Book excerpt: Gold nanoparticles (AuNPs) have been widely used in optical, catalytic, chemical and biomedical applications due to their distinct physicochemical properties. Molecular recognition and the binding of chemical and biological entities have attracted particular research interest for the relevance in drug delivery and diagnostics. By addressing material design as well as advanced characterisation methods, this thesis aims to shed light on fundamental aspects of AuNPs, including the synthesis, functionalisation and implications for relevant applications. A versatile AuNP synthesis enabled the decoupling of core size refinement and ligand shell variation in sub-10 nm thiol-capped AuNPs, thus providing a novel solution of comparative fine-tuning of the ligand shell composition. The analysis of AuNP size distribution was addressed with a comparative study employing single particle analysis (electron microscopy imaging) and ensemble methods (X-ray scattering and analytical ultracentrifugation). Subsequently, nanoscale colloidal stability and molecular interaction with small molecules were investigated both in solution by small-angle X-ray scattering and on a surface by quartz crystal microbalance with dissipation monitoring. This work establishes a versatile platform for AuNP engineering with target size and surface functionality and provides a powerful toolbox for systematic studies on colloidal behaviour as well as ligand-shell mediated nanoparticle-stimuli interactions for biomedical applications.

In that way, three different projects were developed; one project using fluorescence and two projects using Raman spectroscopy as measuring technique. The fluorescence spectroscopy project used the fluorescence lifetime imaging microscope (FLIM) to evaluate the relative position of the molecules methylene blue (MB) and cucurbit[7]uril (CB) on the gold nanoparticle (AuNP) surface. Although the inclusion complex is favored in solution, it was found that MB forms an exclusion complex with CB, when CB is attached to the AuNP surface. The first project utilizing Raman spectroscopy, specifically surface enhanced Raman scattering (SERS), took advantage of a confined system (a reverse micelle) to evaluate the Raman signal of water molecules in close proximity to the AuNP surface. It was observed that the SERS water signal had a big shift to higher energies compared with the Raman signal of the bulk water; indicating the water molecules in the system are subjected to different bond-stretching energies. The second Raman project studied the modification of two different AuNS (specifically AuNP and gold nanorod -AuNR) with thiols. Different thiols were used to evaluate the kinetics of the modification of the AuNS surface, also the different AuNS presented different ligands on their surface. In general, and considering the difference in the bonding strength of the ligands present on the AuNS surface (by synthesis) and the size of the thiol, at least 2 h are required to modify the complete AuNS surface.

These targeted therapies have potential to provide a patient with a form of personalized medicine because these therapies are biological molecules that specifically target other molecules involved with a cancer's growth. Past trials using these therapeutic molecules, however, have led to controversial results, where certain patients responded better than others to the therapy for unknown reasons. Elucidating the reason behind these mixed results can be accomplished using metal nanoparticle technologies which could provide a bright signal to monitor the path that these therapeutic molecules take in vivo as well as enhance the molecule's efficacy. Literature has shown that presenting targeting molecules in a dense manner to their target will increase these molecules' binding affinity. This concept has been explored here to increase binding affinity of therapeutic molecules by attaching these molecules in a dense manner on the surface of gold nanoparticles, and correlating this increased affinity with therapeutic efficacy. Additionally, gold nanoparticles provide an easy surface for molecules to be functionalized on and have shown to be effective imaging, x-ray, and photothermal therapy agents. A major roadblock to using these gold nanoparticles clinically is their non-degradability and thus potential to cause long-term negative side effects in vivo. A platform for developing biodegradable gold nanoparticles is also explored here to take advantage of the gold nanoparticles' excellent imaging and drug delivery capabilities while still allowing them to be used safely in the long term.

