Book excerpt: A high resolution spectral domain optical coherence tomography (SD-OCT) system was used to study in-vivo early morphological changes and optical nerve head (ONH) blood flow in Long Evans rat retina, induced by injection of sodium iodate (NaIO3). OCT images were acquired at the same retinal location from healthy rats and from rats at 1, 3, 6 12, 24, 72 and 168 hours post drug administration. Morphological OCT images showed the formation of a new low reflective layer between photoreceptor OS and RPE in all tested rats which was most likely fluid accumulation. A Doppler Optical Micro-Angiography (DOMAG) algorithm was used to carry out quantitative analysis of ONH blood flow. Estimation of flow rate on each ONH vessel was done by measurements of Doppler angle, vessel size and axial velocity. This study has demonstrated that the capability of UHR-OCT to observe optical reflectance and layer thickness changes, rearrangement and detachment of photoreceptor OS and RPE layers, together with flow rate estimation of retinal blood vessels. Therefore, it can serve as markers in future non-invasive, in-vivo studies of disease or drug induced retinal degeneration in ophthalmic research.

The aim of this thesis is to develop multi-modal non-invasive imaging technology for studying retinal degeneration and gene therapy in mice. We have constructed a FD-OCT prototype and combined it with a Scanning Laser Ophthalmoscope (SLO) to permit real time alignment of the retinal field of view. Fluorescence detection was integrated with the SLO to enable the en face localization of a molecular contrast signal, which is important for retinal angiography and retinal auto-fluorescence imaging. The integrated FD-OCT/fSLO system was investigated for imaging the retina of mice In vivo. The results of this research have demonstrated that the state of art FD-OCT/fSLO system OCT is a powerful tool to conduct research retinal degeneration and gene therapy in mice.

Book excerpt: A prototype spectrometer based on Fourier Domain Optical Coherence Tomography (FD OCT) system was used as a non-invasive depth resolved medical imaging tool for diagnostic visualization of the retinal structures In vivo. This thesis investigated so-called Spectral Domain (SD) OCT to detect and monitor pathological signs of retinal degeneration in small animals such as mice, rats and Xenopus laevis larvae. Non-invasive SD OCT imaging was validated against invasive retinal cross sectional images (histology) acquired from a nearby region for comparison of specific retinal layers. The retinal layers were manually segmented for quantitative thickness measurements, and followed by statistical analysis to determine the onset of detectable retinal degeneration. The results of this research have demonstrated that FD OCT is a powerful tool for quantitative in vivo measurement of retinal degeneration in small animals.
