gravitational waves from the merger of a pair of black holes. For a brief fraction of a second, the power emitted by this merger exceeded the combined output of all stars in the visible universe. This has since been followed by another confirmed detection and a third candidate binary black hole merger. These detections heralded the birth of an exciting new field: gravitational-wave astrophysics.

Book excerpt: The research detailed in this monograph was originally motivated by our interest in control problems involving partial and delay differential equations. Our attempts to apply control theory techniques to such prob lems in several areas of science convinced us that in the need for better and more detailed models of distributed/ continuum processes in biology and mechanics lay a rich, interesting, and challenging class of fundamen tal questions. These questions, which involve science and mathematics, are typical of those arising in inverse or parameter estimation problems. Our efforts on inverse problems for distributed parameter systems, which are infinite dimensional in the most common realizations, began about seven years ago at a time when rapid advances in computing capabilities and availability held promise for significant progress in the development of a practically useful as well as theoretically sound methodology for such problems. Much of the research reported in our presentation was not begun when we outlined the plans for this monograph some years ago. By publishing this monograph now, when only a part of the originally intended topics are covered (see Chapter VII in this respect), we hope to stimulate the research and interest of others in an area of scientific en deavor which has exceeded even our optimistic expectations with respect to excitement, opportunity, and stimulation. The computer revolution alluded to above and the development of new codes allow one to solve rather routinely certain estimation problems that would have been out of the question ten years ago.

Book excerpt: Most of the modeling performed in biology aims at achieving a quantitative description and understanding of the intracellular signaling pathways within a "typical cell". However, in many biologically important situations even genetically identical cell populations show a heterogeneous response. This means that individual members of the cell population behave differently. Such situations require the study of cell-to-cell variability and the development of models for heterogeneous cell populations. The main contribution of this thesis is the development of unifying modeling frameworks for signal transduction and proliferation processes in heterogeneous cell populations. These modeling frameworks allow for the detailed description of individual cells as well as differences between them. In contrast to many existing modeling approaches, the proposed frameworks allow for a direct comparison of model predictions with available data. Beyond this, the proposed population models can be simulated efficiently and, by exploiting the model structures, we are able to develop model-tailored Bayesian parameter estimation methods. These methods enable the calculation of the optimal parameter estimates, as well as the evaluation of the parameter and prediction uncertainties. The proposed tools allow for novel insights in population dynamics, in particular the model-based characterization of population heterogeneity and cellular subgroups. This is illustrated for two different application examples: pro- and anti-apoptotic signaling, which is interesting in the context of cancer therapy, and immune cell proliferation.

Book excerpt: Markov Chain Monte Carlo (MCMC) methods are sampling based techniques, which use random numbers to approximate deterministic but unknown values. They can be used to obtain expected values, estimate parameters or to simply inspect the properties of a non-standard, high dimensional probability distribution. Bayesian analysis of model parameters provides the mathematical foundation for parameter estimation using such probabilistic sampling. The strengths of these stochastic methods are their robustness and relative simplicity even for nonlinear problems with dozens of parameters as well as a built-in uncertainty analysis. Because Bayesian model analysis necessarily involves the notion of prior knowledge, the estimation of unidentifiable parameters can be regularised (by priors) in a straight forward way. This work draws the focus on typical cases in systems biology: relative data, nonlinear ordinary differential equation models and few data points. It also investigates the consequences of parameter estimation from steady state data; consequences such as performance benefits. In biology the data is almost exclusively relative, the raw measurements (e.g. western blot intensities) are normalised by control experiments or a reference value within a series and require the model to do the same when comparing its output to the data. Several sampling algorithms are compared in terms of effective sampling speed and necessary adaptations to relative and steady state data are explained.