This interaction with the biological medium modulates the surface of the nanoparticles, conferring a “biological identity” to their surfaces (referred to as a “corona”), which determines the subsequent cellular/tissue responses. The new interface between the nanoparticles and the biological medium/proteins, called “bio-nano interface,” has been very rarely studied in detail to date, though the interest in this topic is rapidly growing. In this book, the importance of the physiochemical characteristics of nanoparticles for the properties of the protein corona is discussed in detail, followed by comprehensive descriptions of the methods for assessing the protein-nanoparticle interactions. The advantages and limitations of available corona evaluation methods (e.g. spectroscopy methods, mass spectrometry, nuclear magnetic resonance, electron microscopy, X-ray crystallography, and differential centrifugal sedimentation) are examined in detail, followed by a discussion of the possibilities for enhancing the current methods and a call for new techniques. Book excerpt: Materials that can interact with and transit membranes without toxic bilayer disruption or poration are of great interest in the drug delivery field. These materials can presumably bypass endocytosis to directly enter the cell cytosol through the plasma membrane, which is often the desired site of action for therapeutics, thereby avoiding potential and likely cargo degradation if trapped in endosomes. Alternatively, if these materials are endocytosed, they are often able to escape the endosome by interacting with and transiting the endosomal membrane, and moving into the cytosol. Here, we present a unique membrane-interacting, amphiphilic gold nanoparticle (amph-AuNP) system, which we show can interact with, embed within, and even penetrate through multiple adjacent lipid bilayers without evidence of membrane disruption or poration. By virtue of these key properties, we also present these amph-AuNPs as effective carriers for therapeutic molecules. Using a one-step reaction, we synthesized small -2-4 nm core size amph- AuNPs with an amphiphilic ligand shell comprised of one or two alkanethiols. Each amph-AuNP is coated by a mixture of long-chain mercaptoundecanesulfonate (MUS) terminated by a water-soluble sulfonate group, and in some cases, short-chain hydrophobic octanethiol (OT). First, we describe our efforts to adapt and develop a method to synthesize giant multilamellar model membranes to study amph-NP-membrane interaction in a well-defined setting. We show that giant membranes can be synthesized and fine-tuned by varying lipid composition and buffer salt concentration, can be fluorescently labeled with lipid tracers, and can be analyzed robustly with confocal microscopy and flow cytometry. Second, we describe our systematic analysis of amph- AuNP and membrane characteristics that influence mechanisms of NP association with bilayers. We study effects of general membrane properties such as electrostatics and phase that govern NP-membrane interactions, and found that NP penetration of bilayers was blocked under conditions where strong electrostatic repulsion or gel-phase lipids were employed. We further studied effects of AuNP core diameter, surface charge, and surface hydrophobicity on NP-membrane interactions at the nanoscale. We found that MUS particles with an optimal gold core size -2- 3nm in diameter and MUS:OT particles of a broader size range were capable of inducing hemifusion between liposomal membranes, while MUS:OT 2:1 particles of intermediate hydrophobicity were capable of spontaneously aggregating within the bilayer of vesicles to form Janus egg-like morphologies. Third, we built on these NP membrane-embedding properties to explore and characterize NP-embedding in erythrocyte membranes, with particular attention to the glycocalyx and membrane fluidity, for the future application of constructing therapeutic erythrocyte 'pharmacytes' in situ. Finally, we describe work in engineering amph-AuNPs to carry short antigenic peptide cargoes for in vivo vaccine applications, where immunization experiments have shown much promise for antigen-ferrying amph-AuNPs in eliciting robust and long-lasting CD8+ T cell responses.

Numerous reports have been dedicated to AuNP interaction with protein or organothiol alone. Competitive protein and organothiol (OT) interaction is, however, mostly an unexplored area. The research reported here focused on developing a fundamental understanding of sequential and simultaneous protein and organothiol interaction with AuNPs in which protein and OT are added either simultaneously or sequentially into the colloidal AuNP solutions. In studies of OT interactions with bovine serum albumin (BSA) stabilized AuNPs, we found that the protein coating layer is highly porous and permeable for small molecules such as mercaptobenzimidazole (MBI), cysteine (Cys), homocysteine (Hcy), and glutathione (GSH). Based on the amounts of MBI adsorbed and the kinetics of MBI adsorption onto BSA stabilized AuNPs, we were able to get an insight into protein conformational changes on the AuNPs. The competitive and sequential studies of protein and OT interactions with AuNPs involving eight model organothiols showed that the protein and OT co-adsorption onto AuNPs is a kinetically controlled process. The AuNP stability against ligand-adsorption-induced AuNP aggregation differed significantly among the AuNP/OT and AuNP/BSA/OT mixtures where the AuNP stability order increased from (AuNP/OT)/BSA to AuNP/(BSA/OT), and finally (AuNP/BSA)/OT samples (the two components inside the parenthesis are mixed first followed by the addition of the third component). The studies on the role of cysteine in protein-AuNP interactions found that the cysteine has no significant effect on the kinetics of protein adsorption onto AuNPs. However the stability of the protein-AuNP complex against the organothiol-adsorption induced AuNP aggregation increased as the number of cysteine residues increased from zero to two. Besides providing new insights on protein interaction with AuNPs, this research is important for AuNP biological/biomedical applications because AuNPs in biofuids encounter a mixture of proteins and OTs in addition to other molecular species.

The goal of each volume is to give the reader, whether at a university or in industry, a comprehensive overview of an area where new insights are emerging that are of interest to a larger scientific audience. Thus each review within the volume critically surveys one aspect of that topic and places it within the context of the volume as a whole. The most significant developments of the last 5 to 10 years should be presented using selected examples to illustrate the principles discussed. A description of the physical basis of the experimental techniques that have been used to provide the primary data may also be appropriate, if it has not been covered in detail elsewhere. The coverage need not be exhaustive in data, but should rather be conceptual, concentrating on the new principles being developed that will allow the reader, who is not a specialist in the area covered, to understand the data presented. Discussion of possible future research directions in the area is welcomed. Review articles for the individual volumes are invited by the volume editors.
