Book excerpt: This volume contains papers presented at the international conference on "Transgenic Mice and Mutants in MHC Research", held in Bar Harbor, Maine, in June, 1989. The meeting brought together eighty researchers working in the field. While mouse H-2 mutants have been known for many years their studies continue to contribute a great deal to our understanding of structure/function relationships and evolution of MHC molecules. Recently a new direction of research has emerged on regulation, expression and function of MHC genes using transgenic animals carrying exogenous MHC genes from the same or other species or engineered MHC genes. With the introduction of transgenic mice more specific questions about the various functions of MHC genes can be answered as, for example, the role of soluble and membrane-bound MHC molecules in self tolerance and elimination of specific T cell clones, recognition of human MHC antigens by the mouse immune system, the role of individual human MHC genes in disease susceptibility. New approaches for evaluation of the role of MHC linked and unlinked genes in susceptibility of mice to malignant tumors and their metastases have been also reported. Our purpose has been to provide a forum for discussion of these new developments. Many questions remain to be answered but the necessary tools have become available. We thank the contributors and hope that the readers will benefit from this Pro ceedings. Book excerpt: The major histocompatibility complex (MHC) is a highly polymorphic and diverse multigene locus in all jawed vertebrate species that has an integral role in adaptive/innate immune systems, transplantation, and infectious and autoimmune diseases. The MHC supra-locus in mammalian vertebrates is usually partitioned into three distinct regions, known as classes I, II, and III, which, to varying extents, can be found conserved in nonmammalian jawed vertebrates, such as bony fish, amphibians, and bird lineages. The MHC gene region is characterized particularly by the expression of class I and class II glycoproteins that bind peptides derived from intracellular or extracellular antigens to circulating T-cells. While this expressed antigenic specificity remains the predominant interest with respect to MHC function and polymorphism in a population, a broader concept has emerged that examines the MHC as a multifunctional polymorphic controller that facilitates and regulates genome diversity with a much greater array of functions and effects than just MHC-restricted antigen recognition. This volume of 19 reprints presented by various experts and collected from the Special Issue of Cells on “MHC in Health and Disease” covers a broad range of topics on the genomic diversity of the MHC regulatory system in various vertebrate species, including MHC class I, II, and III genes; innate and adaptive immunity; neurology; transplantation; haplotypes; infectious and autoimmune diseases; fecundity; conservation; allelic lineages; and evolution. Taken together, these articles demonstrate the immense complexity and diversity of the MHC structure and function within and between different vertebrate species.

In science, too, laws of hybrid vigor seem to operate at the interface between two disciplines, an interface that often proves to be fertile ground for germinating concepts and new outlooks. The fringes of research into the major histocompatibility complex (Mhc) have provided such an interface several times in the past and the encounters have invigorated fields such as transplantation biology, cellular immunology, and immunogenetics. In the last few years, a new interface has been emerging between Mhc and evolutionary genetics, and particularly the branch of evolutionary genetics dealing with molecular evolution. Mhc research relies upon molecular evolutionary genetics, with its grand superstructure of mathematical formulations, to come to grips with the events leading to and maintaining the Mhc polymorphism. Without the armament of rigorous statistical procedures developed by evolutionary geneticists, the intricate relationships among Mhc genes cannot be resolved. It will undoubtedly be a molecular geneticist who is the final arbiter in the dispute concerning the nature of the selection pressure molding the Mhc genes. And it is doubtful whether the true function of Mhc can ever be comprehended without the vantage point afforded by the elucidation of its evolutionary history.

Book excerpt: The major histocompatibility complex (MHC) is a gene region found in all jawed vertebrates. These highly polymorphic, tightly linked genes are responsible for immune regulation by presenting self and non-self antigens to the immune system. The MHC is the most polymorphic vertebrate genome region because its genes face considerable selective pressure since survival depends on the capacity to respond to bacterial, viral and parasitic diseases. The MHC organization in different species has clearly evolved in separate and unique ways since the time that birds and mammals diverged from a common ancestor 310 million years ago. The size and organization of chicken MHC genes is somewhat different from the MHC regions found in mice and humans. Given the wide variability in gene numbers revealed in this study, I wanted to determine whether genetic recombination, crossing over, in the MHC-Y region might be contributing to haplotypic variation. YF molecules seem to have a high recombination frequency between its multiple copies, perhaps on the order of 1% (1 map unit) or higher. This may be due to many similar genes present within the MHC-Y region. Proximity of a 41 base pair repeat known as PO41 might also contribute to instability of the region.

In addition to the PPLO specialists, the conference invited Julius Marmur to compare PPLO DNA to DNA of other organisms; David Garfinkel, who was one of the first to develop computer models of metabolism; Cyrus Levinthal to talk about coding; and Henry Quastler to discuss information theory constraints on very small cells. The conference was an announcement of the role of PPLO in the fundamental understanding of molecular biology. Looking back 40-some years to the Connecticut meeting, it was a rather bold enterprise. The meeting was international and inter-disciplinary and began a series of important collaborations with influences resonating down to the present. If I may be allowed a personal remark, it was where I first met Shmuel Razin, who has been a leading figure in the emerging mycoplasma research and a good friend. This present volume is in some ways the fulfillment of the promise of that early meeting. It is an example of the collaborative work of scientists in building an understanding of fundamental aspects of biology.
